Round 1

Reviewer 1 Report

  1. Title: Change “selected thermotolerant yeast strain” with “Kluyveromyces marxianus SS106”
  2. The main issues reported in the introduction (Lines 37-45) are deeply investigated in the recent Review which could be added as reference: Processes 2020, 8(7), 768 https://doi.org/10.3390/pr8070768
  3. The introduction lacks of a literature analysis about previously works on this sequential methodology or similar
  4. Materials and method: Raw material- Could be possible add the compositional analysis of the used feedstock? (%cell %hemic %Lign….)
  5. Figure 3 Nominate right axis
  6. Table 2 explain the superscript a,b,c.
  7. Table 3: See previously comment
  8. Conclusion explain better the advantages obtained with this technique

Reviewer 3 Report

The manuscript by Morakot Krajang et al. deals with the isolation of thermotolerant yeast strains to be applied in the production of ethanol from  raw cassawa starch. The isolate SS106, identified as K. marxianus, was selected among 15 isolates for further analyses. The authors consider SS106 as a suitable strain for single-step ethanol production using a combination of raw cassava starch hydrolysis and fermentation.

I have found both the introduction and discussion too long. Please, shorten them and be more focused on the topic and obtaned results.

The manuscript contains a lot of typesetting errors and, most importantly, English grammar must be strongly revised.

A work-flow figure would better explain and clarify the entire experimental procedure, from yeast isolation to process optimization.

line 53, "S. cerevisiae" in italics

line 84, "a DE of 10-13..." please, use full words at first mention.

line 84, "According to our studies...", please add references

lines 229-232, what about methods?

line 230, K. is Kluyveromyces
