Pseudomonas putida strain U can be grown using, as sole carbon sources, the biogenic amines putrescine or cadaverine, as well as their catabolic intermediates, ɣ-aminobutyrate or δ-aminovalerate, respectively. Several paralogs for the genes that encode some of the activities involved in the catabolism of these compounds, such as a putrescine-pyruvate aminotransferase (spuC1 and spuC2 genes) and a ɣ-aminobutyrate aminotransferase (gabT1 and gabT2 genes) have been identified in this bacterium. When the expression pattern of these genes is analyzed by qPCR, it is drastically conditioned by supplying the carbon sources. Thus, spuC1 is upregulated by putrescine, whereas spuC2 seems to be exclusively induced by cadaverine. However, gabT1 increases its expression in response to different polyamines or aminated catabolic derivatives from them (i.e., ɣ-aminobutyrate or δ-aminovalerate), although gabT2 does not change its expression level concerning no-amine unrelated carbon sources (citrate). These results reveal differences between the mechanisms proposed for polyamine catabolism in P. aeruginosa and Escherichia coli concerning P. putida strain U, as well as allow a deeper understanding of the enzymatic systems used by this last strain during polyamine metabolism. A new Maximum Power Point Tracking (MPPT) method, consisting in combining the Incremental Conductance (INC) algorithm with the Hysteresis control, was developed and applied to a standalone photovoltaic (PV) system to generate the maximum power of the PV array. The INC allows one to search for the Maximum Power Point (MPP). The hysteresis improves the accuracy of tracking the MPP very fast even after severe changes in weather conditions and has no oscillations around the MPP. The five-level S-Packed U Cells (SPUC5) inverter is used to transform the produced DC voltage to AC voltage; it generates five-level output voltage with a small number of switches and only DC source voltage. The capacitors of the SPUC5 are controlled by the Pulse Width Modulation (PWM) in order to balance their voltages. The proposed PV system was established and trained in the MATLAB/Simulink environment under various irradiation conditions. A comparison between different MPPT methods, INC-PWM and INC-PI, was investigated in order to examine the effectiveness of the developed MPPT technique in particular, and of all the PV system components. The results of the simulation validate the effectiveness of the suggested MPPT algorithm as well as the used SPUC5 inverter. Full article

Distribution of unloading zones determines stability of slopes, and the unloading of a large-scale rocky slope formed by rapid river erosion and surface uplift on the Qinghai–Tibet Plateau is particularly severe. Generally, unloading assessment relies on traditional methods, which are mainly related to phenomena. However, unloading is a process of deformation and failure regarding the rock mass. Based on deformation and failure, strain rate and crack rate established through theoretical analysis can measure the relationship between accumulative width of open cracks and unloading deformation and the relationship between accumulative number of cracks and unloading failure, respectively. Thus, a method that combines strain rate and crack rate, namely strain per unit crack (i.e., SPUC), is proposed. The SPUC was applied to assess the unloading zones of a large-scale rocky slope on the Qinghai–Tibet Plateau. The results show that the SPUC curve regularly varied and can be easily divided into three parts. Strong and weak unloading zones can be recognized from the SPUC curve. The reasonability of SPUC in the unloading zones assessment was confirmed by comparing and verifying with traditional methods. We found that SPUC has good universality and can compensate for the defect of using strain rate or crack rate to assess unloading zones. Full article

In this paper, the authors propose a new three-phase, nine-level inverter with self-balancing of capacitors voltages. The proposed inverter is the result of a serial connection of the SPUC topology and the NPC converter. A single DC source is used, and each phase is made up of nine power switches and three capacitors. Two control techniques are proposed to maintain capacitors voltages at desired values, the first of which is a PWM technique ensures the self-balancing of capacitors voltages in open loop without using any filters or PI regulators, and the second is based on the hysteresis control which offers a nearly sinusoidal waveform of the load current without using any voltage sensors. The authors make use of the Matlab Simulink environment to perform the simulation of the proposed concept. The dynamics of the latter was verified against load change. Full article

In this work, an advanced pulse width modulation (PWM) technique was developed to provide the auto-balancing of the capacitors voltages of the five-level split-packed U-Cells (SPUC5) single-phase inverter, and then, the latter was applied to a photovoltaic (PV) system in standalone mode to evaluate its performance in this kind of application. The SPUC5 inverter makes use of only five switches (four active bidirectional switches and one four quadrant switch), one DC source and two capacitors to generate five levels of output voltage and a current with a quasi-sinusoidal waveform which reduces the total harmonic distortion (THD) without the need to add filters or sensors, and also reduces its cost compared to the other multilevel inverters. In the proposed system; the incremental conductance (INC) algorithm is combined with a DC/DC boost converter to reach the maximum power (MP) of the PV array by tracking the MP point (MPP). The offered concept has been constructed and then simulated in the MATLAB/Simulink environment to evaluate its efficiency. According to the results, the self-balancing of the capacitors voltages has been achieved. A comparative study was performed with the traditional PWM technique. The proposed PV system has been validated by experimental results. Full article

Sphingopyxis sp. USTB-05, which we previously identified and examined, is a well-known bacterial strain for biodegrading cyanobacterial hepatotoxins of both nodularins (NODs) and microcystins (MCs). Although the pathways for biodegrading the different types of [D-Asp1] NOD, MC-YR, MC-LR and MC-RR by Sphingopyxis sp. USTB-05 were suggested, and several biodegradation genes were successfully cloned and expressed, the comprehensive genomic analysis of Sphingopyxis sp. USTB-05 was not reported. Here, based on second and third generation sequencing technology, we analyzed the whole genome of Sphingopyxis sp. USTB-05, which is 4,679,489 bp and contains 4,312 protein coding genes. There are 88 protein-coding genes related to the NODs and MCs biodegradation, of which 16 genes (bioA, hmgL, hypdh, speE, nspC, phy, spuC, murD, glsA, ansA, ocd, crnA, ald, gdhA, murC and murI) are unique. These genes for the transformation of phenylacetic acid CoA (PA-CoA) to CO2 were also found in Sphingopyxis sp. USTB-05. This study expands the understanding of the pathway for complete biodegradation of cyanobacterial hepatotoxins by Sphingopyxis sp. USTB-05. Full article
