Exercise training is a non-pharmacological strategy known to modulate these molecular alterations. This study investigated the effects of high-intensity interval training (HIIT) on transcriptomic changes in the cerebral cortex of spontaneously hypertensive rats (SHR) fed a high-fat diet (HFD). Rats were assigned to either a HIIT intervention group (HIIT-HFD-SHR) or a sedentary control group (HFD-SHR). Cortical RNA was extracted, sequenced using the Illumina NovaSeq 6000 platform, and analyzed with DESeq2. Functional enrichment was conducted using Metascape. RNA-seq identified 1223 differentially expressed genes (DEGs) (adjusted p < 0.05), with 51 remaining significant under stringent criteria (adjusted p < 0.001, |log2FC| > 0.5). Among these, eight key genes were closely associated with the regulation of apoptosis and autophagy, including seven downregulated (Egr1, Atf3, Tgm2, Lgals1, Nr4a1, Plekhf1, Nupr1) and one upregulated (Trim39). This transcriptomic analysis following HIIT also modulated circadian rhythm, long-term memory processes, and hypoxia response in the hypertensive brain. These findings indicate that HIIT decreases apoptosis and autophagy and improves circadian rhythm, long-term memory, and hypoxia in hypertensive rats’ brains. Full article

Traditional colloid SERS substrates are mostly based on metal nanoparticles (MNPs), which have complex and time-consuming fabrication processes, poor structural control, and are susceptible to oxidation. As a result, solid-state SERS substrates have emerged as an effective alternative. Here, we propose using two-step mild and hard anodization to fabricate ordered anodic aluminum oxide (AAO) substrates with high total pore circumference for SERS detection. Hybrid pulse anodization (HPA) enables the fabrication of AAO at room temperature using 40 V in the first step and 40, 110, and 120 V in the second step of anodization. The different voltages applied in the second step effectively control the pore diameter, thereby achieving various nanostructures. The enhancement mechanism primarily originates from the high total pore circumference of nanostructures, which generates abundant hot spots around the pore peripherals, thereby significantly amplifying the SERS signal. Sorbic acid is a common preservative widely used in food products and employed as a test substance on high regularity AAO substrates at concentrations of 1000 ppm to 10 ppb. The resulting SERS spectra exhibited distinct characteristic peaks at 1640–1645 cm−1. The analytical enhancement factor is calculated as 1.02 × 105 at the AAO substrate prepared by 110 V with the Si substrate as the reference. By appropriately tuning the process parameters, a limit of detection (LOD) as low as 10 ppb of sorbic acid was achieved. Full article

Our previous study demonstrated that thiophene-substituted synthetic curcumin analogs possessed better antibacterial activity and stability than natural curcumin, demethoxycurcumin, or bisdemethoxycurcumin in antibacterial photodynamic therapy (aPDT). In addition, the activity of the furan-substituted analogs was weaker than that of the thiophene-substituted compounds. As oxygen, sulfur, and selenium belong to the same group in the periodic table, the antibacterial and anticancer activities of these three different elemental analogs were compared and investigated. The thiophene-substituted analog (compound 3) exhibited the most potent antibacterial activity in aPDT experiments. However, the furan-substituted analog (compound 1) exhibited the most potent anticancer activity. These results indicate that the differences in atomic radii or energy levels in these compounds produce different cell-attack results on generated free radicals. Ruthenium(II) complexes have a good reputation for use in PDT for cancer treatment. Our results show that complexation of ruthenium(II) with thiophene-substituted curcumin analogs does not enhance their antibacterial or anticancer activity. Full article

Thioester-containing proteins (TEPs), which are distinguished by the thioester motif (GCGEQ), are essential to arthropods’ defense against infections. Although TEPs have been extensively investigated in Anopheles, Aedes, and Drosophila, their functions in Culex mosquitoes remain inadequately explored. Interestingly, we discovered that Culex TEPs exhibit functional antagonism to their orthologs in other species, actively facilitating viral infection in this vector. In this study, we identified nine TEP genes in Culex quinquefasciatus, three of which were found to critically facilitate Japanese encephalitis virus (JEV) infection, with CqTEP27 exhibiting the most pronounced proviral effect. Mechanistically, CqTEP27 may have suppressed the production of several antimicrobial peptides (AMPs), which