Danggui Buxue Decoction Alleviates Inflammation and Oxidative Stress in Mice with Escherichia coli-Induced Mastitis

(1) Background: Bovine mastitis is a lactational disease caused by infection and milk stagnation in the mammary glands. Danggui buxue decoction (DBD), a traditional remedy for blood tonification, anti-inflammation, and antioxidation, has not been used previously to treat mastitis. (2) Methods: In this study, an Escherichia coli mastitis model was established by infecting lactating Kunming mice with clinically isolated bovine mastitis-derived E. coli. Based on this, the effects of DBD on inflammation and oxidative stress in mastitis model mice were evaluated by conducting routine blood tests, H&E staining, qRT-PCR analysis, ELISA, and microcolorimetry. (3) Results: We found that DBD treatment reduced body weight loss, abnormal organ indices, abnormal blood cell counts, pathological damage to breast tissue, and the upregulation of the expression of inflammatory factor in mice caused by E. coli infection. We also found that DBD increased the expression of antioxidants and antioxidant genes and decreased the expression of oxidation products and oxidation-related genes in breast tissue. The therapeutic effect of DBD on inflammation and oxidative stress (OS) in mice occurred through the regulation of the TLR4/NF-κB and Nrf2/HO-1 signaling pathways. The therapeutic efficacy of a herbal mixture, being multi-target, multi-function and multi-pathway, is the niche of traditional Chinese medicine (TCM). Systems biology can dissect the network of signaling mechanisms in a complex biological system. In preparing TCM decoctions, the boiling of herbs together in water is a common practice; however, the rationale of this specific preparation has not been fully revealed. An approach of mass-spectrometry-based multi-omics was employed to examine the profiles of the cellular pathways, so as to understand the pharmacological efficacy of Danggui Buxue Tang (DBT), a Chinese herbal mixture containing Astragali Radix and Angelicae Sinensis Radix, in cultured rat osteoblasts and mesenchymal stem cells. The results, generated from omics analyses, were compared from DBT-treated osteoblasts to those of treating the herbal extract by simple mixing of extracts from Astragali Radix and Angelicae Sinensis Radix, i.e., herbal mixture without boiling together. The signaling pathways responsible for energy metabolism and amino acid metabolism showed distinct activation, as triggered by DBT, in contrast to simple mixing of two herbal extracts. The result supports that boiling the herbs together is designed to maximize the osteoblastic function of DBT, such as in energy and lipid metabolism. This harmony of TCM formulation, by having interactive functions of two herbs during preparation, is being illustrated. The systems biology approach provides new and essential insights into the synergy of herbal preparation. Well-defined multiple targets and multiple pathways in different levels of omics are the key to modernizing TCM. Full article

Danggui Buxue Decoction Sensitizes the Response of Non-Small-Cell Lung Cancer to Gemcitabine via Regulating Deoxycytidine Kinase and P-glycoprotein

This study aimed to investigate whether the anti-tumor effect of gemcitabine (GEM) in non-small-cell lung cancer (NSCLC) treatment was affected by Danggui Buxue decoction (DBD), and explore the potential mechanisms. The combined use of GEM and DBD showed an enhanced tumor growth inhibition effect in a murine Lewis lung carcinoma (LLC) model. LC-MS/MS results showed that the pharmacokinetic behaviors of a GEM active metabolite, gemcitabine triphosphate (dFdCTP), were found to be altered remarkably in the peripheral blood mononuclear cells (PBMC) of DBD co-administration rats. In addition, after co-administration of DBD with GEM, Western Blot and qPCR results confirmed that the expression of deoxycytidine kinase (dCK) in tumor tissues of LLC-bearing mice were markedly increased. DBD co-administration also reversed the upregulation of P-glycoprotein (P-gp) in tumor tissues induced by GEM. Moreover, DBD could notably up-regulate the IL-12p70 and GM-CSF expression in mice serum, suggesting potential immunomodulatory activities in tumor-bearing mice. Meanwhile, DBD inhibited the P-gp efflux activity in A549 cells. Therefore, the regulation of dCK and P-gp played important roles in the alternation of GEM pharmacokinetics and the enhancement of the anti-tumor effect of GEM. DBD being a potential dCK promoter could work as an adjuvant agent to boost the anticancer effect of GEM. Full article
