This article reviews the multifaceted roles of itaconate in immune regulation and inflammatory metabolism. Itaconic acid is a dicarboxylic acid with anti-inflammatory, antioxidant, and anti-tumor properties. It is initially produced by the heating decomposition of citric acid and is closely related to the tricarboxylic acid cycle. In immune regulation, itaconate regulates macrophage function through a variety of mechanisms, including metabolic reprogramming, polarization regulation, inhibition of cytokine production, and regulation of oxidative stress. It can also affect the function of T cells and B cells. In terms of inflammatory metabolism, itaconate can regulate the production of inflammatory factors, inhibit the activity of succinate dehydrogenase, and affect cellular energy metabolism and lipid metabolism. Its mechanism of action involves the inhibition of succinate dehydrogenase, covalent modification of proteins, influence on epigenetic modification, and playing a role through the G protein-coupled receptor OXGR1 (Oxoglutarate Receptor 1). Itaconic acid derivatives have shown good effects in anti-inflammation and anti-oxidation and have broad application prospects in clinical treatment, including the treatment of inflammatory diseases, anti-tumor and anti-microbial infection. However, the long-term safety and side effects of itaconic acid as a therapeutic agent still need to be further studied. Future studies will further explore the synthesis and function of itaconic acid in different cell types, its physiological effects in non-inflammatory conditions, and its potential application in clinical treatment in order to develop new therapeutic strategies and improve the treatment effect of chronic inflammatory and metabolism-related diseases. Intestinal bacteria form one of the most complex microbial communities in the human body, playing a crucial role in maintaining host health and contributing to the development of various diseases. Here, we provide a comprehensive overview of the composition and function of intestinal bacteria, the factors affecting their homeostasis, and their association and mechanisms with a range of diseases (e.g., inflammatory bowel diseases, colorectal cancer, metabolic diseases). Additionally, their advanced potential in disease diagnosis and treatment is highlighted. Therapies, such as chemotherapy, radiotherapy, and immunotherapy, are significantly impacted by intestinal bacteria, with research indicating that bacteria can enhance chemoimmunotherapy efficiency by affecting T cell recruitment and immune cell infiltration. Fecal microbiota transplantation has emerged as a promising option for treating recurrent Clostridium difficile infections and certain metabolic and neurological disorders. Gut bacteria-related serum metabolites serve as non-invasive indicators for diagnosing CRC, while fecal immunochemical tests offer promising applications in CRC screening. Future research is needed to better understand the causal relationships between intestinal bacteria and diseases, develop more precise diagnostic tools, and evaluate the effectiveness and safety of microbiome-targeted therapies in clinical treatment. This study provides deeper insights into the role of intestinal bacteria in human health and disease, providing a scientific basis for innovative therapeutic strategies that have the potential to transform the landscape of healthcare. Identification of Potential Landslides in the Gaizi Valley Section of the Karakorum Highway Coupled with TS-InSAR and Landslide Susceptibility Analysis

Landslides have become a common global concern because of their widespread nature and destructive power. The Gaizi Valley section of the Karakorum Highway is located in an alpine mountainous area with a high degree of geological structure development, steep terrain, and severe regional soil erosion, and landslide disasters occur frequently along this section, which severely affects the smooth flow of traffic through the China-Pakistan Economic Corridor (CPEC). In this study, 118 views of Sentinel-1 ascending- and descending-orbit data of this highway section are collected, and two time-series interferometric synthetic aperture radar (TS-InSAR) methods, distributed scatter InSAR (DS-InSAR) and small baseline subset InSAR (SBAS-InSAR), are used to jointly determine the surface deformation in this section and identify unstable slopes from 2021 to 2023. Combining these data with data on sites of historical landslide hazards in this section from 1970 to 2020, we constructed 13 disaster-inducing factors affecting the occurrence of landslides as evaluation indices of susceptibility, carried out an evaluation of regional landslide susceptibility, and identified high-susceptibility unstable slopes (i.e., potential landslides). The