Varietal volatile compounds are characteristic of each variety of grapes and come from the skins of the grapes. This work focuses on the development of a methodology for the analysis of free compounds in grapes from Trincadeira, Cabernet Sauvignon, Syrah, Castelão and Tinta Barroca from the 2021 and 2022 harvests, using HS-SPME-GC × GC-TOFMS. To achieve this purpose, a previous optimization step of sample preparation was implemented, with the optimized conditions being 4 g of grapes, 2 g of NaCl, and 2 mL of H2O. The extraction conditions were also optimized, and it was observed that performing the extraction for 40 min at 60 °C was the best for identifying more varietal compounds. The fiber used was a triple fiber of carboxen/divinylbenzene/polydimethylsiloxane (CAR/DVB/PDMS). In addition to the sample preparation, the analytical conditions were also optimized, enabling the adequate separation of analytes. Using the optimized methodology, it was possible to identify fifty-two free volatile compounds, including seventeen monoterpenes, twenty-eight sesquiterpenes, and seven C13-norisoprenoids. It was observed that in 2021, more free varietal volatile compounds were identifiable compared to 2022. According to the results obtained through a linear discriminant analysis (LDA), the differences in volatile varietal signature are observed both among different grape varieties and across different years. With the rising prominence of organic farming systems in European Union countries, motivated by agricultural policies, there is pressure for effective disease management strategies. To address this challenge, the use of plant resistance inducers (PRIs) and elicitors has emerged as a promising approach. In this study, we compared the impact of integrated production with organic agriculture farming practices, specifically applying PRIs and elicitors in the latter, on the expression levels of stress-responsive genes in two grapevine cultivars, ‘Alicante Bouschet’ and ‘Trincadeira’. Our findings revealed that the organic farming system led to upregulation of eight of the 12 studied genes in at least one cultivar, indicating a significant influence of production mode. The upregulated genes were associated with plant stress-responsive genes (PR1, PR2, PR4, and TLP9), sugar metabolism (HT5), phenylpropanoids (STS1), enzymes related to jasmonic acid synthesis and response to biotic stresses, respectively (LOX, PER42). Also, the ‘Alicante Bouschet’ cultivar consistently displayed significantly higher levels of transcript accumulation on most of the stress-related genes compared to the ‘Trincadeira’ cultivar in both production modes. Our study provides valuable insights into the effectiveness of PRIs and elicitors in increasing plant expression levels of stress-responsive genes, leading to greater resilience to pathogen attacks and emphasizing their position in organic agriculture. Adaptation strategies in the wine sector consist of the use of cultural techniques to limit damages caused by climate change, using, among other resources, varieties better adapted to the scenarios of abiotic stress exacerbation, namely water and thermal stress, as well as those more tolerant to heatwaves. With the intention to determine the aromatic characterization of ten monovarietal wines produced from cultivars with high productive performance in a global warming scenario (‘Petit Verdot’, ‘Marselan’, ‘Merlot’, ‘Touriga Franca’, ‘Syrah’, ‘Vinhão’, ‘Bobal’, ‘Preto Martinho’, ‘Trincadeira’, and ‘Alicante Bouschet’), grown in Esporão vineyard (Alentejo, Portugal) and submitted to deficit irrigation (Ks ± 0.5), their aromatic character has been analyzed. Each grape variety was vinified at a small scale, in duplicate, and the wines were evaluated by a sensory panel, which rated several sensory attributes (visual, olfactory, and gustatory). Sensory analysis revealed a discrete appreciation for the monovarietal wines tasted, showing a differentiation at the olfactory level that was not too marked, although present, between the samples. The free volatile compounds were analysed using gas chromatography-olfactometry (GC-O), identified using a gas chromatography-mass spectrometry (GC-MS) technique and semi-quantified using the gas chromatography-flame ionization detector (GC-FID) technique. Based on the interpolation of the results of the various statistical analyses carried out, 49 probable odor active compounds (pOACs) were identified and based on the odor activity values (OAVs), 24 of them were recognized as odor active compounds (OACs) originated mainly during the fermentation processes. An aromatic characterization of the varieties has been proposed. Full article