increased JEV replication. Our work also highlights the potential of targeting susceptibility factors such as CqTEP27 to block pathogen acquisition. Notably, the rate of mosquito infection was significantly decreased by membrane blood feeding antisera against CqTEP27. Therefore, vaccination against CqTEP27 offers a workable method of avoiding JEV infection. According to our research, CqTEP27 is a promising target for the development of vaccines that prevent JEV transmission. By preventing viral infection in mosquitoes that feed on immunized hosts, this approach can directly disrupt the natural transmission cycle, offering a novel strategy to reduce the disease burden. Coal gasification slag (CGS), an industrial solid waste produced during high-temperature (1200–1600 °C) coal gasification, was utilized as the primary raw material, combined with minor additions of coal gangue and calcium oxide, to synthesize ceramsite filter via high-temperature sintering (900–1160 °C) for phosphorus-containing wastewater treatment. The resulting ceramsite was evaluated for compressive strength, apparent porosity, water absorption, mineral phase composition, hydrolysis properties, and phosphorus removal performance. Experimental results revealed that increasing sintering temperature and calcium oxide content shifted the dominant crystalline phases from anorthite and hematite to gehlenite, anorthite, wollastonite, and esseneite, promoting the formation of porous structures. This transition increased apparent porosity while reducing compressive strength. Under optimal conditions (1130 °C, 20 wt.% CaO, 1 h sintering), the ceramsite (CM-20-1130) exhibited an apparent porosity of 43.12%, compressive strength of 3.88 MPa, apparent density of 1.084 g/cm3, and water absorption of 33.20%. The high porosity and abundant gehlenite and wollastonite phases endowed CM-20-1130 with enhanced hydrolysis capacity. Static phosphorus removal experiments demonstrated a maximum phosphorus removal capacity of 2.77 mg/g, driven by the release of calcium and hydroxide ions from gehlenite and wollastonite, which form calcium-phosphate precipitates on the ceramsite surface, enabling efficient phosphorus removal from simulated wastewater. Background: Persistent rotational instability and graft failure remain major challenges following anterior cruciate ligament reconstruction (ACLR). The addition of anterolateral ligament reconstruction (ALLR) and suture tape augmentation may enhance graft protection and early stability. This study aimed to assess the short-term clinical outcomes of combined ACLR and ALLR with suture tape augmentation, hypothesizing that this technique would yield favorable results in patients with ACL injury. Methods: Twenty-four patients (16 males, 8 females; mean age 30.8 years) with high grade pivot shift who underwent combined ACLR and ALLR with suture tape augmentation between 2022 and 2024 were retrospectively reviewed. Objective assessments included pivot-shift grading and anterior tibial translation (ATT) using a GNRB arthrometer. Patient-reported outcome measures (PROMs) comprised the International Knee Documentation Committee (IKDC), Marx activity score, and Single Assessment Numeric Evaluation (SANE) scores. Subgroup analysis compared graft diameters < 8 mm versus ≥8 mm. Results: The mean follow-up period was 23.4 months. Significant improvements were observed postoperatively in IKDC (45.9→83.7), Marx (4.4→9.9), SANE (49.2→85.2), and ATT (4.7→1.1 mm) (all p < 0.001). Full range of motion was regained at a mean of 3.2 months. Twenty-one patients presented no pivot shift and three with grade one postoperatively. No significant differences were found between the small (<8 mm) and large (≥8 mm) graft groups. MRI at one year showed intact grafts without adverse reactions. Conclusions: Combined ACL and anterolateral ligament reconstruction with suture tape augmentation demonstrated promising short-term stability and functional recovery without significant complications. This technique may enhance biomechanical reinforcement of both ACL and ALL grafts and represents a viable option for patients with smaller graft diameters. Full article

