The recent birth of genetically modified canids phenotypically resembling the extinct dire wolf (Aenocyon dirus) was hailed as a landmark in synthetic biology. Using genome editing and cloning, the biotech company Colossal Biosciences created three such animals from gray wolf cells, describing the project as an effort in “functional de-extinction”. This case raises significant questions regarding animal welfare, moral justification, and regulatory governance. We used the five domains model framework to assess the welfare risks for the engineered animals, the surrogate mothers used in reproduction, and other animals potentially affected by future reintroduction or escape scenarios. Ethical implications are examined through utilitarian, deontological, virtue, relational, and environmental ethics. Our analysis suggests that the project suffers from ontological ambiguity: it is unclear whether the animals created are resurrected species, hybrids, or novel organisms. While the current welfare of the engineered animals may be manageable, their long-term well-being, particularly under rewilding scenarios, is likely to be compromised. The moral arguments for reviving long-extinct species are weak, particularly in cases where extinction was not anthropogenic. Legally, the current EU frameworks lack the clarity and scope to classify, regulate, or protect genetically engineered extinct animals. We recommend that functional de-extinction involving sentient beings be approached with caution, supported by revised welfare tools and regulatory mechanisms. Full article

This paper investigated and experimentally analyzed the penetration behavior of liquid copper along austenite grain boundaries (GBs) at high temperatures. The microstructure of the liquid copper channel network along GBs and triple junctions (TJs), as well as the TJ wetting, was observed and interpreted through diffusion-controlled premelted GB formation. The concentration distribution results along GBs show that copper diffusion in both the near-surface premelted GBs and the non-surface-layer solid-state GBs conform to the diffusion equation, though the diffusion coefficients differ by approximately one order of magnitude. However, the copper concentration at premelted GBs cannot be fully described by an error solution. Using a modified diffusion equation when considering the concentration dependence of the GB diffusion coefficient provides a more accurate description, aligning better with experimental characteristics. Electron backscatter diffraction measurements indicate that the copper orientation at premelted GBs remains consistent with that of surface copper coating, whereas that at solid-state GBs undergoes significant changes. This finding is consistent with the argument that the corresponding material states at premelted GBs are different from those at solid-state GBs, thus providing experimental evidence for the diffusion equation solutions presented above. It provides a theoretical reference for understanding and preventing liquid metal embrittlement. Full article

The interdisciplinary study of Daoism and literature can be broadly categorized into three main approaches: analyzing literary works as sources of Daoist material, examining Daoist scriptures as literary texts, and exploring the influence of Daoist beliefs on writers. This paper proposes a new perspective that complements these existing frameworks: by examining Daoist scriptures, we can better assess whether certain literary expressions or rhetorical devices were innovative or simply widespread conventions among writers of a particular period. Using the works of Lu Xiujing 陸修靜 (406–477 CE) as a case study, this paper builds on Haun Saussy’s argument that the use of “fragrance” as a rhetorical device to modify virtue—often surprising to modern scholars—was, in fact, a familiar trope for ancient Chinese writers. However, the paper critiques Saussy’s reliance on the works of the famous poet Qu Yuan 屈原 (c. 342–278 BCE) as evidence. Unlike Qu Yuan, who was primarily a literary figure, Lu Xiujing, as a Daoist scholar, frequently employed metaphors related to smell and taste to express abstract moral or doctrinal concepts. For Lu and his contemporaries, such expressions were not regarded as remarkable literary techniques but rather as conventional modes of discourse. This suggests that, at least in early medieval China, such rhetorical usage among writers was not seen as novel but as commonplace. Full article

This study was aimed at elucidating the role of the carboneous component in Co–carbon-containing catalysts for the hydrogenation of organic compounds. A Co-C/SiO2 catalyst was synthesized via pyrolysis of a Co(II) complex with 1,2-diaminobenzene on silica and subsequently irradiated with 2.3 MeV electrons, producing Co-C/SiO2*. A comprehensive characterization using XRD, TEM, IR, and Raman spectroscopy indicated minimal structural changes in the cobalt nanoparticles and carboneous materials. However, the Raman analysis revealed a slight decrease in the structural defect content in the carboneous component upon irradiation, which could occur due to defect healing. The catalytic tests, including the hydrogenation of 26 compounds, demonstrated an enhanced performance of the irradiated catalyst in 17 cases. This performance improvement was attributed to a modification of the carboneous material. It could be concluded that the carboneous components in Co-C composites contributed to their catalytic performance, probably by modifying the electronic structure of Co nanoparticles. In turn, the results provide arguments against the supposition that hydrogenation occurs on the structural defects of the carboneous material. Full article

