Round 1

Reviewer 1 Report

General comments:

In this manuscript, the authors have developed GC-MS method for determination of lidocaine in postmortem whole blood using Liquid-Liquid Extraction for sample pre-treatment. The method was applied to real whole blood samples from cases where unsuccessful Cardiopulmonary Resuscitation was attempted.

My remarks and recommendations are as follows:

  1. The introduction should be described in more detail. More references to the existing methods for the determination of lidocaine in postmortem blood samples should be also provided.
  2. Negligible novelty and significance of the research. There is a number of published analytical methods (also GC-MS) for determination of lidocaine in postmortem samples including blood. Nevertheless, the authors did not provide the comparison of the proposed method with already known ones. What is the novelty and advantages of this work compared to papers published earlier? Novelty and significance of the work should be explained and highlighted.
  3. It would be good to give some results of the statistical analysis regarding the results of quantification in real samples (e.g. mean, median, standard deviation).

Additionally:

  1. I don't think the abbreviation of Cardiopulmonary Resuscitation should be used in the title. In my opinion, the full name should be used.
  2. There are some typos in the text. For example: in abstract it should be written ,,Cardiopulmonary Resuscitation’’ not ,,CardiPulmonary Resuscitation’’. The text of manuscript should be carefully checked.

from paper it is not clear what kind of guidelines were used to validate the method

The article describes a method for the detection of lidocaine in postmortem whole blood using LLE followed by GC/MS analysis.

Lidocaine chemical structure should be presented. Pharmacological and pharmacokinetic data about lidocaine are missing in the article. The Introduction section is supefically written. What about other methods of detecting lidocaine in biological matrices ?

Statistical presentation of the results can be improved. Validation data should be included in a table for a more rapid identication by the reader.
