This paper presents a comprehensive analysis of the adsorbent capacity of five distinctly different biosorbents derived from untreated biomasses. The optimal adsorption capacity of seaweed (Laminaria digitata), horse chestnut husk, hazelnut husk, rapeseed residue, and whitewood to remove methylene blue (MB) dye was assessed by analysing the effects of particle size, pH, temperature, and initial dye concentrations. Furthermore, the adsorption kinetics, isotherms, and adsorption thermodynamics were investigated. The results showed that relatively high MB adsorption capacity was achieved by Laminaria digitata (~180 mg/g), in addition to a reasonable MB adsorption capacity of horse chestnut husk (~130 mg/g), hazelnut husk (~110 mg/g), and rapeseed residue (~80 mg/g). However, whitewood provides a relatively low adsorption capacity of below 20 mg/g. The best fit with experimental results regardless of biosorbent type was a pseudo-second-order kinetic model with the lowest mean absolute percentage error (ε, MAPE < 2.5%) and the highest correlation coefficients (R2 > 0.99). Although the pseudo-second-order kinetic model is often associated with chemisorption, the low enthalpy values (<29.30 kJ/mol) typically suggest that the adsorption process is more characteristic of physisorption, which involves weaker van der Waals forces rather than the stronger covalent bonds of chemisorption. This proposed a multi-step adsorption process involving both physisorption and chemisorption. The adsorption isotherm of Langmuir showed superior fitting results for Laminaria digitata and hazelnut husk. In contrast, rapeseed residue and horse chestnut husk fit better with the Freundlich adsorption isotherm. The Langmuir adsorption isotherms showed a maximum adsorption capacity of ~500 mg/g for Laminaria digitata, followed by horse chestnut husk (~137 mg/g), hazelnut husk (~120 mg/g), and rapeseed residue (~85 mg/g). The Gibbs free energy was negative for Laminaria digitata < horse chestnut husk < hazelnut husk < 0, which suggests that the removal of MB is thermodynamically favourable, as the adsorption process occurs spontaneously. The results of the study indicate that MB dye removal using untreated biomasses has the potential to be a low-cost valorisation option in the holistic whole life cycle valorisation pathway for Laminaria digitata, horse chestnut husk, and hazelnut husk. Opportunities for the valorisation of agro-industrial residues of the chestnut (Castanea sativa Mill.) production chain have been fostered with the production of multifunctional polyphenol-rich extracts with the potential to be introduced as natural additives or active components in several products. Nonetheless, it is crucial to explore the feasibility of different extracts from the various by-products for these applications through the exhaustive study of their composition and bioactivities without losing sight of the sustainable character of the process. This work aimed at the screening of the phenolic compound composition and bioactivities of different green extracts of chestnut burs, shells and leaves, as the first step to establish their potential application as natural ingredients, primarily as food preservatives. To this end, maceration (MAC) as a conventional extraction method besides ultrasound and microwave-assisted extractions (UAE and MAE) was employed to obtain the extracts from chestnut by-products using water (W) and hydroethanolic solution (HE) as solvents. Phenolic compounds were analysed by HPLC-DAD-(ESI-)MS/MS; the antioxidant capacity was assessed by colourimetric assays, and the antimicrobial activity was evaluated against several strains of food-borne bacteria and fungi. The leaf extracts obtained by MAC-HE and UAE-HE presented the highest concentration of phenolic compounds (70.92 ± 2.72 and 53.97 ± 2.41 mg.g−1 extract dw, respectively), whereas, for burs and shells, the highest recovery of total phenolic compounds was achieved by using UAE-HE and UAE-W (36.87 ± 1.09 and 23.03 ± 0.26 mg.g−1 extract dw, respectively). Bis-HHDP-glucose isomers, chestanin and gallic acid were among the most abundant compounds. Bur extracts (MAC-HE and UAE-HE) generally presented the highest antioxidant capacity as measured by TBARS, while the best results in DPPH and reducing power assays were found for shell extracts (MAE-W and MAC-HE). Promising antibacterial activity was noticed for the aqueous extracts of burs, leaves and hydroethanolic extracts of shells, with emphasis on the MAE-W extract of burs that showed bactericidal activity against E. cloacae, P. aeruginosa and S. aureus (MBC 5 mg.mL−1). Overall, it can be concluded that chestnut by-products, including burs, shells and leaves, are sources of polyphenolic compounds with significant antioxidant and antimicrobial activities. The choice of extraction method and solvent greatly influenced the composition and bioactivity of the extracts. These findings highlight the potential of chestnut by-products for the development of natural additives, particularly for food preservation, while also emphasizing the importance of sustainable utilization of agricultural waste materials. Further research is warranted to optimize extraction techniques and explore additional applications for these valuable bioactive compounds. Full article