Grapevine trunk diseases (GTDs) are among the most important problems that affect the longevity and productivity of vineyards in all the major growing regions of the world. They are slow-progression diseases caused by several wood-inhabiting fungi with similar life cycles and epidemiology. The simultaneous presence of multiple trunk pathogens in a single plant together with the inconsistent GTDs symptoms expression, their isolation in asymptomatic plants, and the absence of effective treatments make these diseases extremely complex to identify and eradicate. Aiming to gain a better knowledge of GTDs and search sustainable alternatives to limit their development, the present work studied the fungal community structure associated with GTDs symptomatic and asymptomatic grapevines, following a metagenomic approach. Two important cultivars from the Alentejo region with different levels of susceptibility to GTDs were selected, namely, ‘Alicante Bouschet’ and ‘Trincadeira’. Deep sequencing of fungal-directed ITS1 amplicon led to the detection of 258 taxa, including 10 fungi previously described as responsible for GTDs. Symptomatic plants exhibited a lower abundance of GTDs-associated fungi, although with significantly higher diversity of those pathogens. Our results demonstrated that trunk diseases symptoms are intensified by a set of multiple GTDs-associated fungi on the same plant. The composition of fungal endophytic communities was significantly different according to the symptomatology and it was not affected by the cultivar. This study opens new perspectives in the study of GTDs-associated fungi and their relation to the symptomatology in grapevines. Full article

Quality Assessment of Red Wine Grapes through NIR Spectroscopy

Red wine grapes require a constant follow-up through analytical chemistry to assure the greatest wine quality. Wet chemical procedures are time-consuming and produce residues that are hard to eliminate. NIR (near infrared radiation) spectroscopy has been referred as an accurate, rapid, and cost-efficient technique to evaluate quality in many fruit species, both in field and in industry. The main objective of this study was to develop predictive models using NIR spectroscopy to quantify important quality attributes in wine grapes. Soluble solids content (SSC), titratable acidity (TA), total phenolic content, total flavonoids, total anthocyanins, and total tannins were quantified in four red wine grape varieties, ‘Aragonês’, ‘Trincadeira’, ‘Touriga Nacional’, and ‘Syrah’. Samples were collected during 2017 and 2018 along véraison. Prediction models were developed using a near-infrared portable device (Brimrose, Luminar 5030), and spectra were collected from entire grapes under near field conditions. Models were built using a partial least square regression (PLSR) algorithm and SSC, TA, total anthocyanins, and total tannins exhibited a determination coefficient of 0.89, 0.90, 0.87, and 0.88, respectively. The Residual Prediction Deviation (RPD) values of these models were higher than 2.3. The prediction models for SSC, TA, total anthocyanins, and total tannins have considerable potential to quantify these attributes in wine grapes. Total flavonoids and total phenolic content were predicted with a slightly lower capacity, with R2 = 0.72 and 0.71, respectively, and both with a RPD of 1.6, indicating a very low to borderline potential for quantitative predictions in flavonoids and phenols models. Volatile Metabolism of Wine Grape Trincadeira: Impact of Infection with Botrytis cinerea

The aroma of grapes is cultivar dependent and is influenced by terroir, vineyard practices, and abiotic and biotic stresses. Trincadeira is a non-aromatic variety associated with low phenolic content and high sugar and organic acid levels. This cultivar, widely used in Portuguese wines, presents high susceptibility to Botrytis cinerea. This work aimed to characterise the volatile profile of Trincadeira grapes and how it changes under infection with B. cinerea. Thirty-six volatile organic compounds were identified, from different functional groups, namely alcohols, ester acetates, fatty acid esters, fatty acids, aldehydes, and products of the lipoxygenase pathway. Both free and glycosidic volatile organic compounds were analysed by Gas Chromatography and Gas Chromatography coupled to Mass Spectrometry for component quantification and identification, respectively. A multivariance analysis showed a clear discrimination between healthy and infected grapes with 2-trans-hexenal and isoamyl-acetate among the compounds identified as negative and positive markers of infection, respectively. Ester acetates such as 2-phenylethyl acetate, isoamyl acetate, and 2-methylbutyl acetate were present in higher contents in infected samples, whereas the contents of several fatty acid esters, such as ethyl decanoate and ethyl dodecanoate, decreased. These data were integrated with quantitative PCR data regarding genes involved in volatile metabolism and showed up-regulation of a gene coding for Hydroperoxide Lyase 2 in infected grapes. Altogether, these changes in volatile metabolism indicate an impact on the grape quality and may be related to defence against B. cinerea. The presence/absence of specific compounds might be used as infection biomarkers in the assessment of Trincadeira grapes’ quality. Full article
