Mineral Supplementation in Jade Perch (Scortum barcoo) Aquaponics with Lettuce: A Comparison with Hydroponics and RAS

The global demand for local and high-quality food sources has increased in recent decades, driven by consumer preferences and a growing population. RAS, hydroponics and a ‘hybrid’ version, aquaponics, are food production techniques that could support such growing demand. The current study evaluated coupled aquaponics (Jade perch and lettuce) with and without mineral supplementation in comparison to standard cultivation methods, i.e., RAS and hydroponics. The mineral supplementation in aquaponics was carried out using a hydroponics commercial blend fertilizer (10% of the dosage utilised in the hydroponics treatment). Fish/plant performance, nutrient dynamics and water quality were evaluated for 9 weeks using twelve experimental units (~720 L in total; fish tank/reservoir + filtration systems/sump + floating raft-based plant tank). After two crops of plants, the results showcased the positive impact of mineral supplementation in aquaponics; however, the hydroponics treatment outperformed the two aquaponics treatments, e.g., with higher total plant wet weight and total plant length (p < 0.05). Fish in aquaponics with and without mineral supplementation presented similar growth performance as compared to RAS (p > 0.05). By comparing these different farming approaches, this study sought to provide insights into optimising aquaponics systems and advancing the integration of Jade perch into aquaculture practices. Jade perch (Scortum barcoo) has an XX/XY sex-determination system (SDS); however, its sex chromosomes and sex-determining region remain unknown. The recent availability of the jade perch chromosome-level genomic data provides a valuable resource for pinpointing the location of functional genes and the whole genomic structure. In this study, we conducted. In silico chromosome mapping of male-specific/linked loci of jade perch and identified a potential 11.18 Mb male-linked region localized on chromosome 19 (SBA19). Repeat annotations of the male-linked region revealed an abundance of transposable elements, particularly Ty3/Gypsy and novel repeats. Sequence analysis of this region identified a remnant of amh gene, which is considered a potential candidate for SDS in many teleosts. A duplicate copy of amh remnant was located at SBA6. These duplicated amh copies were highly similar to those of XX/XY SDS in teleosts, in which one copy of amh was identified on the Y sex chromosome. Taken all together, we hypothesize SBA19 as the putative sex chromosome and the 11.18 Mb male-linked region to be a potential male-determining region. Full article

This study investigated the effects of different protein sources and butyric acid contents in aquafeed on the growth, survival rate, lipid peroxidation enzymes, and intestinal villi of 7.3 g jade perch, Scortum barcoo. The experimental treatment groups were the whole fish meal protein (FM) group, the FM + butyric acid (FMB) group, the 50% fish meal protein and 50% black soldier fly meal protein (FBM) group, the FBM + butyric acid (FBMB) group, the whole black soldier fly meal protein (BM) group, and the BM + butyric acid (BMB) group. The experimental results showed that the jade perch fed with the FMB feed had a significantly higher weight gain rate and antioxidant enzyme activity than the other treatment groups. The red blood cell count, hemoglobin concentration, and hematocrit contents in the blood of jade perch were not affected by the protein source, but they increased with the addition of butyric acid. The intestinal villi length of the jade perch that were fed diets containing butyric acid were significantly higher than that of the jade perch that were fed diets without butyric acid supplementation. The results of this study show that the addition of butyric acid to a balanced feed composed of protein can contribute to the growth of jade perch. Full article

Due to the high meat yield and rich nutritional content, jade perch (Scortum barcoo) has become an important commercial aquaculture species in China. Jade perch has a slow growth rate, taking 3–4 years to reach sexual maturity, and has almost no difference in body size between males and females. However, the study of its gonad development and reproduction regulation is still blank, which limited the yield increase. Herein, the gonad transcriptomes of juvenile males and females of S. barcoo were identified for the first time. A total of 107,060 unigenes were successfully annotated. By comparing male and female gonad transcriptomes, a total of 23,849 differentially expressed genes (DEGs) were identified, of which 9517 were downregulated, and 14,332 were upregulated in the testis. In addition, a large number of DEGs involved in sex differentiation, gonadal development and differentiation and gametogenesis were identified, and the differential expression patterns of some genes were further verified using real-time fluorescence quantitative PCR. The results of this study will provide a valuable resource for further studies on sex determination and gonadal development of S. barcoo. Full article
