Sour cherry is one of the most popular stone fruits in Poland. In the organic production system of sour cherries, no artificial pesticides and fertilizers are allowed, which is one of the organic production requirements increasingly appreciated by producers and consumers. The taste of fruits is created by the sugar and organic acid content and their ratio. Vitamin C is known for its health-promoting properties. The aim of the present study was to analyze and compare the concentrations of vitamin C, sugars, and organic acids and their profiles in organic vs. conventional sour cherry fruits representing different cultivars, in a three-year experiment. In the presented experiment, four sour cherry cultivars, ‘Kelleris’ 16, ‘Oblacińska’, ‘Pandy 103’, and ‘Debreceni Bötermö’, were cultivated in two horticultural systems, organic and conventional, and the content of sugars and organic acids was analyzed in the fruit with HPLC methods. Organically cultivated sour cherry fruits were characterized by significantly higher concentrations of sugars and vitamin C only in the first year of the experiment, when the mean concentrations of fructose, glucose, and sucrose in these fruits reached 4.15 g/100 g F.W., 0.37 g/100 g F.W., and 0.27 g/100 g F.W., respectively, and the concentration of vitamin C reached 17.28 mg/100 g F.W. In the two subsequent years, conventional cherries were more abundant in these compounds. Among the tested sour cherry cultivars, ‘Oblačińska’ performed the best in terms of sugar content. The mean value for total sugars for ‘Oblačińska’ cv. was 5.53 g/100 g F.W. In the case of vitamin C, the highest levels (av. 28.13 mg/100 g F.W.) were noted in the fruits of ‘Pandy 103’ cv. The strong year-to-year variability underscores the need for multi-year experiments and, where possible, multi-site trials, to disentangle cultivar × system × environment interactions. Because the quality of sour cherry for fresh consumption and for processing depends mostly on sugar content, for organic production, ‘Oblačińska’ cv. is strongly recommended. Full article

Passive acoustic monitoring (PAM) using hydrophones enables collecting acoustic data to be collected in large and diverse quantities, necessitating the need for a reliable automated classification system. This paper presents a data-processing pipeline and a set of neural networks designed for a humpback-whale-detection system. A collection of audio segments is compiled using publicly available audio repositories and extensively curated via manual methods, undertaking thorough examination, editing and clipping to produce a dataset minimizing bias or categorization errors. An array of standard data-augmentation techniques are applied to the collected audio, diversifying and expanding the original dataset. Multiple neural networks are designed and trained using TensorFlow 2.20.0 and Keras 3.13.1 frameworks, resulting in a custom curated architecture layout based on research and iterative improvements. The pre-trained model MobileNetV2 is also included for further analysis. Model performance demonstrates a strong dependence on both feature representation and network architecture. Mel spectrogram inputs consistently outperformed MFCC (Mel-Frequency Cepstral Coefficients) features across all model types. The highest performance was achieved by the pretrained MobileNetV2 using mel spectrograms without augmentation, reaching a test accuracy of 99.01% with balanced precision and recall of 99% and a Matthews correlation coefficient of 0.98. The custom CNN with mel spectrograms also achieved strong performance, with 98.92% accuracy and a false negative rate of only 0.75%. In contrast, models trained with MFCC representations exhibited consistently lower robustness and higher false negative rates. These results highlight the comparative strengths of the evaluated feature representations and network architectures for humpback whale detection. Full article

Background/Objectives: Infectious keratitis remains a major cause of blindness worldwide, and many cases progress to therapeutic keratoplasty despite advances in antimicrobial therapy. This systematic review aims to evaluate the outcomes of therapeutic keratoplasty in microbial keratitis and examine factors influencing anatomical success, graft survival, and visual rehabilitation. Methods: A systematic review was conducted following PRISMA guidelines, including English-language studies, published between 2000 and 2025. Studies with ≥10 eyes and ≥6 months follow-up were included. Data on infection control, graft clarity, anatomical success, visual acuity, and complications were extracted. Results: Fourteen studies encompassing 1527 eyes were analyzed. TPK accounted for 89% of procedures; DALK was used selectively for anterior or mid-stromal infections. Overall infection control ranged from 69 to 100%, with globe preservation in 85–100% of cases. Bacterial keratitis had higher cure rates and graft clarity than fungal or Acanthamoeba keratitis. Larger grafts (>8 mm) and deep stromal involvement were associated with increased graft rejection and postoperative complications. DALK offered higher graft survival and lower immunologic risk when the endothelium was spared. Visual outcomes were generally limited, reflecting preoperative disease severity, timing of surgery, and postoperative immunomodulation constraints. Early surgical intervention improved anatomical outcomes in severe fungal keratitis. Conclusions: Therapeutic keratoplasty is an effective globe-preserving intervention in advanced microbial keratitis, but with limited functional outcomes. Further prospective studies are needed to refine surgical indications, postoperative management, and long-term functional results. Full article

