Cover Story: Hepatic ischemia-reperfusion injury (IRI) remains a major challenge in liver surgery and transplantation, contributing to graft dysfunction and adverse clinical outcomes. Its complex pathophysiology requires reliable experimental models to investigate underlying mechanisms and identify potential therapeutic targets. This review critically examines conventional 2D cell cultures and animal models, discussing their strengths, limitations, and translational relevance. It also explores the potential of advanced 3D platforms, including organoids and multicellular systems, to better recapitulate the complexity of human hepatic IRI. By highlighting current challenges and future opportunities, this review provides a perspective on the development of more physiologically relevant experimental models. View this paper
