Background: Ophioglossum vulgatum Linn. is a medical herb widely distributed in Southwest China. It has been used for the treatment of various diseases, including wounds or dermatitis, since ancient times, but little is known about its pharmacological and pharmaceutical chemistry. Methods: The ethyl acetate fraction of O. vulgatum (OpvE) was prepared with the reflex extraction and fractional extraction method. Its components were detected and identified with the UPLC-Q/TOF-MS system and the SCIEX OS database. The related biological activities and the underlying mechanisms were predicted by computational analysis. HaCaT cells were treated with gradient concentrations of OpvE, and a CCK-8 assay was performed to determine the cell viability. The OpvE-pretreated HaCaT cells were exposed to H2O2 or LPS for antioxidative and anti-inflammatory assessment. DPPH, GSH, SOD, and MDA kits were used to evaluate oxidative stress. A serially diluted microbiota assay and a disc diffusion assay were used to evaluate anti-Staphylococcus aureus activities. The transcription of genes was semi-quantitatively studied by reversed real-time PCR. Protein levels were determined with western blotting. Results: The extract ratio of OpvE was 2.00 ± 0.12% (g/g). A total of 21 ingredients were identified. The computational analysis found that the PI3K/Akt signaling pathway might be a crucial target of OpvE. OpvE (7.5~125 μg/mL) stimulated HaCaT cell proliferation and migration by stimulating the over-expressed collagen type I alpha 1 Chain (COL1A1) and fibronectin 1 (FN1) and upregulating PI3K/AKT/GSK3-β signaling pathway. In the antioxidative assay test, 250 μg/mL OpvE scavenged obvious 97.28% DPPH-released free radicals. Pretreatment of OpvE inhibited H2O2-induced oxidative stress and protected against LPS-induced inflammatory injury by respectively regulating the Nrf2/HO-1/COX2 and TLR4/MYD88 signaling pathways. OpvE also showed anti-S. aureus properties with a MIC of 1.2 mg/mL, and with this concentration, OpvE produced an 8.3 ± 0.16 mm inhibition zone on a bacterial plate. Conclusions: This work highlighted the phytochemical character and some bioactivities, as well as the underline mechanism, which would support the further studies and application of O. vulgatum Linn. Many plant mitochondrial (mt) genomes have been sequenced but few in ferns. Ophioglossum vulgatum represents a typical species of fern genus Ophioglossum with medicinal and scientific value. However, its mt genome structure remains to be characterized. This study assembled and annotated the complete O. vulgatum mt genome and presented its structural characters and repeat sequences firstly. Its mt and chloroplast (cp) transfer sequences were explored, and the phylogenetic significance of both mt and cp genomes was also evaluated at the family level. Our results showed that the complete mt genome of O. vulgatum is a single circular genome of 369,673 bp in length, containing 5000 dispersed repetitive sequences. Phylogenetic trees reconstructed from cp and mt genomes displayed similar topologies, but also showed subtle differences at certain nodes. There exist 4818 bp common gene fragments between cp and mt genomes, of which more than 70% are located in tRNA intergenic regions (in mt). In conclusion, we assembled the complete mt genome of O. vulgatum, identified its remarkable structural characters, and provided new insights on ferns. The complementary results derived from mt and cp phylogeny highlighted that some higher taxonomic-level phylogenetic relationships among ferns remain to be resolved. Full article

The distribution of the threatened fern Ophioglossum vulgatum L., a plant with extremely small populations (PSESPs) in Sardinia, is characterized by small disjunct populations with only a few individuals, and little is known about its status in the wild. To provide information for the conservation of O. vulgatum and with the aim to develop an in situ conservation strategy, we investigated its distribution, population size, and habitat. Field surveys confirmed that the species grows in only five localities. Two representative populations were selected for this study (Funtanamela and Gedili), and in each population, all plants were mapped and monitored monthly from April to August over an 8-year period. During the study, the populations had a very low number of reproductive plants and the populations appeared to be in decline, with the total number of plants per population slightly decreased in Gedili while a sharp reduction was recorded in Funtanamela due to wild boar threat. A fence was built in order to protect the site from further damage, but no noticeable signals of recovery were observed. The most urgent conservation requirement for this species is to preserve the threatened habitat of the remnant populations. Further field surveys and research are also required for an improved understanding of the species’ status. Full article

In an extensive screening, the antiviral, antitrypanosomal and anticancer properties of extracts from 82 plants used in traditional Chinese medicine and European phytomedicine were determined. Several promising plants that were highly effective against hepatitis B virus (HBV), bovine viral diarrhoea virus (BVDV)—a flavivirus used here as a surrogate in vitro model of hepatitis C virus, trypanosomes (Trypanosoma brucei brucei) and several cancer cell lines were identified. Six aqueous extracts from Celosia cristata, Ophioglossum vulgatum, Houttuynia cordata, Selaginella tamariscina, Alpinia galanga and Alpinia oxyphylla showed significant antiviral effects against BVDV without toxic effects on host embryonic bovine trachea (EBTr) cells, while Evodia lepta, Hedyotis diffusa and Glycyrrhiza spp. demonstrated promising activities against the HBV without toxic effects on host human hepatoblastoma cells transfected with HBV-DNA (HepG2 2.2.15) cells. Seven organic extracts from Alpinia oxyphylla, Coptis chinensis, Kadsura longipedunculata, Arctium lappa, Panax ginseng, Panax notoginseng and Saposhnikovia divaricata inhibited T. b. brucei. Moreover, among fifteen water extracts that combined high antiproliferative activity (IC50 0.5–20 µg/mL) and low acute in vitro toxicity (0–10% reduction in cell viability at IC50), Coptis chinensis presented the best beneficial characteristics. In conclusion, traditional herbal medicine from Europe and China still has a potential for new therapeutic targets and therapeutic applications. Full article