Castanea sativa is very common in Italy, and the large amount of waste material generated during chestnut processing has a high environmental impact. Several studies demonstrated that chestnut by-products are a good source of bioactive compounds, mainly endowed with antioxidant properties. This study further investigates the anti-neuroinflammatory effect of chestnut leaf and spiny bur extracts, together with the deepest phytochemical characterisation (by NMR and MS) of active biomolecules contained in leaf extracts, which resulted in being more effective than spiny bur ones. BV-2 microglial cells stimulated with lipopolysaccharide (LPS) were used as a model of neuroinflammation. In BV-2 cells pre-treated with chestnut extracts, LPS signalling is partially blocked via the reduced expression of TLR4 and CD14 as well as the expression of LPS-induced inflammatory markers. Leaf extract fractions revealed the presence of specific flavonoids, such as isorhamnetin glucoside, astragalin, myricitrin, kaempferol 3-rhamnosyl (1-6)(2″-trans-p-coumaroyl)hexoside, tiliroside and unsaturated fatty acids, all of which could be responsible for the observed anti-neuroinflammatory effects. Interestingly, the kaempferol derivative has been identified in chestnut for the first time. In conclusion, the exploitation of chestnut by-products is suitable for the achievement of two goals: satisfaction of consumers’ demand for new, natural bio-active compounds and valorisation of by-products. Efficient natural resource management prevents and reduces negative impacts, such as environmental damage, misappropriation of resources, and conflicts; several strategies can be leveraged to conserve, protect, and enhance natural resources. Multicriteria decision analysis (MCDA) is useful in providing solutions to addressing decision choice problems. In this study, the natural resource under evaluation is the chestnut forest, with the objective of valorising its supply chains. The methodology applied is A’WOT, which allows previously identified factors, using a qualitative SWOT matrix, to be ordered through an objective quantification using the AHP (analytic hierarchy process), which is a multicriteria decision support method. The survey was conducted with a group of chestnut resource (n = 20) experts. The SWOT matrix identified a total of 20 factors: 6 strengths and 6 weaknesses and 4 factors each for opportunities and threats. The results express a clear stakeholder interest, which identifies the significant role of civil society in directing management choices for the provision and enhancement of ecosystem and vocational services. This study evaluated the adaptability of decision support tools applied to a real case of forest resource management to identify and order factors useful to enhance the resource and stimulate the supply chains to achieve greater added value. In a general sense, the methodological potential emerged to replicate or improve the research in other geographical regions, whether regional or extraregional, or even on a larger scale, such as on a national level. The chestnut tree has characterised the social history of the Italian hills and mountains. However, in recent years, this resource has been abandoned because of the change in socio-economic needs. The aim of this study is twofold: (i) To give an overview of this resource, assessing the strengths-weaknesses-opportunities-threats of the chestnut tree through the use of the SWOT methodology; (ii) to provide action strategies for resource valorisation, in accordance with the SDGs set out in the UN 2030 Agenda, by using the TOWS methodology. Our results suggest four strategies, specifically; ‘resource-based’, ‘network-based’, ‘policy-based’ and ‘market-based’, capable of valorising the resource from environmental, economic and societal points of view. The active management of chestnut forests, together with the development of a value chain can guarantee sustainable production, technological and process innovation and could empower companies to face global markets in a more competitive way and lead to the creation of high value-added products, generating a strong economic and social engine for the revival of the sector. Moreover, the commitment of the private sector to the reorganisation of the supply chain must also be accompanied by coherent public policy action. This is essential for the adoption of plans and programmes consistent with sustainable development objectives, ensuring their prioritisation and implementation. Full article
