Special Issue Information

Dear Colleagues,

In the last decade, a great deal of effort has been made to investigate the role of protein homeostasis in neurodegenerative disorders. Protein homeostasis or "proteostasis" represents the process that regulates the balance of the intracellular functional and "healthy" proteins. Proteostasis is fundamental to preserving physiological metabolic processes in the cell and it enables the cell to respond to any given stimulus.

Increasing evidence has recently shown that oxidative stress (OS), associated with mitochondrial dysfunction and with the failure of antioxidant responses, is an early signature of neurodegenerative disorders, promoting protein oxidation and the formation of toxic protein aggregates. In turn, systems involved in the surveillance of protein synthesis/folding/degradation mechanisms, such as the integrated stress response (ISR), the unfolded stress response (UPR), and autophagy are impaired in neurodegenerative disorders.

Targeting altered protein homeostasis and energy reservoirs is now a major focus of future research and could be more advantageous and effective in neurodegenerative disorders.