The gear transmission system is a safety-critical component in rail transit, typically designed for a service life exceeding 20 years. Failure analysis of such systems remains a key focus for railway engineers. This study systematically investigates four representative cases of premature gear failure in high-speed trains using a standardized analytical procedure that includes visual inspection, chemical analysis, metallographic examination, scanning electron microscopy, and hardness testing. The results identify four primary root causes: subsurface slag inclusions in raw materials, inadequate heat treatment leading to a non-martensitic layer (∼60 μm) at the tooth root, grinding-induced temper burns (crescent-shaped "black spots") accompanied by a hardness drop of ∼100–150 HV, and insufficient lubrication. The interdependencies between these factors and failure mechanisms, e.g., fatigue cracking, spalling, and thermal scuffing, are analyzed. This work provides an evidence-based framework for improving gear reliability and proposes targeted countermeasures, such as ultrasonic inclusion screening and real-time grinding temperature control, to extend operational lifespans. Using Landsat series imagery and the deep learning model CITNet, this study conducted high-accuracy classification and spatiotemporal change analysis of wetlands on Chongming Island from 2000–2020 and explored the driving mechanisms by integrating climatic and anthropogenic factors. The results demonstrate that the total wetland area decreased by approximately 125.5 km2 over the two decades. Among natural wetlands, tidal mudflats and shallow seawater zones continuously shrank, while herbaceous marshes exhibited a “decline recovery” trajectory. Artificial wetlands expanded before 2005 but contracted significantly thereafter, mainly due to aquaculture pond reduction. Wetland transformation was dominated by wetland-to-non-wetland conversions, peaking during 2005–2010. Driving factor analysis revealed a “human pressure dominated, climate modulated” pattern: nighttime light index (NTL) and GDP demonstrated strong negative correlations with wetland extent, while minimum temperature and the Palmer Drought Severity Index (PDSI) promoted herbaceous marsh expansion and accelerated artificial wetland contraction, respectively. The findings indicate that wetland changes on Chongming Island result from the combined effects of policy, economic growth, and ecological processes. Sustainable management should focus on restricting urban expansion in ecologically sensitive zones, optimizing water resource allocation under drought conditions, and incorporating climate adaptation and invasive species control into restoration programs to maintain both the extent and ecological quality of wetlands. Long COVID, characterized by persistent symptoms following acute SARS-CoV-2 infection, has emerged as a significant public health challenge with wide-ranging clinical and socioeconomic implications. Developing an effective risk assessment strategy is essential for the early identification and management of individuals susceptible to prolonged symptoms. This study uses a quantitative approach to characterize the dose–response relationships between spike protein concentrations and effects, including Long COVID symptom numbers and the release of proinflammatory mediators. A mathematical model is also developed to describe the time-dependent change in spike protein concentrations post diagnosis in twelve Long COVID patients with a cluster analysis. Based on the spike protein concentration–Long COVID symptom numbers relationship, we estimated a maximum symptom number (~20) that can be used to reflect a persistent predictor. We found that among the crucial biomarkers associated with Long COVID proinflammatory mediator, CXCL8 has the lowest 50% effective dose (0.01 μg mL−1), followed by IL-6 (0.39), IL-1β (0.46), and TNF-α (0.56). This work provides a comprehensive risk assessment strategy with dose–response tools and mathematical modeling developed to estimate potential spike protein concentration. Our study suggests persistent Long COVID guidelines for personalized care strategies and could inform public health policies to support early interventions that reduce long-term disability and healthcare burdens with possible other post-infection syndromes. Atrial fibrillation (AF) is a common arrhythmia associated with major adverse cardiovascular events. Early detection and short-horizon risk prediction are therefore clinically critical. Prior attention-based electrocardiogram (ECG) models typically treated subtype classification and short-horizon onset risk prediction as separate tasks and optimized attention in only one representational dimension rather than in a coordinated hierarchy. We propose a hierarchical multiattention temporal fusion network (HMA-TFN). The proposed framework jointly integrates lead-level, morphology-level, and rhythm-level attention, enabling the model to simultaneously highlight diagnostically informative leads, capture waveform abnormalities, and characterize long-range temporal dependencies. Moreover, the model is trained for dual tasks—AF subtype classification and 30-min onset prediction. Experiments were conducted on three open-source databases and the Fuzhou University–Fujian Provincial Hospital (FZU-FPH) clinical database, comprising thousands of dual-lead ECG recordings from a diverse subject population. Experimental results show that HMA-TFN achieves 95.77% accuracy in classifying paroxysmal AF (PAAF) and persistent AF (PEAF), and 96.36% accuracy in predicting PAAF occurrence 30 min in advance. Ablations show monotonic gains as each attention level is added, delivering 14.0% accuracy over the baseline for subtyping and 5.2% for prediction. Grad-CAM visualization highlights clinically relevant features such as absent P-waves, confirming model interpretability. On the FZU-FPH clinical database, it achieves a generalization performance of 94.31%, demonstrating its strong potential for clinical application. Full article