Background/Objectives: One potential risk factor that remains especially contentious in the anterior cruciate ligament (ACL) injury literature is the role of neuromuscular fatigue in ACL injury risk. Therefore, the purposes of this review are (i) to present the research and practical concepts of lower extremity neuromuscular fatigue; and (ii) to review the literature relating to neuromuscular fatigue as an ACL injury risk factor and mechanism. Methods: A structured review was performed in the Medline database using a search strategy that included terms such as “anterior cruciate ligament injury” and “knee injuries” combined with terms such as “injury” and “fatigue”. Articles were included if they included young healthy participants (18–35) and made a comparison between non-fatigued and fatigued states that were assessed with at least one lower extremity biomechanical variable associated with ACL injury risk. Results: Overall, there were 67 studies included, accounting for 1440 participants (627 male and 813 female) across a variety of sports and activities. Of these, 53 (79%) reported a post-fatigue change in the kinematics, kinetics, neuromuscular, and/or other (e.g., proprioceptive) outcomes that indicate that the participants would be at an increased risk of an ACL injury. The most common argument against fatigue as a risk factor is that ACL injuries do not tend to occur later in a game or season, when it is assumed that athletes would be most fatigued. Conclusions: The evidence presented in this review suggests that localized neuromuscular fatigue is a risk factor, among multiple factors, for ACL injuries, providing another modifiable risk factor that should be considered when developing ACL injury risk reduction interventions. This paper investigates the dynamical behavior of a discrete fractional-order modified Leslie–Gower model with a Michaelis–Menten-type harvesting mechanism and a Holling-II functional response. We analyze the existence and stability of the nonnegative equilibrium points. For the interior equilibrium points, we study the conditions for period-doubling and Neimark–Sacker bifurcations using the center manifold theorem and bifurcation theory. To control the chaos arising from these bifurcations, two chaos control strategies are proposed. Numerical simulations are performed to validate the theoretical results. The findings provide valuable insights into the sustainable management and conservation of ecological systems. Full article

Low transitive constructions are ubiquitous in English conversation and as such can be considered “typical” clauses. This article furthers this claim by showing that these constructions are also most frequent in a different genre: arguments between participants at organized protests. It has been argued that one reason these constructions are so frequent is that they function to display participants’ stances. Arguments are a type of interaction where stance displays abound. In fact, they are defined as sequences of utterances that display opposing stances. Thus, the study goes on to examine how the most frequent of the low transitive constructions in the data—clauses with the nonverbal predicate be—function to display opposition across utterances. Du Bois has analyzed stance as resonance across utterances, created from structural parallelism, and he argues that slight changes between linguistic forms can create differing focal points that index contrasts. This framework is used to analyze how participants use the multiple semantic functions of be clauses across interactional sequences to display and modify their stances in response to their opponent. It is suggested that the versatility and ambiguity of be clauses are especially useful in arguments where participants do not share a set of common beliefs. Full article

We have collected theoretical arguments supporting the functional role of nano-metallic coatings of solar cells, which enhance solar cell efficiency via by plasmon-strengthening the absorption of sun-light photons and reducing the binding energy of photoexcitons. The quantum character of the plasmonic effect related to the absorption of photons (called the optical plasmonic effect) is described in terms of the Fermi golden rule for the quantum transitions of semiconductor-band electrons induced by plasmons from a nano-metallic coating. The plasmonic effect related to the lowering of the exciton binding energy (called the electrical plasmonic effect) is of particular significance for metalized perovskite solar cells and is also characterized in quantum mechanics terms. The coupling between plasmons in nanoparticles from a coating with band electrons in a semiconductor substrate significantly modifies material properties (dielectric functions) both of the particles and the semiconductor, beyond the ability of the classical electrodynamics to describe. Hereditary sensory and autonomic neuropathy type 1 is an autosomal dominant neuropathy caused by the SPTLC1 or SPTLC2 variants. These variants modify the preferred substrate of serine palmitoyl transferase, responsible for the first step of de novo sphingolipids synthesis, leading to accumulation of cytotoxic deoxysphingolipids. Diagnosis of HSAN1 is based on clinical symptoms, mainly progressive loss of distal sensory keep, and genetic analysis. Aim: Identifying new SPTLC1 or SPTLC2gain-of-function” variants raises the question as to their pathogenicity. This work focused on characterizing six new SPTLC1 variants using in silico prediction tools, new meta-scores, 3D modeling, and functional testing to establish their pathogenicity. Methods: Variants from six patients with HSAN1 were studied. In silico, CADD and REVEL scores and the 3D modeling software MITZLI were used to characterize the pathogenic effect of the variants. Functional tests based on plasma sphingolipids quantification (total deoxysphinganine, ceramides, and dihydroceramides) were performed by tandem mass spectrometry. Results: In silico predictors did not provide very contrasting results when functional tests discriminated the different variants according to their impact on deoxysphinganine level or canonical sphingolipids synthesis. Two SPTLC1 variants were newly described as pathogenic: SPTLC1 NM_006415.4:c.998A>G and NM_006415.4:c.1015G>A. Discussion: The combination of the different tools provides arguments to establish the pathogenicity of these new variants. When available, functional testing remains the best option to establish the in vivo impact of a variant. Moreover, the comprehension of metabolic dysregulation offers opportunities to develop new therapeutic strategies for these genetic disorders. Full article

