The oleoresin and volatile fraction produced by conifers, such as Pinus pinaster, play a crucial role in plant defence, acting as precursors to resin acids and adapting in response to environmental stress or pathogen attacks. Abietadiene (abieta-7,13-diene), the biosynthetic precursor to abietic acid, has been identified as the most abundant compound in extracts from pinewood nematode (PWN)-infected P. pinaster trees. As abietadiene is not commercially available, this study aimed to achieve, for the first time, its isolation, structure elucidation, and detailed characterization from readily available forestry residues. Abietadiene was successfully isolated using thin-layer chromatography (TLC), and its purity and identity were evaluated using multiple analytical techniques: gas chromatography (GC), liquid chromatography (LC), nuclear magnetic resonance (NMR), and Fourier-transform infrared spectroscopy (FTIR). GC analysis indicated a purity of over 70% for the isolated compound, while LC provided the higher purity value of 98%. The identity of abietadiene was unequivocally confirmed through LC, FTIR, and NMR analysis. This work represents the first isolation and comprehensive characterization of abietadiene from a natural source, making detailed chemical data on this compound available to the scientific community. These findings may be used for future studies on the biological interactions and ecological roles of abietadiene, particularly in the context of plant defence and pathogen resistance. Full article

There is a high interest in utilizing natural bioactive products derived from plants as a substitute for synthetic chemicals in the industry. This research focuses on the phytochemical composition of essential oils (EOs) of Ammi visnaga L. and Trachyspermum ammi L and their insecticidal activity against Sitophilus oryzae (L.), a common pest found in stored cereals. The EOs were extracted through steam distillation and analyzed using gas chromatography coupled with mass spectrometry (GC-MS). The EOs of A. visnaga consisted of twenty-four components, with Abietadiene (41.23%) being the most abundant, followed by linalool (25.54%) and limonene (19.04%). On the other hand, the EOs of T. ammi consisted of twenty-eight main components, with isothymol being the most abundant (51.88%). The results revealed that the EOs of T. Ammi (DL50 = 0.1 µL EOs/L of air) were more toxic than A. visnaga (0.38 µL EOs/L of air), with the toxicity varying based on doses and exposure periods. To further understand the molecular mechanisms underlying this activity, molecular docking and dynamic simulations were performed using the major chemical constituents of the oils. The simulation results indicated that the major compounds, Abietadiene and isothymol, interact with the catalytic sites of the target proteins, inhibiting acetylcholinesterase and chitin synthase. These interactions form energetically favorable systems that remain stable throughout the molecular dynamic period. This research provides valuable insights into the potential of these EOs as natural insecticides and highlights the importance of molecular modeling in understanding the biological activities of plant-derived compounds. Biological Properties of Extracts Obtained from In Vitro Culture of Plectranthus scutellarioides in a Cell Model

Plectranthus scutellarioides (L.) R.Br. is a medicinal plant that has long been used in traditional medicine to treat conditions such as abscesses, ulcers, and ear and eye infections. It is known to have a wide range of biological properties, such as antibacterial, antioxidant, antifungal, anti-inflammatory, anti-diabetic and anti-cancer effects. In this study, we established in vitro cultures from both the aerial parts and roots of Plectranthus scutellarioides. Subsequently, we compared the basic phytochemical profile of the obtained extracts and conducted a biological analysis to assess their potential for inducing apoptosis in breast (MCF-7) and lung (A549) cancer cells. Phytochemical analysis by HPLC-MS revealed the presence of compounds belonging to phenolic acids (ferulic, syringic, vanillic, rosmarinic, chlorogenic, caffeic, coumaric, dihydroxybenzoic acids), flavonoids (eriodyctiol and cirsimaritin), and terpenes such as 6,11,12,14,16-Pentahydroxy-3,17diacetyl-8,11,13-abietatrien-7-one, 6,11,12,14,16-Pentahydroxy-3,17-diacetyl5,8,11,13-abietatetraen-7-one, and 3,6,12-Trihydroxy-2-acetyl-8,12-abietadien7,11,14-trione. The results show that both extracts have a cytotoxic and genotoxic effect against MCF-7 and A549 cancer cells, with a different degree of sensitivity. It was also shown that both extracts can induce apoptosis by altering the expression of apoptotic genes (Bax, Bcl-2, TP53, Fas, and TNFSF10), reducing mitochondrial membrane potential, increasing ROS levels, and increasing DNA damage. In addition, it has been shown that the tested extracts can alter blood coagulation parameters. Our results indicate that extracts from in vitro cultures of Plectranthus scutellarioides aerial parts and roots have promising therapeutic application, but further research is needed to better understand the mechanisms of their action in the in vitro model. The seasonal variation of the chemical composition of the essential oil from fresh leaves of Tetradenia riparia (Hochst.) Codd grown in southern Brazil was analyzed by GC-MS, and the analgesic and antimicrobial activities of this oil were assayed. The yield of essential oil ranged from 0.17% to 0.26%, with the maximum amount in winter and the minimum in spring. The results obtained from principal components analysis (PCA) revealed the existence of high chemical variability in the different seasons. The samples were clearly discriminated into three groups: winter, autumn, and spring-summer. Samples collected during winter contained the highest percentages of calyculone (24.70%), abietadiene (13.54%), and viridiflorol (4.20%). In autumn, the major constituents were ledol (8.74%) and cis-muurolol-5-en-4-α-ol (13.78%). Samples collected in spring-summer contained the highest percentages of fenchone (12.67%), 14-hydroxy-9-epi-caryophyllene (24.36%), and α-cadinol (8.33%). Oxygenated sesquiterpenes were predominant in all the samples analyzed. The observed chemovariation might be environmentally determined by a seasonal influence. The essential oil, when given orally at a dose of 200 mg/kg, exhibited good analgesic activity on acetic acid-induced writhing in mice, inhibiting the constrictions by 38.94% to 46.13%, and this effect was not affected by seasonal variation. The antimicrobial activity of the essential oil against the bacterial strains: Staphylococcus aureus, Bacillus subtilis, Enterococcus faecalis, Escherichia coli, Salmonella enterica, Pseudomonas aeruginosa, Klebsiella pneumonia, Proteus mirabilis, Morganella morganii, and Enterobacter cloacae, and the pathogenic fungus Candida albicans was assessed by the disc diffusion method and determination of the minimum inhibitory concentration. The results obtained, followed by measurement of the minimum inhibitory concentration (MIC), indicated that S. aureus, B. subtilis, and Candida albicans were the most sensitive microorganisms, showing largest inhibition, and the lowest MIC values varied from 15.6 to 31.2 µg/mL, 7.8 to 15.6 µg/mL, and 31.2 to 62.5 µg/mL, respectively.
