This is a comprehensive review article about a role of FXR in various liver disorders, and covers from basic biology to clinical applications. Also, the authors explain present status of progress of clinical trials of OA, a ligand of FXR, as a pharmacological therapy for PBC and NAFLD. The explanation has been made from a quite neutral standpoint, and looks very fair. I believe that it can satisfy many readers of various backgrounds.

This compelling article discusses the role of the farnesoid X receptor as the true guardian of liver function by multifactorial preserving liver integrity and functionality.

The manuscript is well written, addressing the topic in-depth and with an up-to-date bibliography.

Some aspects that could improve the work would be:

Adding a list of abbreviations.

Sections 2 and 3 have explanatory figures. In Sections 4 and 5, it would be more illustrative if summarized with a graph or a table.

Reference 149 is missing the publication date of the post.
