Influence of Silver Nanoparticles on the Growth of Ascitic and Solid Ehrlich Adenocarcinoma: Focus on Copper Metabolism

The link between copper metabolism and tumor progression motivated us to use copper chelators for suppression of tumor growth. We assume that silver nanoparticles (AgNPs) can be used for lowering bioavailable copper. Our assumption is based on the ability of Ag(I) ions released by AgNPs in biological media and interfere with Cu(I) transport. Intervention of Ag(I) into copper metabolism leads to the replacement of copper by silver in ceruloplasmin and the decrease in bioavailable copper in the bloodstream. To check this assumption, mice with ascitic or solid Ehrlich adenocarcinoma (EAC) were treated with AgNPs using different protocols. Copper status indexes (copper concentration, ceruloplasmin protein level, and oxidase activity) were monitored to assess copper metabolism. The expression of copper-related genes was determined by real-time PCR in the liver and tumors, and copper and silver levels were measured by FAAS. Intraperitoneal AgNPs treatment beginning on the day of tumor inoculation enhanced mice survival, reduced the proliferation of ascitic EAC cells, and suppressed the activity of HIF1α, TNF-α and VEGFa genes. Topical treatment by the AgNPs, which was started together with the implantation of EAC cells in the thigh, also enhanced mice survival, decreased tumor growth, and repressed genes responsible for neovascularization. The advantages of silver-induced copper deficiency over copper chelators are discussed. Full article

The Use of Hydrogels in the Treatment of Metal Cultural Heritage Objects

Currently gels are widely used in the restoration of paintings, graphic arts, stuccowork and stonework, but their use in metal restoration is less widespread. In this study, several polysaccharide-based hydrogels (agar, gellan and xanthan gum) were selected for use in metal treatments. The use of hydrogels allows to localize a chemical or electrochemical treatment. This paper presents several examples of treatment of metal objects of cultural heritage, i.e., historical or archaeological objects. The advantages, disadvantages and limits of hydrogel treatments are discussed. The best results are obtained for the cleaning of copper alloys via associating an agar gel with a chelating agent (EDTA (ethylenediaminetetraacetic acid) or TAC (tri-ammonium citrate)). The hot application allows to obtain a peelable gel, particularly adapted for historical objects. Electrochemical treatments using hydrogels have been successful for the cleaning of silver and for the dechlorination of ferrous or copper alloys. The use of hydrogels for the cleaning of painted aluminum alloys is possible but it has to be coupled with mechanical cleaning. However, for the cleaning of archaeological lead, the cleaning using hydrogels was not very effective. In this work, two thiourea ligands bearing a phosphine group in one arm and in the other a phenyl group (T2) or 3,5-di-CF3 substituted phenyl ring (T1) have been prepared and their coordination to Au and Ag has been studied. A different behavior is observed for gold complexes, a linear geometry with coordination only to the phosphorus atom or an equilibrium between the linear and three-coordinated species is present, whereas for silver complexes the coordination of the ligand as P^S chelate is found. The thiourea ligands and their complexes were explored against different cancer cell lines (HeLa, A549, and Jurkat). The thiourea ligands do not exhibit relevant cytotoxicity in the tested cell lines and the coordination of a metal triggers excellent cytotoxic values in all cases. In general, data showed that gold complexes are more cytotoxic than the silver compounds with T1, in particular the complexes [AuT1(PPh3)]OTf, the bis(thiourea) [Au(T1)2]OTf and the gold-thiolate species [Au(SR)T1]. In contrast, with T2 better results are obtained with silver species [AgT1(PPh3)]OTf and the [Ag(T1)2]OTf. The role played by the ancillary ligand bound to the metal is important since it strongly affects the cytotoxic activity, being the bis(thiourea) complex the most active species. This study demonstrates that metal complexes derived from thiourea can be biologically active and these compounds are promising leads for further development as potential anticancer agents. Complexation of the 8,8′-bis(methylsulfanyl) derivatives of cobalt and iron bis(dicarbollides) [8,8′-(MeS)2-3,3′-M(1,2-C2B9H10)2] (M = Co, Fe) with copper, silver, palladium and rhodium leads to the formation of the corresponding chelate complexes, which is accompanied by a transition from the transoid to the cisoid conformation of the bis(dicarbollide) complex. This transition is reversible and can be used in design of coordination-driven molecular switches based on transition metal bis(dicarbollide) complexes. The solid-state structures of {(Ph3P)ClPd[8,8′- (MeS)2-3,3′-Co(1,2-C2B9H10)22-S,S′]} and {(COD)Rh[8,8′-(MeS)2-3,3′-Co(1,2-C2B9H10)22-S,S′]} were determined by single crystal X-ray diffraction. Three accurate methods were developed for the quantitative determination of cefprozil in pure form and in its dosage forms. The first method was based upon the interaction of the drug with 3-methyl-benzothiazolinone-2-hydrazone (MBTH) in the presence of ceric ammonium sulfate or ferric chloride as an oxidizing agent, where the formed color was measured at λ 521 nm or 624 nm, respectively. The second inethod was based on the chelate formation with palladium (II) chloride (PdCl2) in the presence of buffered medium, where the formed complex was determined at λ 345 nm. The third method was based upon the reaction of the neutral solution of the hydrolysis product of drug with each of silver nitrate & lead acetate standard solutions, forming drug-metal complex and the metal ion contents were determined directly or indirectly by atomic absorption spectroscopy (AAS). The reaction conditions of the proposed methods were studied and optimized. The precision of the proposed methods was achieved by determining different samples of bulk powder and pharmaceutical dosage forms.
