This study aims to select fungi isolates to reduce olive mill wastewater (OMWW) chemical oxygen demand (COD) and phenolic compounds (PC), as well as their genotoxicity effect. Treatment with mold, isolated by an innovative technique using phenolic compound-selective media, showed a reduction rate of about 4% for COD and 2% for PC during one month of incubation without optimization of the treatment conditions. Whereas this percentage reached 98% and 96% for COD and PC, respectively, after only 12 days of treatment, when the C:N ratio was adjusted to 30 by adding urea as a nitrogen source at 150 rpm agitation speed. Genetic sequence homology of the most efficient mold isolate showed 100% similarity to Penicillium chrysogenum. High-performance liquid chromatography analysis of phenolic extracts of untreated OMWW showed the presence of five compounds—hydroxytyrosol at 1.22 g.L−1, tyrosol at 0.05 g.L−1, caffeic acid at 0.16 g.L−1, p-coumaric acid at 0.05 g.L−1 and oleuropein at 0.04 g.L−1—that were eliminated during the degradation process at 88.82%. Genotoxicity, assessed by the Vicia-faba root cell, showed a significant decrease in micronucleus frequency of about 96% after fungal treatment. These results confirm the positive role of fungal treatment of OMWW to eliminate genotoxicity and their ability to improve the agronomic potential. Full article

Two field experiments were conducted at Kafr El Sheikh Gov., Egypt, during two winter growing seasons (2020/2021 and 2021/2022). The objective of this study was to improve some chemical and physical properties of soil and the yield and water productivity of faba beans (Viciafaba L.), Cv. Sakha-4 by application of gypsum, compost, and some nanoparticles in salt-affected soils. The experimental treatments were arranged in a split-plot design with three replications. The main plots had the following soil amendments: T1: control treatment, T2: 10 tons compost/hectare, T3: soil gypsum requirement (GR) of 8.59 ton ha−1, and T4: GR + 10 tons compost/hectare. The subplots were treated with foliar application as follows: no treatment, manganese nanoparticles (Mn-NPs), selenium nanoparticles (Se-NPs), and Mn-NPs + Se-NPs. According to the findings, the application of compost + GR significantly decreased soil salinity (EC), exchangeable sodium percentage (ESP), and soil bulk density (BD). However soil porosity, soil penetration resistance (SPRa), and basic soil infiltration (IR) were significantly increased. On the other hand, the results revealed significant positive effects onthe 100-grain weight as well as proline, chlorophyll, superoxide dismutase, and catalase contents due to the interaction between gypsum + compost and Mn-NPs + Se-NPs, which enhanced the productivity of both the seed and straw yields of faba beans compared to the alternative treatments. In addition, the seed yield and irrigation water productivity (PIW, kg m3) of faba beans were significantly increased with addition of gypsum and compost and foliar application of nanoparticles. The highest values of these parameters were achieved due to the interaction between gypsum + compost and Mn-NPs + Se-NPs. It can be concluded that application of GR of 8.59 ton ha−1 and 10 ton ha−1 compost as well as foliar application of Mn-NPs and Se-NPs may be a key strategy for improving some chemical and physical properties of soil and the yield and water productivity of faba beans in salt-affected soil under these experimental conditions. Green pesticides are highly desirable, as they are environmentally friendly and efficient. In this study, the antifungal peptide P852 was employed to suppress Fusarium wilt in the Faba bean. The disease index and a range of physiological and metabolomic analyses were performed to explore the interactions between P852 and the fungal disease. The incidence and disease index of Fusarium wilt were substantially decreased in diseased Faba beans that were treated with two different concentrations of P852 in both the climate chamber and field trial. For the first time, P852 exhibited potent antifungal effects on Fusarium in an open field condition. To explore the mechanisms that underlie P852′s antifungal effects, P852 treatment was found to significantly enhance antioxidant enzyme capacities including guaiacol peroxidase (POD), superoxide dismutase (SOD), catalase (CAT), and the activities of antifungal enzymes including chitinase and β-1,3-glucanase, as well as plant dry and fresh weights, and chlorophyll content compared to the control group (p ≤ 0.05). Metabolomics analysis of the diseased Faba bean treated with P852 showed changes in the TCA cycle, biological pathways, and many primary and secondary metabolites. The Faba bean treated with a low concentration of P852 (1 μg/mL, IC50) led to upregulated arginine and isoquinoline alkaloid biosynthesis, whereas those treated with a high concentration of P852 (10 μg/mL, MFC) exhibited enhanced betaine and arginine accumulation. Taken together, these findings suggest that P852 induces plant tolerance under Fusarium attack by enhancing the activities of antioxidant and antifungal enzymes, and restoring plant growth and development. Strategic implementation of vermicompost as safe biofertilizer besides defensing saline soils offer dual function solving problems in developing countries. The current study aims to utilize vermicompost (VC) for amelioration of 200mM NaCl in Vicia faba Aspani cultivar and investigate the molecular role of salt overly sensitive pathway (SOS1). The experiment was conducted following a completely randomized design with three replicates. Treatments include 0; 2.5; 5; 10; 15% dried VC intermingled with soil mixture (clay: sand; 1:2) and/or 200 mM NaCl. The results show that salinity stress decreased broad bean fresh and dry weight; and K+/Na+. However, malonedialdehyde and H2O2 contents; increased. Application of 10% VC and salinity stress increases Ca2+ (41% and 50%), K+/Na+ (125% and 89%), Mg2+ (25% and 36%), N (8% and 11%), indole acetic acid (70% and 152%) and proteins (9% and 13%) for root and shoot, respectively, in comparison to salt treated pots. Moreover, all examined enzymatic antioxidants and their substrates increased, except glutathione reductase. A parallel decrease in abscisic acid (75% and 29%) and proline (59% and 58%) was also recorded for roots and leaves, respectively. Interestingly, the highly significant increase in gene expression of SOS1 (45-fold) could drive defense machinery of broad bean to counteract 200 mM NaCl. Full article
