Optimization of the Brewing Process and Analysis of Antioxidant Activity and Flavor of Elderberry Wine

Fruit wines have high nutritional value and good palatability. However, fruit wine made from a single fruit type does not have good enough flavor and nutritional quality. Therefore, flavorsome fruit wines made from a variety of fruits should be developed as a matter of urgency. In this study, the raw material of elderberry wine was used to explore the production technology of mixed juice wines; the fruits selected were apple, lychee, pear, blueberry, and elderberry. We utilized a single-factor experiment and the response surface method (RSM) approach to optimize the fermentation procedures; the results show that the solid–liquid ratio was 1:7.5, the amount of yeast inoculation was 0.68 g/L, the fermentation temperature was 20 °C, and the added sugar content was 120 g/L. Under these process conditions, a verification test was carried out in a 35 L fermenter. The results showed that the alcohol content, residual sugar content, total acidity, total phenol content, and total flavonoid content of the elderberry wine were, respectively, 7.73% vol, 8.32 g/L, 9.78 g/L, 8.73 mg/mL, and 1.6 mg/mL. In total, 33 volatile components were identified in the resulting elderberry wine. It achieved a harmonious aroma and fruit flavor, a homogeneous and transparent liquid phase, a pleasant taste, and a sensory evaluation score of 95. The antioxidant activity experiments showed that elderberry had a certain antioxidant capacity, and that fermented elderberries had significantly higher antioxidant ability than unfermented ones. The “soilless” cultivation of blueberry (Vaccinium corymbosum L.) in containers with peat as substrate allows overcoming the problem of unsuitable soils, thus enhancing the spread of this crop in new areas. Since the use of peat presents several critical environmental and economic sustainability issues, the evaluation of alternative solutions is required. The effectiveness of compost produced with solid digestate and residues from the vine-wine chain to replace part of the peat was therefore tested. Micropropagated plants of cultivar Duke grown in three substrates consisting of a mixture of commercial peat with three compost fractions (10, 20, 40%) were compared with plants grown in 100% unfertilized or fertilized peat (0.3 g of Osmocote per pot). Plant height did not significantly differ between the five theses at the end of the trial, whereas the total number of nodes per plant was higher than in the control theses, due to a greater development of secondary shoots. The nutritional status of the plants, monitored with Dualex, during the growing season, was generally not significantly different in the innovative substrates compared to peat alone. In mid-summer the plants grown in substrates with compost showed the best nitrogen balance index (NBI values). Plants cultivated with medium-high percentages of compost (20–40%) showed a lower degree of defoliation at the end of the trial, dependent on a slower decline of vegetative activity. The final destructive measures of fresh and dry weight of biomass and of its partitioning between roots and shoots highlight that the use of compost did not negatively affect the production of biomass, but rather, in the theses with the highest percentages of compost (20–40%), root development was stimulated. Blueberry wine residues produced during the wine-brewing process contain abundant anthocyanins and other bioactive compounds. To extract anthocyanins from blueberry wine residues more efficiently, a novel procedure of ultrasound-assisted deep eutectic solvent extraction (UADESE) was proposed in this work. The extraction process was optimized by response surface methodology coupled with genetic algorithm. The optimum extraction parameters to achieve the highest yield of anthocyanins (9.32 ± 0.08 mg/g) from blueberry wine residues by UADESE were obtained at water content of 29%, ultrasonic power of 380 W, extraction temperature of 55 °C, and extraction time of 40 min. The AB-8 macroporous resin combined with Sephadex LH-20 techniques was used to purify the crude extract (CE) obtained under optimum extraction conditions and analyze the anthocyanins composition by HPLC-ESI-MS/MS. The cyanidin-3-rutinoside with purity of 92.81% was obtained. The HepG2 antitumor activity of CE was better than that of the purified anthocyanins component. Moreover, CE could increase the intracellular reactive oxygen species levels and the apoptosis, and arrest HepG2 cells in the S phases. These findings provided an effective and feasible method for anthocyanins extraction, and reduced the environmental burden of this waste. Full article
