Cover Story: Oxidative stress in the RPE/retina has a critical role in the pathogenesis of AMD. Paraoxonase 2 (PON2) is an antioxidant protein with lactonase activity. We show that PON2 is expressed in hRPE cells including mitochondria and is regulated by stress. Knockdown of PON2 causes mitochondrial dysfunction by altering mitochondrial bioenergetics. NaIO3-induced stress causes significant retinal damage and function, which is exacerbated in PON2-deficient mice. Our findings suggest that PON2 may play a beneficial role in the pathophysiology of retinal diseases. View this paper