Cetaceans are artiodactyls adapted to live in the marine environment, and this group includes whales, dolphins, and porpoises. Although mitochondrial nucleotide diversity has been reported separately for many cetacean groups, the proportion of deleterious mutations in these populations is unknown. Furthermore, a comparison of mitogenomic diversities across all cetaceans is also lacking. To investigate this, we conducted a comparative genomic analysis of 2244 mitochondrial genomes from 65 populations across 32 cetacean species. We observed a 78-fold variation in mitogenomic diversity among cetacean populations, suggesting a large difference in genetic diversity. We used the ratio of nonsynonymous-to-synonymous diversities (dN/dS) to measure the proportion of deleterious mutations in the mitochondrial exomes. The dN/dS ratio showed a 22-fold difference between the cetacean population. Based on genetic theories, the large differences observed in the two measures could be attributed to differences in the effective sizes of the cetacean populations. Typically, small populations have low heterozygosity and a high dN/dS ratio, and the reverse is true for large populations. This was further confirmed by the negative correlation observed between heterozygosity and dN/dS ratios of cetacean populations. While our analysis revealed similarities in mitogenomic diversity between the endangered and least-concern cetacean species, the dN/dS ratio of the former was found to be higher than that of the latter. The findings of this study are useful for identifying the relative magnitude of reductions in the population sizes of different cetacean species. This will help conservation management efforts prioritise the use of limited resources, time, and effort to protect the cetacean populations that need immediate attention. Full article

Objective: The objective was to evaluate the optimal timing of cervical cerclage insertion for perinatal outcomes, such as birthweight, gestational week, and pregnancy prolongation in women with diagnosed cervical insufficiency (CI). Methods: This retrospective study was conducted at the 1st Department of Obstetrics and Gynaecology of the Medical University of Warsaw, over a 10-year period. Maternal and perinatal outcomes were compared between 75 women divided into three groups based on the gestational week (GW) at cerclage insertion: (1) before 18 GW (n = 31), (2) 18–22 GW (n = 31), (3) after 22 GW (n = 13). Only single pregnancies were included in the final analysis in order to maintain the homogeneity of the population. The primary outcomes included the week of delivery and pregnancy prolongation following cervical cerclage insertion. Numerous secondary outcomes were also evaluated, including neonatal mortality, need for NICU hospitalization, Apgar score, birthweight, maternal white blood cell (WBC) count and C-reactive protein (CRP) levels. Results: Birth week was significantly associated with GW at insertion—35.8 ± 3.8 vs. 34.8 ± 5.2 vs. 32 ± 5.7, respectively, p = 0.016. Moreover, statistical difference was also found regarding birthweight of the analysed groups—2723.8 ± 951.6 g vs. 2518.5 ± 1167.9 g vs. 1886.7 ± 1011.2 g, respectively, p < 0.001, and pregnancy prolongation following cerclage insertion 20.4 ± 4.2 vs. 14.7 ± 5.5 vs. 7.3 ± 5.7 weeks, respectively, p < 0.001. Conclusions: Earlier cerclage placement (<18 weeks) is associated with significantly improved perinatal outcomes. However, this association largely reflects the benefit of prophylactic intervention over emergency ‘rescue’ procedures (common in the >22-week group). The sharp decline in outcomes after 22 weeks highlights the risks of advanced cervical dilation, suggesting that clinical management should prioritize risk assessment within the prophylactic window. Pathogens that have developed resistance to antibiotics pose a threat to public health. The primary goal in preventing foodborne infections is to inhibit the growth of and, subsequently, eliminate antibiotic-resistant pathogens at every stage from production to consumption. Escherichia coli, which has acquired resistance to most known antibiotics, is frequently found in chicken meat. In many countries, due to unregulated antibiotic use in poultry farming, poor hygiene in slaughterhouses, or cross-contamination, extended-spectrum beta-lactamase (ESBL)-producing E. coli has been identified as the causative agent in poultry-associated food poisoning. The need for more effective antimicrobial agents against this pathogen, which is resistant to existing antibiotics, has led to increased attention being paid to postbiotics produced by lactic acid bacteria, particularly bacteriocins. This study aimed to determine the antimicrobial effects of postbiotics obtained from kefir-derived Lactiplantibacillus plantarum and Lactococcus lactis against ESBL-positive E. coli. To achieve this, E. coli strains were isolated from raw chicken meat samples collected from the market using culture-based methods, and their antimicrobial resistance profiles were determined using the disk diffusion method. The ESBL positivity of the isolates was assessed using the double-disk synergy test. The antimicrobial activities of the postbiotics against the identified ESBL-positive E. coli strains were tested using the macro-dilution method to determine minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values. ESBL-positive E. coli was detected in 48% of raw chicken meat samples. The antimicrobial effects of postbiotics were examined by disk diffusion, and postbiotics produced by 18 Lb. plantarum strains and 20 Lc. lactis strains showed strong antimicrobial activity. Significant differences in the antimicrobial effects of postbiotics were observed between the two species. Lb. plantarum postbiotics exhibited both bacteriostatic (concentration 60%) and bactericidal (concentration 80%) effects on ESBL-positive E. coli strains, whereas Lc. lactis postbiotics showed only bacteriostatic effects (80% concentration). Postbiotics derived from probiotic bacteria offer promising effects against multidrug-resistant E. coli due to their heat resistance, activity across different pH values, strong antimicrobial effects, affordability, and ease of production. This study aims to examine the impacts of AI investment on economic growth, while controlling for labor force participation and gross fixed capital formation in Germany. The analysis is based on data collected on the state of economic development in Germany from the first quarter of 2012 to the fourth quarter of 2022. The study used a nonlinear ARDL bounds approach for these investigations. The outcomes clearly reveal that positive shocks to labor force participation and investment in AI significantly enhance economic growth (GDP) in Germany, whereas a positive shock to gross fixed capital formation (GFCF) has no considerable effect on economic growth. Likewise, negative shocks to gross fixed capital formation and AI investment increase GDP growth. Negative shock to the labor force reduces GDP growth. Recommendations are made that Germany must maintain its measured approach while offering long-term commitment. More spectacular AI expenditure increases are not required; prioritize steadiness and integration. Establish long-time-horizon AI development partnerships between government, universities, and industry with stable funding streams on long-term horizons. The fragile link between traditional capital formation and growth means that Germany needs to redefine productive investment. Full article