results show that DS-InSAR and SBAS-InSAR have good agreement in terms of deformation distribution and deformation magnitude and that compared with single-orbit data, double-track SAR data can better identify unstable slopes in steep mountainous areas, providing a spatial advantage. The landslide susceptibility results show that the area under the curve (AUC) value of the artificial neural network (ANN) model (0.987) is larger than that of the logistic regression (LR) model (0.883) and that the ANN model has a higher classification accuracy than the LR model. A total of 116 unstable slopes were identified in the study, 14 of which were determined to be potential landslides after the landslide susceptibility results were combined with optical images and field surveys. These 14 potential landslides were mapped in detail, and the effects of regional natural disturbances (e.g., snowmelt) and anthropogenic disturbances (e.g., mining projects) on the identification of potential landslides using only SAR data were assessed. The results of this research can be directly applied to landslide hazard mitigation and prevention in the Gaizi Valley section of the Karakorum Highway. In addition, our proposed method can also be used to map potential landslides in other areas with the same complex topography and harsh environment. Full article

Temperature and precipitation are crucial indicators for investigating climate changes, necessitating precise measurements for rigorous scientific inquiry. While the Fifth Generation of European Centre for Medium-Range Weather Forecasts Atmospheric Reanalysis (ERA5), ERA5 of the Land Surface (ERA5-Land), and China Meteorological Forcing Dataset (CMFD) temperature and precipitation products are widely used worldwide, their suitability for the Altay region of arid and semi-arid areas has received limited attention. Here, we used the Altay region as the study area, utilizing meteorological station data and implementing the residual revision method for temperature and the coefficient revision method for precipitation to rectify inaccuracies in monthly temperature and precipitation records from ERA5-Land, ERA5, and CMFD. We evaluate the accuracy of these datasets before and after correction using bias, Taylor diagrams, and root-mean-square error (RMSE) metrics. Additionally, we employ Tropical Rainfall Measuring Mission satellite precipitation data (TRMM) as a benchmark to assess the performance of ERA5-Land, ERA5, and CMFD monthly precipitation before and after correction. The results revealed significant differences in the temperature and precipitation capture capabilities of ERA5-Land, ERA5, and CMFD in the Altay region. Overall, these data exhibit substantial errors and are not directly suitable for scientific research. However, we applied residual and coefficient revision methods. After this revision, ERA5-Land, ERA5, and CMFD showed significantly improved temperature and precipitation capture capabilities, especially for ERA5-Land. In terms of temperature, post-revision-CMFD (CMFDPR) demonstrated better temperature capture capabilities. All three datasets showed weaker performance in mountainous regions compared to plains. Notably, post-revision-ERA5 (ERA5PR) seemed unsuitable for capturing temperature in the Altay region. Concerning rain, CMFDPR, post-revision-ERA5-Land (ERA5-LandPR) and ERA5PR outperformed TRMM in capturing precipitation. CMFDPR and ERA5-LandPR both outperform ERA5PR. In summary, the revision datasets effectively compensated for the sparse distribution of meteorological stations in the Altay region, providing reliable data support for studying climate change in arid and semi-arid areas. The arid terrestrial ecosystem carbon cycle is one of the most important parts of the global carbon cycle, but it is vulnerable to external disturbances. As the most direct factor affecting the carbon cycle, how land cover change affects vegetation carbon sources/sinks in arid terrestrial ecosystems remains unclear. In this study, we chose the arid region of northwest China (ARNWC) as the study area and used net ecosystem productivity (NEP) as an indicator of vegetation carbon source/sink. Subsequently, we described the spatial distribution and temporal dynamics of vegetation carbon sources/sinks in the ARNWC from 2001–2018 by combining the Carnegie-Ames-Stanford Approach (CASA) and a soil microbial heterotrophic respiration (RH) model and assessed the effects of land cover change on them through modeling scenario design. We found that land cover change had an obvious positive impact on vegetation carbon sinks. Among them, the effect of land cover type conversion contributed to an increase in total NEP of approximately 1.77 Tg C (reaching 15.55% of the original value), and after simultaneously considering the effect of