Amyloid-Beta Peptides Trigger Premature Functional and Gene Expression Alterations in Human-Induced Neurons

Alzheimer’s disease (AD) is the most prevalent cause of dementia in the elderly, characterized by the presence of amyloid-beta (Aβ) plaques, neurofibrillary tangles, neuroinflammation, synapse loss and neurodegeneration in the brain. The amyloid cascade hypothesis postulates that deposition of Aβ peptides is the causative agent of AD pathology, but we still lack comprehensive understanding of the molecular mechanisms connecting Aβ peptides to neuronal dysfunctions in AD. In this work, we investigate the early effects of Aβ peptide accumulation on the functional properties and gene expression profiles of human-induced neurons (hiNs). We show that hiNs acutely exposed to low concentrations of both cell-secreted Aβ peptides or synthetic Aβ1–42 exhibit alterations in the frequency of calcium transients suggestive of increased neuronal excitability. Using single-cell RNA sequencing, we also show that cell-secreted Aβ up-regulates the expression of several synapse-related genes and down-regulates the expression of genes associated with metabolic stress mainly in glutamatergic neurons and, to a lesser degree, in GABAergic neurons and astrocytes. These neuronal alterations correlate with activation of the SEMA5, EPHA and NECTIN signaling pathways, which are important regulators of synaptic plasticity. Altogether, our findings indicate that slight elevations in Aβ concentrations are sufficient to elicit transcriptional changes in human neurons, which can contribute to early alterations in neural network activity. We coupled together high-resolution versions of the ocean–sea ice model NEMO and the ice sheet model BISICLES configured to the Totten Glacier area and ran a series of simulations over the recent past (1995–2014) and under warming conditions (2081–2100; SSP4-4.5) with NEMO in stand-alone mode and with the coupled model to assess the effects of the coupling. During the recent past, the ocean–ice sheet coupling has increased the time-averaged value of the basal melt rate in both the Totten and Moscow University ice shelf cavities by 6.7% and 14.2%, respectively. The relationship between the changes in ice shelf thickness and ice shelf basal melt rate suggests that the effect of the coupling is not a linear response to the melt rate but rather a more complex response, driven partly by the dynamical component of the ice sheet model. The response of the ice sheet–ocean coupling due to the ocean warming is a 10% and 3% basal melt rate decrease in the Totten and Moscow University ice shelf cavities, respectively. This indicates that the ocean–ice sheet coupling under climate warming conditions dampens the basal melt rates. Our study highlights the importance of incorporating ocean–ice sheet coupling in climate simulations, even over short time periods. Full article

Pest Activity and Protection Practices: Four Decades of Transformation in Quebec Apple Orchards

A group of commercial orchards from Quebec (Canada) was followed from 1977 to 2019 as part of a project to implement Integrated Pest Management (IPM) practices. Collected data comprised activity of major fruit pests (from monitoring traps), fruit damage at harvest and pesticide applications, from which the annual costs and impacts of protection programs over 42 years were calculated. Activity and fruit damage in commercial orchards were compared to patterns observed in a reference insecticide-free orchard. Some insects (European apple sawfly, codling moth, apple maggot) were more prevalent in the insecticide-free orchard than in commercial orchards, while others were more prevalent in commercial orchards (oblique-banded leafroller) or as prevalent in both orchard types (tarnished plant bug). Annual fruit damage in the insecticide-free orchard was mostly from the apple maggot (up to 98%), the plum curculio (up to 90%) and the codling moth (up to 58%). The average situation was different in commercial orchards, whose damage was mostly from the plum curculio (up to 7.6%), the tarnished plant bug (up to 7.5%) and the oblique-banded leafroller (up to 1.7%). While the number of registered pesticides, the number of applications and the total cost of pesticides gradually increased from 2002 to 2019, the risks incurred, as measured by indicators of environmental and health impacts, followed a downward trend for insecticides and acaricides and varied slightly for fungicides. Several instruments have been developed by clinicians and academics to assess clinical recovery. Based on their life narratives, measurement tools have also been developed and validated through participatory research programs by persons living with mental health problems or illnesses to assess personal recovery. The main objective of this project is to explore possible correlations between clinical recovery, personal recovery, and citizenship by using patient-reported outcome measures. All study participants are currently being treated and monitored after having been diagnosed either with (a) psychotic disorders or (b) anxiety and mood disorders. They have completed questionnaires for clinical evaluation purposes (clinical recovery) will further complete the Recovery Assessment Scale and Citizenship Measure (personal-civic recovery composite index). Descriptive and statistical analyses will be performed to determine internal consistency for each of the subscales, and assess convergent-concurrent validity between clinical recovery, citizenship and personal recovery. Recovery-oriented mental health care and services are particularly recognizable by the presence of Peer Support Workers, who are persons with lived experience of recovery. Upon training, they can personify personalized mental health care and services, that is to say services that are centered on the person’s recovery project and not only on their symptoms. Data from our overall research strategy will lay the ground for the evaluation of the effects of the intervention of Peer Support Workers on clinical recovery, citizenship and personal recovery. The objectives were to assess whether social support for healthy eating and perceived food environment are associated with diet quality, and to investigate if sociodemographic characteristics moderate these associations. A probability sample of French-speaking adults from the Province of Québec, Canada, was recruited in the context of the PREDISE study. Participants reported their perceptions of supportive and non-supportive actions related to healthy eating from close others at home and outside of home (n = 952), and of the accessibility to healthy foods (n = 1035). The Canadian Healthy Eating Index (C-HEI) was calculated based on three Web-based 24 h food recalls. Multiple linear regression models showed that supportive (B = 1.50 (95% CI 0.46, 2.54)) and non-supportive (B = −3.06 (95% CI −4.94, −1.18)) actions related to healthy eating from close others at home were positively and negatively associated with C-HEI, respectively, whereas actions from close others outside of home were not. The negative association between non-supportive actions occurring at home and C-HEI was stronger among participants with lower (vs. higher) levels of education (p interaction = 0.03). Perceived accessibility to healthy foods was not associated with C-HEI (p > 0.05). These results suggest that the social environment may have a stronger influence on healthy eating than the perceived physical environment. This adds support for healthy eating promotion programs involving entire families, especially for more socioeconomically disadvantaged individuals, whose efforts to eat healthily may be more easily thwarted by non-supportive households.
