There is a plethora of individual studies of correlations of the frequency HIV-1 DNA integration into different genomic regions. Factors studied include the density of specific cellular proteins associated with different regions of chromatin, DNA features, and regions of chromatin associated with specific cytological features. It is quite hard to synthesize a global view of the the contribution of these factors and how they interact with each other. The authors provide a much-needed metanalysis that attempts to do this based on all the currently available data. It is a thorough analysis and an extremely important contribution to the field and is also of general interest. I have only a minor suggestion:

 

  1. It would be helpful to include a few sentences in the Introduction to explain what are nuclear-speckle-associated chromatin and spatial position of interference of nuclear genome (SPIN).

This is an interesting article, which demonstrates that the integration of the HIV-1 genome occurs into specific gene sets of host chromosomes and controlled by host proteins. The study used relevant controls, the data are well presented, and the conclusions are in accordance with the results.
