Exploring the Potential of Low-Temperature Vacuum Drying to Improve the Bioactive Compound Content and Health-Promoting Properties of Chilean Wild Murta

For the first time, the effect of low-temperature vacuum drying (LTVD) on wild murta (Ugni molinae Turcz) was evaluated, in comparison with freeze-drying (FD) and vacuum drying (VD), to assess their capacity to preserve bioactive compounds and associated bioactivities. Murta was dried using LTVD at 20, 30, and 40 °C under a constant vacuum of 10 mbar, where FD and VD at 60 °C (VD 60) were included as comparative methods. The content of fatty acids and tocols, along with the retention of bioactive compounds and their antioxidant, anti-inflammatory, cytotoxic, and α-glucosidase inhibitory activities, were systematically analyzed. LTVD- and VD-dried murta exhibited higher polyunsaturated-to-saturated fatty acid ratios (>9.0) and markedly greater tocol contents, whereas FD maintained a more balanced ratio (<5.0) but with lower tocol levels. While FD was most effective in preserving catechin, higher levels of other phenolic compounds were observed in samples dried by LTVD at 20 and 40 °C, as well as VD 60, possibly due to the release of bound forms during processing. The drying method significantly influenced murta bioactivity. LTVD 30 preserved the highest antioxidant capacity, while topical anti-inflammatory effects on skin lesions varied by pathway, with LTVD 40 being the most effective in the TPA model and FD in the AA model. These effects were evaluated only using a topical inflammation model in BALB/c mice of both sexes; dietary effects were not assessed in this study. Regarding other bioactivities, VD 60 extracts excelled in both cytotoxic and α-glucosidase inhibitory effects, whereas FD extracts were the most effective against AGS cells and LTVD 20 against α-glucosidase. In conclusion, LTVD emerges as a promising alternative to FD and VD, showing potential to preserve bioactive compounds and key bioactivities of wild murta, although further studies are needed to elucidate the underlying mechanisms. Cauliflower (Brassica oleraceae L. var. Botrytis Linnaeus) has various health benefits due to its rich bioactive compound content. However, this fresh vegetable faces challenges related to its perishability and short shelf life. This study explores the effect of five drying methods, namely vacuum drying (VD), convective drying (CD), infrared drying (IRD), low-temperature vacuum drying (LTVD) and vacuum freeze-drying (VFD), on the bioactive compounds and health-promoting properties of cauliflower. Analyses of amino acids, hydroxycinnamic acid and its derivatives, glucosinolates, and isothiocyanates, as well as evaluations of their anti-inflammatory, antiproliferative, and neuroprotective properties, were conducted based on these five drying methods. The results revealed that samples treated with VFD and IRD had a higher content of amino acids involved in GSL anabolism. Moreover, VFD samples retained hydroxycinnamic acid derivatives and glucosinolates to a greater extent than other methods. Nonetheless, the CD and VD samples exhibited higher antiproliferative and neuroprotective effects, which were correlated with their high sulforaphane content. Overall, considering the retention of most bioactive compounds from cauliflower and the topical inflammation amelioration induced in mice, VFD emerges as a more satisfactory option. Full article

Stress-inducing husbandry and rearing conditions, bacterial infections or parasitic diseases may all lead to chronic inflammation. The immune response will then channel energy away from growth, reproduction and other important physiological processes, to fuel immune-related metabolic responses. The present study aims to unravel the mechanisms and contribute with new information on the molecular, cellular and humoral parameters of European seabass (Dicentrarchus labrax) undergoing chronic inflammation that can be used as health indicators for application in fish health management. European seabass individuals were intra-peritoneally injected with either Freund’s Incomplete Adjuvant (FIA) to induce inflammation or Hanks Balanced Salt Solution (HBSS) to serve as sham. Fish were sampled at 24 h, 7, 14 and 21 days post-injection and blood, plasma and head-kidney were collected. The results found were clear indicators of an inflamed peritoneal cavity and an ongoing systemic immune response that persisted for at least 21 days. Locally, inflammation was characterized by an intense recruitment of immune cells that was still evident 21 days after injection, thus illustrating the chronic character of the immune response. Cellular response was also noticed peripherally with leukocyte numbers rising in the blood of FIA-injected fish. Furthermore, the cellular-mediated respiratory burst peaked at 21 days post-FIA injection, suggesting that phagocytes were still actively fighting the phlogistic agent. Regarding the head-kidney molecular analysis, cxcr4 and il34 appear to be good markers of a chronic inflammation response due to their importance for pathways with high relevance in chronic inflammation settings. In addition, opioid receptor nopr seems to be a good marker of a chronic inflammation response due to its role in detecting noxious stimuli. The present study can serve as a baseline to assess long-term immune-related responses in future studies. For that, more research is nonetheless required to select more responsive and specific molecular markers. Full article