A novel rare mutation in the pore region of Nav1.5 channels (p.L889V) has been found in three unrelated Spanish families that produces quite diverse phenotypic manifestations (Brugada syndrome, conduction disease, dilated cardiomyopathy, sinus node dysfunction, etc.) with variable penetrance among families. We clinically characterized the carriers and recorded the Na+ current (INa) generated by p.L889V and native (WT) Nav1.5 channels, alone or in combination, to obtain further insight into the genotypic–phenotypic relationships in patients carrying SCN5A mutations and in the molecular determinants of the Nav1.5 channel function. The variant produced a strong dominant negative effect (DNE) since the peak INa generated by p.L889V channels expressed in Chinese hamster ovary cells, either alone (−69.4 ± 9.0 pA/pF) or in combination with WT (−62.2 ± 14.6 pA/pF), was significantly (n ≥ 17, p < 0.05) reduced compared to that generated by WT channels alone (−199.1 ± 44.1 pA/pF). The mutation shifted the voltage dependence of channel activation and inactivation to depolarized potentials, did not modify the density of the late component of INa, slightly decreased the peak window current, accelerated the recovery from fast and slow inactivation, and slowed the induction kinetics of slow inactivation, decreasing the fraction of channels entering this inactivated state. The membrane expression of p.L889V channels was low, and in silico molecular experiments demonstrated profound alterations in the disposition of the pore region of the mutated channels. Despite the mutation producing a marked DNE and reduction in the INa and being located in a critical domain of the channel, its penetrance and expressivity are quite variable among the carriers. Our results reinforce the argument that the incomplete penetrance and phenotypic variability of SCN5A loss-of-function mutations are the result of a combination of multiple factors, making it difficult to predict their expressivity in the carriers despite the combination of clinical, genetic, and functional studies. We explore the dependence of vacuum energy on the boundary conditions for massive scalar fields in (2 + 1)-dimensional spacetimes. We consider the simplest geometrical setup given by a two-dimensional space bounded by two homogeneous parallel wires in order to compare it with the non-perturbative behaviour of the Casimir energy for non-Abelian gauge theories in (2 + 1) dimensions. Our results show the existence of two types of boundary conditions which give rise to two different asymptotic exponential decay regimes of the Casimir energy at large distances. The two families are distinguished by the feature that the boundary conditions involve or not interrelations between the behaviour of the fields at the two boundaries. Non-perturbative numerical simulations and analytical arguments show such an exponential decay for Dirichlet boundary conditions of SU(2) gauge theories. The verification that this behaviour is modified for other types of boundary conditions requires further numerical work. Subdominant corrections in the low-temperature regime are very relevant for numerical simulations, and they are also analysed in this paper. Full article

Solar photovoltaics (PV) continues to grow rapidly across the world and now accounts for a very considerable proportion of all non-fossil-fuel electricity. With the continuing urgency of greenhouse gas abatement, the growth of solar PV is inevitable. Competition with other land uses and the desire to optimize the efficiency of the panels by making use of water cooling are compelling arguments for offshore floating PV (OFPV), a trend that could also benefit from the existing infrastructure recently built for offshore wind farms. Building on our earlier work, we present a larger dataset (n = 82) located around the globe to assess global yield (dis)advantages while also accounting for a modified form of water cooling of the offshore panels. Using our results regarding the Köppen–Geiger (KG) classification system and using a statistical learning method, we demonstrate that the KG climate classification system has limited validity in predicting the likely gains from OFPV. Finally, we also explore a small subset of sites to demonstrate that economics, alongside geography and technology, impacts the feasibility of locating PV panels offshore. Full article

We present a modified version of the Szilard engine, demonstrating that an explicit measurement procedure is entirely unnecessary for its operation. By considering our modified engine, we are able to provide a new interpretation of Landauer’s original argument for the cost of erasure. From this view, we demonstrate that a reset operation is strictly impossible in a dynamical system with only conservative forces. Then, we prove that approaching a reset yields an unavoidable instability at the reset point. Finally, we present an original proof of Landauer’s principle that is completely independent from the Second Law of thermodynamics. Full article

In this work, we develop a new iterative method for computing the digits of π by argument reduction of the tangent function. This method combines a modified version of the iterative formula for π with squared convergence that we proposed in a previous work and a leading arctangent term from the Machin-like formula. The computational test we performed shows that algorithmic implementation can provide more than 17 digits of π per increment. Mathematica codes, showing the convergence rate for computing the digits of π, are presented. Full article