Newborn screening (NBS) for sickle cell disease (SCD) has been performed in the United States (US) for decades, significantly reducing infant morbidity and mortality. A landmark clinical trial demonstrated that early identification of SCD enabled timely and life-saving prophylactic penicillin; this led to recommendations for universal NBS across the US. Early use of hydroxyurea as a safe and effective treatment for SCD further improved clinical outcomes by preventing acute and chronic disease complications. These advances add to the importance of early diagnosis through NBS, providing an opportunity for early treatment intervention. In recent years, high-resource countries—including those in Europe, the UK, and Canada—have adopted NBS for SCD using diverse strategies. Simultaneously, pilot programs in lower-resource settings such as Africa, Brazil, and India have demonstrated local feasibility and impact through implementation efforts. An overarching equity gap for achieving global NBS for SCD is the variable access to simple, accurate, and affordable testing. Other challenges include timing of NBS testing, targeted populations, laboratory methods, and parental education with genetic counseling. Questions remain about the equitable enrollment of affected infants worldwide into comprehensive care to ensure early treatment. These challenges raise concerns about sustainability, underscore the need for long-term funding and a strategic plan, and highlight persistent inequities from the lack of global NBS standards. As a packaging material, molded pulp has experienced sustained growth in demand because of its high recyclability and biodegradability. However, refined structural modeling of their behavior during transportation remains limited. This study established finite element representations of molded pulp packaging structural cells and developed a spring–mass dynamic model that incorporates the parameters of the packaged product. The proposed model can predict the combined modal characteristics of molded pulp packaging and the protected item while significantly reducing the computational requirements compared with traditional FEM (finite element method) analyses. Experimental validation shows that the prediction error of the first order modal frequency is approximately 6%, which meets the actual needs. The model provides a foundation for the subsequent optimization of molded pulp structures under road transport conditions. Full article

