Reference intervals for clinical chemistry and haematology analytes are essential for the interpretation of blood results, but limited data are available for Humboldt penguins (Spheniscus humboldti) in zoological collections as well as in the wild. The aim of the present study was therefore to establish reference intervals for a series of clinical chemistry and haematology analytes in a single zoological population of Humboldt penguins. Furthermore, possible variations of the analytes influenced by the age and sex of the individuals were investigated. Lithium heparinized plasma and whole blood samples from 39 animals were analysed and statistically evaluated. The following clinical chemistry analytes were significantly higher in females: glutamate dehydrogenase (p = 0.0003), alanine transaminase (p = 0.0005), alkaline phosphatase (p = 0.0245), aspartate aminotransferase (p = 0.0043), creatine kinase (p = 0.0016), lactate dehydrogenase (p < 0.0001), inorganic phosphorus (p = 0.0223), and sodium (p = 0.0415). No significant differences between males and females were found for any haematology analyte. Cholesterol (p = 0.0233; −0.39), white blood cell count (p = 0.0208; −0.40), and absolute heterophiles (p = 0.0148; −0.42) had significant negative correlations with the age of the penguins. The results indicate that the factors of sex and age should be considered when interpreting the results of haematological and clinical biochemistry assays, and further studies are needed to determine whether there are more differences in other age groups or seasons. Pollution from plastic waste thrown into the ocean affects all levels of the food chain. Marine species of birds are affected by plastic particles of different sizes, especially the mesoplastics (1 to 10 mm) found in their digestive tract, which mainly cause obstructions. In the case of microplastics (1.000 µm to 1 mm), their presence in the digestive tract of these species has been widely reported. We studied fecal samples of the Dominican gull (Larus dominicanus) (n = 14), Magellanic penguins (Spheniscus magellanicus) (n = 8), and Humboldt penguin (Spheniscus humboldti) (n = 1) obtained from the Wildlife Rehabilitation Center of the Biobío region, Chile. Microfibers of various colors were present in the feces of Dominican gulls and Magellanic penguins, corresponding mainly in composition to polypropylene (PP) (83%) and rayon (77%). These results demonstrate that microplastic particles occur in the coastal environments of central Chile and suggest that they are probably circulating in the food chain. Full article

Aspergillus fumigatus is the main causative agent of avian aspergillosis and results in significant health problems in birds, especially those living in captivity. The fungal contamination by A. fumigatus in the environment of Humboldt penguins (Spheniscus humboldti), located in a Belgian zoo, was assessed through the analysis of air, water, sand and nest samples during four non-consecutive days in 2021–2022. From these samples, potential azole-resistant A. fumigatus (ARAF) isolates were detected using a selective culture medium. A total of 28 veterinary isolates obtained after necropsy of Humboldt penguins and other avian species from the zoo were also included. All veterinary and suspected ARAF isolates from the environment were characterized for their azole-resistance profile by broth microdilution. Isolates displaying phenotypic resistance against at least one medical azole were systematically screened for mutations in the cyp51A gene. A total of 14 (13.6%) ARAF isolates were identified from the environment (n = 8) and from Humboldt penguins (n = 6). The TR34/L98H mutation was observed in all resistant environmental strains, and in two resistant veterinary strains. To the best of our knowledge, this is the first description of this mutation in A. fumigatus isolates from Humboldt penguins. During the period 2017–2022, pulmonary aspergillosis was confirmed in 51 necropsied penguins, which reflects a death rate due to aspergillosis of 68.0%, mostly affecting adults. Microsatellite polymorphism analysis revealed a high level of diversity among environmental and veterinary A. fumigatus isolates. However, a cluster was observed between one veterinary isolate and six environmental strains, all resistant to medical azoles. In conclusion, the environment of the Humboldt penguins is a potential contamination source of ARAF, making their management even more complex. Full article

