This manuscript is well-written and logically clear.  I cannot many more comments but simply recommend it for acceptation. However, I do hope the authors can provide some more background regarding the importance to determine Lewis blood group status and the rationale for large population study.

The authors have developed a probe-based fluorescence melting curve analysis for the FUT3 gene analysis (Diagnostics 2022, 12(12), 3039). In this manuscript, the authors continue to use the same strategy to investigate the SNPs in both FUT2 and FUT3 genes among samples from the Japanese population. Compare to those established methods such as Sanger sequencing, this probe-based FMCA is suitable for high throughput screening with a friendly cost, and presents more robust and accurate results.

 

 

The whole manuscript is generally well organized, and the logic is clear. In all, I am willing to recommend publishing this paper with no more revisions.
