Systemic lupus erythematous (SLE) is a chronic autoimmune disorder, broadly characterized by systemic inflammation along with heterogeneous clinical manifestations, severe morbidity, moribund organ failure and eventual mortality. In our study, SLE patients displayed a higher percentage of activated, inflamed and hyper-polarized CD8+ T cells, dysregulated CD8+ T cell differentiation, significantly elevated serum inflammatory cytokines and higher accumulation of cellular ROS when compared to healthy controls. Importantly, these hyper-inflammatory/hyper-polarized CD8+ T cells responded better to an antioxidant than to an oxidant. Terminally differentiated Tc1 cells also showed plasticity upon oxidant/antioxidant treatment, but that was in contrast to the SLE CD8+ T cell response. Our studies suggest that the differential phenotype and redox response of SLE CD8+ T cells and Tc1 cells could be attributed to their cytokine environs during their respective differentiation and eventual activation environs. The polarization of Tc1 cells with IL-21 drove hyper-cytotoxicity without hyper-polarisation suggesting that the SLE inflammatory cytokine environment could drive the extreme aberrancy in SLE CD8+ T cells. Cordia dichotoma Forst. (F. Boraginaceae) has been traditionally used for the management of a variety of human ailments. In our earlier work, the antidiabetic activity of methanolic bark extract of C. dichotoma (MECD) has been reported. In this paper, two flavonoid molecules were isolated (by column chromatography) and identified (by IR, NMR and mass spectroscopy/spectrometry) from the MECD with an aim to investigate their antidiabetic effectiveness. Molecular docking and ADMET studies were carried out using AutoDock Vina software and Swiss ADME online tool, respectively. The isolated flavonoids were identified as 3,5,7,3′,4′-tetrahydroxy-4-methoxyflavone-3-O-l-rhamnopyranoside and 5,7,3′-trihydroxy-4-methoxyflavone-7-O-l-rhamnopyranoside (quercitrin). Docking and ADMET studies revealed the promising binding affinity of flavonoid molecules for human lysosomal α-glucosidase and human pancreatic α-amylase with acceptable ADMET properties. Based on computational studies, our study reports the antidiabetic potential of the isolated flavonoids with predictive pharmacokinetics profile. Full article

Estimation of latent heat flux at the agricultural field scale is required for proper water management. The current generation thermal sensors except Landsat-8 provide data on the order of 1000 m. The aim of this study is to test three approaches based on contextual models using only remote sensing datasets for the disaggregation of latent heat flux over India. The first two approaches are, respectively, based on the estimation of the evaporative fraction (EF) and solar radiation ratio at coarser resolution and disaggregating them to yield the latent heat flux at a finer resolution. The third approach is based on disaggregation of the thermal data and estimating a finer resolution latent heat flux. The three approaches were tested using MODIS datasets and the validation was done using the Bowen Ratio energy balance observations at five sites across India. From the validation, it was observed that the first two approaches performed similarly and better than the third approach at all five sites. The third approach, based on the disaggregation of the thermal data, yielded larger errors. In addition to better performance, the second approach based on the disaggregation of solar radiation ratio was simpler and required lesser data processing than the other approaches. In addition, the first two approaches captured the spatial pattern of latent heat flux without introducing any artefacts in the final output. Full article