To mitigate the effects of anthropogenic environmental impacts, conservation education (CE) aims to increase public awareness of biodiversity and encourage the uptake of pro-environmental behaviours (PEB). One way of achieving this goal is the use of controlled animal interactions with animal ambassadors to promote CE goals. This study aimed to determine how differing experiences with ambassador Humboldt penguins (Spheniscus humboldti) affected visitors’ knowledge of conservation actions (CA) and their likelihood to perform PEBs in a UK zoo. A quasi-experimental repeated measures questionnaire was used to investigate this aim. Visit types were categorised as (a) ‘penguin parade’, (b) ‘animal encounter’, (c) ‘exhibit viewing’ and (d) a ‘control’ groups. A Kruskal–Wallis test was used to determine if responses to rating scale items related to PEBs differed according to experience. No significant (p > 0.05) differences were detectable between groups. Qualitative data provided in open ended questions relating to CAs were coded into themes and the percentage of responses were calculated for each theme. Whilst the ‘exhibit viewing’ group mentioned sustainable fish significantly more (p = 0.017) than the ‘control’ group, no significant difference was found for any other pair-wise comparison (p > 0.05 in all cases). Overall, this study highlights that zoo visitors gained specific conservation-based knowledge and had higher behavioural intentions towards PEBs than they previously had. However, no evidence was found to suggest that animal encounters improved CE outcomes. To ensure best practice is followed and CE outcomes are considered equally alongside ambassador animal welfare concerns, further research is recommended. Full article

Penguins are considered among the most popular animals for zoo and aquarium visitors to observe. Swimming is considered a desirable activity, both for the visitor experience and the welfare of the penguins. However, little is known about the amount of time exhibited penguins spend swimming, or how such swimming is related to regular feeding events. We examined the effects of introducing live prey in the form of trout on 22 Humboldt penguins living at the Woodland Park Zoo. Of primary interest was how the live feeds changed (1) daily and hourly swimming activity, and (2) variability in enclosure use. We hypothesized that the live feedings would increase swimming activity prior to and during the delivery of the live trout, as well as create an overall increase in total swimming activity for live feed days compared to non-live feed days. We also predicted that the penguins would be more likely to use the entire exhibit around these live feeds, since they are likely to chase fish throughout the exhibit. Penguins did show an increase in swimming activity in the hour prior to and during the live feed, with a small decrease in swimming activity following the live feed when compared to non-live feed days. There was also a more than 30% increase in the total swimming activity for live feed days when compared to all other non-live feed days. In addition, a single measure of variability in enclosure use (entropy) showed greater overall enclosure use for the live feed days compared to the non-live feed days. These results demonstrate that live fish can be a useful way of enriching the behavioural welfare of Humboldt penguins. Full article

It is currently unknown if current guidelines for collecting and interpreting blood corticosterone in flying birds can be extrapolated to penguins. It is also difficult to collect blood quickly without causing stress to a penguin. Therefore, immunoreactive fecal glucocorticoid metabolites (FGCMs) may be the most practical and minimally invasive way of monitoring the stress levels of penguins. This study investigated the reliability of FGCMs for monitoring stress levels in captive Humboldt Penguins (Spheniscus humboldti) at Jurong Bird Park, Singapore. Humboldt Penguin feces were randomly sampled and pooled from the exhibit for 2 months. The penguins were restrained and transported on three separate occasions to simulate stressful events. The feces were analyzed using an enzyme immunoassay to measure corticosterone levels. There were significant increases lasting 3 to 7 days in the FGCM levels after a stressful event. This method was then used to test whether accelerometer vests used for behavior quantification caused stress responses in these birds. There was no significant difference in FGCMs between time periods with and without the accelerometer vests. The results indicated that FGCMs can be an accurate indication of capture-, restraint-, and transport-related stress in Humboldt Penguins, and that accelerometer vests do not appear to cause stress. We examined 185 metabolites in 30 adult Humboldt Penguins (Spheniscus humboldti) nesting at the Punta San Juan Marine Protected Area, Peru, in order to examine gender differences in metabolome profiles, particularly those involved in metabolism and energetics. The majority of the compounds identified were fatty (26% of total identified compounds), organic (19%), and amino (16%) acids. We were able to differentiate male and female penguins with 96.6% accuracy on the basis of 12 metabolites, most of which are involved in lipid and carbohydrate metabolism. These included 2-oxoglutarate, erythronic acid, GABA, mannitol, sedoheptulose 7-phosphate, and serine and six metabolites present in higher concentrations in females compared to males (2-aminoadipic acid, O-phosphorylethanolamine, glycerol 2-phosphate, glycerol 3-phosphate, pantothenic acid, and creatinine). Of these, 2-oxoglutarate and glycerol 3-phosphate were key metabolites distinguishing gender. Our results indicated that male and female Humboldt Penguins were characterized by differing metabolic states. Such differences could be important to individual and brood survival in times of environmental stress. Full article
