Identification and Characterization of Resistance to Rust in Lentil and Its Wild Relatives

Lentil rust is a major disease worldwide caused by Uromyces viciae-fabae. In this study, we screened a large germplasm collection of cultivated lentils (Lens culinaris ssp. culinaris) and its wild relatives, both in adult plants in the field with a local rust isolate during 2 seasons and in seedlings under controlled conditions with four fungal isolates of worldwide origin. The main results from our study were the following: (1) a significant number of accessions with resistance based on hypersensitive reaction (reduced Infection Type (IT)) were identified in cultivated lentil and in L. ervoides, L. nigricans and L.c. orientalis. The IT scores showed a clear isolate-specific response suggesting race-specificity, so each fungal isolate might be considered a different race. Resistance was identified against all isolates what might be the basis to develop a standard differential set that should be a priority for rust definition and monitoring. (2) Interestingly, although at lower frequency than in L. ervoides and L. nigricans, the hypersensitive response was also observed within cultivated lentil, with accession 1561 (L.c. culinaris) displaying resistance to the four isolates making this accession a valuable ready-to-use resource for lentil resistance breeding. Resistance to all other rust isolates was also available within L.c. culinaris in an isolate-specific manner. Accession 1308 (L. ervoides) showed resistance against all isolates tested, as well as a reduced number of accessions belonging to other wild Lens species. (3) In addition, our screenings allowed the identification of several accessions with partial resistance (reduced Disease Severity (DS) despite high IT). Adult Plant Resistance resulting in reduced severity in adult plants in the field, despite high susceptibility in seedlings, was more frequently identified in L.c. culinaris, but also in L. nigricans and L.c. orientalis. Lentils (Lens sp.) are one of the main sources of protein for humans in many regions, in part because their rusticity allows them to withstand semi-dry climates and tolerate a wide spectrum of pests. Both are also highly sought-after attributes to face climate change. Wild accessions, rather than cultivated varieties, are typically the holders of most influential alleles for rusticity traits. However, most genomic and transcriptomic research conducted in lentils has been carried out on commercial accessions (L. culinaris), while wild relatives have been largely neglected. Herein, we assembled, annotated, and evaluated the transcriptomes of eight lentil accessions, including the cultivated Lens culinaris and the wild relatives: L. orientalis, L. tomentosus, L. ervoides, L. lamottei, L. nigricans, and two L. odemensis. The assemblies allowed, for the first time, a comparison among different lentil taxa at the coding sequence level, providing further insights into the evolutionary relationships between cultivated and wild germplasm and suggesting a grouping of the seven accessions into at least three conceivable gene pools. Moreover, orthologous clustering allowed a first estimation of the lentil pan-transcriptome. It is composed of 15,910 core genes, encoded in all accessions, and 24,226 accessory genes. The different pan-transcriptome clusters were also screened for Pfam-domain enrichment. The present study has a high novelty, as it is the first pan-transcriptome analysis using six wild species in addition to cultivated species. Because of the amount of transcript sequences provided, our findings will greatly boost lentil research and assist breeding efforts. Full article

Crop Wild Relatives Crosses: Multi-Location Assessment in Durum Wheat, Barley, and Lentil

Crop wild relatives (CWR) are a good source of useful alleles for climate change adaptation. Here, 19 durum wheat, 24 barley, and 24 lentil elites incorporating CWR in their pedigrees were yield tested against commercial checks across 19 environments located in Morocco, Ethiopia, Lebanon, and Senegal. For each crop, the combined analysis of variance showed that genotype (G), environment (E), and genotype x environment (G×E) effects were significant for most of the traits. A selection index combining yield potential (G) and yield stability (G×E) was used to identify six CWR-derived elites for each crop matching or superior to the best check. A regression analysis using a climate matrix revealed that grain yield was mostly influenced by the maximum daily temperature and soil moisture level during the growing stages. These climatic factors were used to define five clusters (i.e., E1 to E5) of mega-environments. The CWR-derived elites significantly outperformed the checks in E1, E2, and E4 for durum wheat, and in E2 for both barley and lentil. The germplasm was also assessed for several food transformation characteristics. For durum wheat, one accession (Zeina) originating from T. araraticum was significantly superior in mixograph score to the best check, and three accessions originating from T. araraticum and T. urartu were superior for Zn concentration. For barley, 21 accessions originating from H. spontaneum were superior to the checks for protein content, six for Zn content, and eight for β-glucan. For lentil, ten accessions originating from Lens orientalis were superior to the check for protein content, five for Zn, and ten for Fe concentration. Hence, the results presented here strongly support the use of CWR in breeding programs of these three dryland crops, both for adaptation to climatic stresses and for value addition for food transformation. Challenged by population increase, climatic change, and soil deterioration, crop improvement is always a priority in securing food supplies. Although the production of grain legumes is in general lower than that of cereals, the nutritional value of grain legumes make them important components of food security. Nevertheless, limited by severe genetic bottlenecks during domestication and human selection, grain legumes, like other crops, have suffered from a loss of genetic diversity which is essential for providing genetic materials for crop improvement programs. Illustrated by whole-genome-sequencing, wild relatives of crops adapted to various environments were shown to maintain high genetic diversity. In this review, we focused on nine important grain legumes (soybean, peanut, pea, chickpea, common bean, lentil, cowpea, lupin, and pigeonpea) to discuss the potential uses of their wild relatives as genetic resources for crop breeding and improvement, and summarized the various genetic/genomic approaches adopted for these purposes.
