Parrotia subaequalis is a rare and endangered deciduous tree native to China, valued for its vibrant autumn foliage and ornamental appeal. Its leaves exhibit striking coloration, ranging from red to yellow and purple, yet the physiological and molecular mechanisms behind this variation remain poorly understood. Here, we combined transcriptomic, metabolomic, and physiological analyses to investigate pigment changes within the yellow leaf phenotype of P. subaequalis. Our findings revealed significant differences in gene expression and metabolic profiles between yellow and green leaves, particularly in starch and sucrose metabolism, photosynthesis, and carbon metabolism. Yellow leaves exhibited reduced photosynthetic capacity and carotenoid levels, alongside increased D-glucose concentration. These findings suggest that visible color transitions are likely driven by coordinated changes in carbohydrate metabolism, photosynthetic function, and organic compound accumulation. This study provides novel insights into the molecular and physiological mechanisms governing leaf pigmentation in an endangered tree, with useful information relevant to their conservation and sustainable utilization. Hynobius amjiensis, also known as the Anji salamander, is an amphibian species currently categorized as endangered due to its limited geographical distribution, primarily in China. To address the critical conservation status of this species, artificial breeding is essential for population expansion. However, progress in artificial breeding efforts has been hindered by the scarcity of research on the reproductive biology of the Anji salamander. In this study, we identified 25 distinct early stages of embryo development. Additionally, we observed that Anji salamander embryos contain a lesser amount of yolk compared to other salamanders or frogs. We further discovered that the Anji salamander employs a highly competitive reproductive strategy, producing a smaller number of high-quality offspring. This strategy aims to generate adaptive individuals through intense intraspecific competition, driven by three factors: extremely confined breeding habitats, a substantial number of eggs, and a reduced yolk content. We introduce the term “mass escape” to describe this strategy, which provides a novel perspective on cannibalization, focusing on the consumption of specific body parts rather than a single-individual activity. This study offers valuable insights into artificial breeding techniques designed to mitigate inherent intraspecific competitive pressure, thereby improving metamorphosis and survival rates. Peatlands deliver a variety of beneficial ecosystem services, particularly serving as habitats for a diverse array of species. Hynobius amjiensis is a critically endangered amphibian initially discovered in a Sphagnum-dominated peatland in Anji, China. The unique habitat requirements of H. amjiensis make it highly vulnerable to environmental changes. Here, we investigated the different breeding pools of H. amjiensis in the Sphagnum-dominated peatland (the type locality) for a one-year period to evaluate the interactions among the egg sacs present, water quality, and microbial communities (16S and 18S rRNA gene amplicon). The numbers of egg sacs were higher in the breeding pools located at the marginal area than those at the core area of the peatland. Similarly, the α-diversity of bacteria, fungi, and protists were lower in the core region compared to those at the edge of the peatland, perhaps due to water eutrophication. The microbial communities and water quality differed significantly among breeding pools and sampling months. The simpler microbial networks of the breeding pools in the core wetland may impact the numbers and health of the egg sacs. This study contributes to a better understanding of the effect of water quality on biodiversity in peatlands, and it can also guide regulations for wetland conservation and the protection of endangered species. It is important to describe lineages before they go extinct, as we can only protect what we know. This is especially important in the case of microendemic species likely to be relict populations, such as Hynobius salamanders in southern China. Here, we unexpectedly sampled Hynobius individuals in Fujian province, China, and then worked on determining their taxonomic status. We describe Hynobius bambusicolus sp. nov. based on molecular and morphological data. The lineage is deeply divergent and clusters with the other southern Chinese Hynobius species based on the concatenated mtDNA gene fragments (>1500 bp), being the sister group to H. amjiensis based on the COI gene fragment, despite their geographic distance. In terms of morphology, the species can be identified through discrete characters enabling identification in the field by eye, an unusual convenience in Hynobius species. In addition, we noted some interesting life history traits in the species, such as vocalization and cannibalism. The species is likely to be incredibly rare, over a massively restricted distribution, fitting the definition of Critically Endangered following several lines of criteria and categories of the IUCN Red List of Threatened Species.