Background and Objectives: Acute and obstructive pyelonephritis (AOP) management, despite advancements in diagnostic imaging and antimicrobial therapy, is characterized by delayed recognition and increasing antimicrobial resistance. This review aimed to summarize current evidence regarding the clinical characteristics, microbiological spectrum, biomarkers, and imaging findings associated with AOP. Materials and Methods: A systematic review was conducted according to PRISMA guidelines and registered in PROSPERO (CRD420251162736). Literature searches were performed across the PubMed, Scopus, and Web of Science databases for articles published between January 2014 and 31 March 2025 using the term “acute obstructive pyelonephritis”. Inclusion criteria comprised original full-text English-language studies, published in the last 10 years and conducted in adults, reporting clinical, laboratory, microbiological, and imaging characteristics. Exclusion criteria are letters to the editor, expert opinions, case reports, conference or meeting abstracts, reviews, and redundant publications; having unclear or incomplete data; and being performed on cell cultures or on mammals. The quality of included studies was assessed using the Newcastle–Ottawa Scale. Results: Twenty-three studies met the inclusion criteria. AOP predominantly affected elderly patients with comorbidities, especially diabetes mellitus and urinary tract obstruction. Predictors of septic shock included thrombocytopenia, hypoalbuminemia, elevated procalcitonin (>1.12 µg/L), presepsin, and a neutrophil-to-lymphocyte ratio ≥ 8.7. Escherichia coli remained the leading pathogen (60–95%) with extended-spectrum β-lactamase (ESBL) rates between 20 and 70%, followed by Klebsiella pneumoniae. CT demonstrated 71–100% sensitivity for detecting obstructive complications, confirming its superiority over ultrasound, while MRI provided comparable diagnostic accuracy in selected cases. Conclusions: AOP requires prompt recognition and early decompression to prevent sepsis-related mortality. Biomarkers such as procalcitonin, presepsin, and neutrophil to lymphocyte ratio enhance risk stratification, while CT remains the gold-standard imaging modality. Effect of First-Line Combination Systemic Therapy on Favorable-Risk Clear Cell Renal Cell Carcinoma: A Retrospective Study

Background/Objectives: For patients with advanced or metastatic clear cell renal cell carcinoma (RCC), combinations of immune checkpoint inhibitors (ICIs) and VEGFR-targeted tyrosine kinase inhibitors (TKIs) are standard first-line therapies. However, the clinical benefit of these regimens in patients with favorable IMDC risk remains unclear. Methods: We retrospectively analyzed 147 patients with favorable-risk metastatic RCC treated with first-line systemic therapy at the Samsung Medical Center between 2019 and 2023. Treatment regimens included TKI monotherapy (n = 110) or ICI–TKI combinations (n = 37). Progression-free survival (PFS), overall survival (OS), and objective response rate (ORR) were evaluated using Kaplan–Meier and Cox regression analyses. Results: At a median follow-up of 46.3 months, the overall median PFS was 20.1 months (95% CI, 14.5–25.7). Median PFS was 26.2 months with ICI–TKI combinations versus 17.0 months with TKI monotherapy (HR 1.32; 95% CI, 0.82–2.12; p = 0.25). In multivariate analysis, TKI monotherapy (HR 14.01; p = 0.002) and liver metastasis (HR 9.17; p < 0.001) were independent predictors of shorter PFS. ORR was significantly higher with combination therapy (68% vs. 46%; p = 0.01). Median OS was not reached in either group, with 3-year OS rates of 89% and 84%, respectively. Conclusions: The findings suggest that even within the favorable-risk population, clinical heterogeneity influences treatment outcomes, emphasizing the need for individualized therapy selection and refined prognostic models. Background: Intravascular hemolysis is a common complication in patients undergoing extracorporeal membrane oxygenation (ECMO), with plasma free hemoglobin (pfHb) serving as a biomarker for detection. Without standardized protocols, laboratories face challenges in interpreting and reporting results. Hemolysis indices may enhance reporting accuracy. Methods: This retrospective observational study at University Hospital Center Zagreb included 61 lithium heparin plasma samples from ECMO patients. pfHb was measured using the Harboe method (fHb) and estimated from hemolysis indices on Abbott Alinity c analyzer (efHb). Total and conjugated bilirubin, hemolysis, icterus, and lipemia indices (HIL) were recorded. Method comparison used Passing-Bablok regression and Bland–Altman analysis. An algorithm for pfHb reporting accounting for HIL interferences was developed. Results: Significant differences were observed between methods, with Harboe yielding higher median fHb (261 mg/L) versus efHb (58 mg/L). Regression analysis showed constant negative bias of −91 mg/L (95% CI: −143 to −16) for efHb relative to fHb. Bland–Altman analysis demonstrated wide limits of agreement. Correlation between fHb and efHb was moderate (Spearman’s rho = 0.618, p < 0.001). The delta between methods increased with higher bilirubin concentrations. An algorithm integrating HIL indices with the Harboe method was developed to guide result validation and reporting. Conclusions: Accurate hemolysis assessment in ECMO patients requires careful interpretation, appropriate method selection, and laboratory–clinician collaboration. The proposed algorithm improves the clinical utility of pfHb testing by accounting for analytical interferences and supporting informed decision-making. Full article

