Title: Single-cell techniques in environmental microbiology
This is an interesting mini review paper or communication as the authors stated. The authors provide an overview of single-cell techniques for the isolation and identification of individual "unculturable" microorganisms from nature that could possibly be used for biological wastewater treatment. Regarding the review of the methods mentioned in chapter 2, I have no objections. Furthermore, in the third chapter, the authors give Applications in environmental microbiology, which is also meaningful.
However, the authors should at least briefly refer to possible applications in the treatment of waste water, or the decomposition of organic pollutants such as the PAH compounds they mentioned.
In my opinion, the purpose would be to isolate and identify a microorganism from nature that decompose a contaminant. So this is natural decomposition or remediation, "in-situ". After isolation, this microbial culture could be used for "ex-situ" remediation, the decomposition under controlled conditions of some wastewater contaminated with organic pollutants.
In this sense, briefly expand the manuscript to get closer to the topic of the mentioned Special Issue.

The article focuses on relevant issues but I find some unclarities. Therefore major revision is needed.
My remarks are :
1. Please add a table comparing achievements in different analyses, Please compare it.
2. Add also in introduction reviews in the field made.
3. Why the review of the topic is important. Add more reasons for the research.
4. Add regression of error part. Please, there compare differences of error in measuring and analysis.