In recent years, the artificial intelligence (AI) + ECG paradigm has attracted considerable interest, but the two intrinsic characteristics of the ECG, namely, inter-lead correlations and multi-label classification, are often overlooked. Given that this oversight may constrain the full potential of AI models to enhance diagnostic performance, this study focuses on investigating methods for fusing information from a 12-lead ECG. A series of comprehensive experiments was conducted to evaluate the performance of various lead configurations, that is, 1-, 3-, 6-, 9-, and 12-lead combinations, with different fusion strategies. Innovatively integrating medical theory, we propose a novel five-lead-grouping strategy and develop a neural network architecture named Lead-5-Group Net (L5G-Net). After ranking the 12 leads with the AUC, we found that the aVR, V5, and V6 leads are particularly informative for single-lead ECG diagnosis. Furthermore, in multi-lead ECG classification, adopting an orthogonal lead-selection strategy which is based on the hypothesis of spatial interdependence among ECG leads was shown to enhance performance by ensuring that the information provided by each lead is complementary. Finally, the proposed L5G-Net demonstrates outstanding performance, achieving a macro-AUC of 0.9357 on the PTB-XL multi-label dataset without the use of data augmentation, attention mechanisms, or other strategies. Furthermore, considerable performance gains were observed after the five-lead-grouping strategy was applied to DenseNet and ResNet. This study investigated the anti-obesity effects of a homogenized, viscous vegetable (VV) mixture prepared from mucilaginous vegetables, with a focus on modulating hepatic lipid metabolism and gut microbiota composition in mice fed with a high-fat (HF) diet. The VV mixture was formulated by blending freeze-dried powders of ten mucilaginous vegetables, classified as moderately thick using a line-spread test and extremely thick according to the IDDSI framework in a 1:9 ratio (VV mixture: water, w/w). Six-week-old male C57BL/6 mice were fed control or HF diets, with or without 10% VV mixture for 8 weeks (n = 7 per group). The HF diet induced significant weight gain, adipose tissue accumulation, hepatic steatosis, and inflammation. The HF diet also significantly reduced hepatic ACO1, CPT1 mRNA expression, and α-diversity with distinct fecal microbiota profiles. On the other hand, VV mixture supplementation reduced serum TC, LDL-C levels and NAFLD scores. VV mixture supplementation also increased hepatic ACO1 and CPT1 mRNA expression, enhanced α-diversity, and enriched SCFA-producing bacteria, particularly the Lachnospiraceae NK4A136 group. In conclusion, the VV mixture attenuated HF diet-induced obesity, possibly through its high viscosity–mediated effects on hepatic fatty acid oxidation and gut microbiota modulation. Full article

This study investigates premature fatigue failures in three EA1N steel axles from permanent magnet direct-drive locomotives during wheel-seat bending tests. Complete fracture occurred in one axle at 3 million cycles, and in the other two axles, cracks appeared and were observed through magnetic particle detection at 3.5 million and 1.6 million cycles, respectively. A comprehensive failure analysis was conducted through metallurgical examination, fractography, mechanical testing, residual stress measurement, and finite element analysis. The fractographic results revealed fractures consistently initiated at the wheel-seat to axle-body transition arc, exhibiting characteristic ratchet marks and beach patterns. The premature fracture mechanism was identified as a high-stress fatigue fracture. The residual stress measurements showed detrimental tensile stresses at the surface. Coupled with the operating stress, the stress on the axle exceeds fatigue strength, which accelerates the initiation and propagation of fatigue cracks. Based on these observations, the failure mechanism was identified, and preventive methods were proposed to reduce the risk of recurrence of the in-service axles. Full article