To address the urgent need for high-fidelity motion data for validating navigation algorithms for Unmanned Underwater Vehicles (UUVs), this paper proposes a data generation method based on a parametric motion model. First, based on the principles of rigid body dynamics and fluid mechanics, a decoupled six-degrees-of-freedom (6-DOF) Linear and Angular Acceleration Vector (LAAV) model is constructed, establishing a dynamic mapping relationship between the rudder angle and speed setting commands and motion acceleration. Second, a segmentation–identification framework is proposed for three-dimensional trajectory segmentation, integrating Gaussian Process Regression and Ordering Points To Identify the Clustering Structure (GPR-OPTICS), along with a Dynamic Immune Genetic Algorithm (DIGA). This framework utilizes real vessel data to achieve motion segment clustering and parameter identification, completing the construction of the LAAV model. On this basis, by introducing sensor error models, highly credible Inertial Measurement Unit (IMU) data are generated, and a complete attitude, velocity, and position (AVP) motion sequence is obtained through an inertial navigation solution. Experiments demonstrate that the AVP data generated by our method achieve over 88% reliability compared with the real vessel dataset. Furthermore, the proposed method outperforms the PSINS toolbox in both the reliability and accuracy of all motion parameters. These results validate the effectiveness and superiority of our proposed method, which provides a high-fidelity data benchmark for research on underwater navigation algorithms. Full article

Topology optimization is a powerful and efficient design tool, but the structures obtained by element-based topology optimization methods are often limited by fuzzy or jagged boundaries. The smooth-edged material distribution for optimizing topology algorithm (SEMDOT) can effectively deal with this problem and promote the practical application of topology optimization structures. This review outlines the theoretical evolution of SEMDOT, including both penalty-based and non-penalty-based formulations, while also providing access to open access codes. SEMDOT’s applications cover diverse areas, including self-supporting structures, energy-efficient manufacturing, bone tissue scaffolds, heat transfer systems, and building parts, demonstrating the versatility of SEMDOT. While SEMDOT addresses boundary issues in topology optimization structures, further theoretical refinement is needed to develop it into a comprehensive platform. This work consolidates the advances in SEMDOT, highlights its interdisciplinary impact, and identifies future research and implementation directions. Full article

Breastfeeding is universally regarded as the cornerstone of infant feeding, as it is the ideal infant feeding choice for optimal nutrition and development. Socio-structural factors of breastfeeding in child health play an important role in guiding women’s decisions on options to feed their babies. A cross-sectional study was conducted among mothers with infants aged 0–6 months in Mthatha, Eastern Cape, with the aim of assessing the role of socio-structural factors in shaping feeding intentions and practices among mothers with infants. Written Informed consent was obtained in accordance with the Declaration of Helsinki from the participants prior to data collection. Data was gathered with a validated designed questionnaire as well as analyzed using Social Sciences (SPSS) version 29. A total of 181 mothers were enrolled. Only 45.9% reported that they exclusively breastfed their babies, of which the highest proportion of exclusive breastfeeding (EBF) was observed among the 21–29 age group at 51.8%, and the lowest among those aged 20 years and below (3.6%). Marital status (p = 0.005) and employment status (p < 0.001) were significantly associated with exclusive breastfeeding, with higher EBF rates observed among married mothers and those who were self-employed. Both the EBF mothers and non-EBF mothers shared a common belief that colostrum was not beneficial for infants (p = 0.854), whereas their views differed significantly on the amount of water given to infants before they reached six months (p = 0.001). There was no significant relationship between EBF status and having a family member who had breastfed in the past six months (p = 0.815); also, a weak association was noted for having a friend who had breastfed recently (p = 0.057). The difference in EBF practice between those receiving antenatal care (ANC) breastfeeding education and those not receiving it was not statistically significant (p = 0.591). A statistically significant association was found between the support level and exclusive breastfeeding status (p < 0.001). This study highlights that the successful practice of exclusive breastfeeding (EBF) is strongly associated with high levels of social support. Interventions are needed to engage active partners, family members, and community members in creating a supportive environment for breastfeeding mothers. Full article