vegetation growth enhancement, it contributed to an increase in total NEP of approximately 14.75 Tg C (reaching 129.61% of the original value). For different land cover types, cropland consistently contributed the most to the increment of NEP, and the regeneration of young and middle-aged forests also led to a significant increase in forest carbon sinks. Thus, our findings provide a reference for assessing the effects of land cover change on vegetation carbon sinks, and they indicated that cropland expansion and anthropogenic management dominated the growth of vegetation carbon sequestration in the ARNWC, that afforestation also benefits the carbon sink capacity of terrestrial ecosystems, and that attention should be paid to restoring and protecting native vegetation in forestland and grassland regions in the future. Background: Hyperthermic intraperitoneal chemotherapy (HIPEC) in T4 colorectal cancer (CRC) remains controversial. The study aimed to explore the safety and efficacy of radical surgery (RS) with HIPEC in T4 CRC. Methods: Adverse events after HIPEC were estimated by Common Terminology Criteria for Adverse Events version 5.0. The efficacy was evaluated using recurrence-free survival (RFS) and overall survival (OS). Propensity score matching (PSM) was used to reduce the effects of confounders between groups. Results: Of the 417 patients (263 men and 154 women), 165 patients were treated with RS + HIPEC and 252 patients with RS alone. There was no significant difference in the incidence of all adverse events after PSM. Overall RFS and OS were not significantly different at 24 months (p = 0.580 and p = 0.072, respectively). However, in patients with T4b stage CRC (92.1% vs. 77.3%, p = 0.048) and tumor size ≥ 5 cm (93.0% vs. 80.9%, p = 0.029), RFS in the two groups showed a significant difference at 24 months. Conclusions: In summary, the safety of HIPEC in T4 CRC was confirmed. Compared with RS, though RS + HIPEC did not benefit the overall cohort at 24 months, RS + HIPEC could benefit patients with T4b stage CRC and tumor size ≥ 5 cm in RFS. Full article

Background: The drug selection of radical surgery (RS), with hyperthermic intraperitoneal chemotherapy (HIPEC), in pT4 colorectal cancer (CRC) remains controversial. Methods: Adverse events after HIPEC were estimated by common terminology criteria for adverse events version 5.0. The efficacy was evaluated using overall survival (OS) and recurrence-free rate (RFR). Propensity score matching (PSM) was used to reduce the influence of confounders between Mitomycin and Lobaplatin groups. Results: Of the 146 patients, from April 2020 to March 2021, 47 were managed with mitomycin and 99 with lobaplatin. There was no significant difference in the incidence of all adverse events between the two groups after PSM. OS and RFR were not significantly different between the two groups at 22 months (p = 0.410; p = 0.310). OS and RFR of the two groups also showed no significant difference for patients with T4a or T4b stage, tumor size < or ≥ 5 cm. Among patients with colon cancer, RFR at 22 months of the two groups was significantly different (100.0% vs. 63.2%, p = 0.028). Conclusions: In summary, the safety of mitomycin and lobaplatin for HIPEC was not different. Compared with lobaplatin, mitomycin for HIPEC after RS could benefit patients with colon cancer in RFR. Colorectal cancer (CRC) is critically related to aging and severely threatens human lives. To better explore the effects of aging on CRC progression and therapy outcome, a reliable aging subtypes identification of CRC is urgently desired. Here, 28 aging-related genes associated with the CRC prognosis were selected by univariate Cox analyses. Based on these 28 genes, CRC patients were divided into the aging subtype and young subtype by non-negative matrix factorization clustering. Aging subtype and young subtype of CRC were identified with distinct molecular features and clinical prognosis. The aging subtype was characterized by upregulation of senescence-associated secretory phenotype, higher frequencies of TP53 and immune checkpoint molecules, and high sensitivity to protein kinase and angiogenesis inhibitors. Furthermore, 14 genes were selected by LASSO penalized Cox regression analyses for aging-related risk signature construction. The constructed aging risk signature exhibited good prediction and the nomogram showed robust discrimination power over the traditional CRC staging system. In conclusion, this study successfully established aging subtype and young subtype of CRC, which is helpful to identify patients with aging characteristics to evaluate prognosis and treatment outcomes. Introducing aging-based subtypes into clinical concern and patient prognostication provides new opportunities for personalized CRC treatment. Full article
