Genetic Variant of the Canine FGF5 Gene for the Hair Length Trait in the Akita: Utility for Hair Coat Variations and Welfare in Conservation Breeding

Background/Objectives: Variations in hair length are observed in many dog breeds, as determined by the canine FGF5 gene. Long-haired Akitas, which are disqualified under breeding standards of Akitas, are sometimes born to short-haired parents and may have been subjected to treatments compromising animal welfare. Here, we aimed to identify an FGF5 variant associated with hair coat variations in Akitas in Japan, and to assess how welfare of this breed can be improved by carefully planned breeding. Methods: DNA samples were obtained from 60 Akitas in 2021 (modern Akitas) and 73 Akitas in the 1970s and the 1980s (classic Akitas). Sanger sequencing was performed on all exons and exon–intron junctions of the FGF5 gene to determine the causative variant of long hair in Akitas. A real-time PCR assay was developed to genotype FGF5:c.578C>T in modern and classic Akitas. Using 54 dogs from modern Akitas, scores (1 to 10) of hair length were compared among the three genotypes (C/C, C/T, and T/T). Results: Sanger sequencing revealed that the canine FGF5:c.578C>T variant was associated with long hair in Akitas in Japan. Genotyping revealed that the frequency of the mutant T allele was 0.350 in modern Akitas, which was significantly higher (p < 0.001) than in classic Akitas (0.212). The three genotypes were not in Hardy–Weinberg equilibrium (HWE) in modern Akitas but were in HWE in classic Akitas. There were significant differences in hair length scores among the three genotypes (p < 0.001) and between the C/C and C/T genotypes (p < 0.005). There was no significant difference in the scores between male and female dogs. Conclusions: This study revealed that a causative variant that determines the long hair trait of Akitas in Japan was the FGF5:c.578C>T variant, which was inherited in an incompletely dominant manner. Akita dog breeders were more likely to select heterozygous C/T dogs based on the appearance of the hair coat for breeding dogs with an ideal fluffy hair coat. This might result in a high mutant T allele frequency and the production of undesired long-haired Akitas with T/T, which may create welfare problems. Genetic testing for this variant is necessary to improve welfare and conserve the Akita breed. Full article

Indoor horticulture requires a substantial quantity of electricity to meet crops extended photoperiodic requirements for optimal photosynthetic rate. Simultaneously, global electricity costs have grown dramatically in recent years, endangering the sustainability and profitability of indoor vertical farms and/or modern greenhouses that use artificial lighting systems to accelerate crop development and growth. This study investigates the growth rate and physiological development of cherry tomato plants cultivated in a pilot indoor vertical farm at the Agricultural University of Athens’ Laboratory of Farm Structures (AUA) under continuous and disruptive lighting. The leaf physiological traits from multiple photoperiodic stress treatments were analyzed and utilized to estimate the plant’s tolerance rate under varied illumination conditions. Four different photoperiodic treatments were examined and compared, firstly plants grew under 14 h of continuous light (C-14L10D/control), secondly plants grew under a normalized photoperiod of 14 h with intermittent light intervals of 10 min of light followed by 50 min of dark (NI-14L10D/stress), the third treatment where plants grew under 14 h of a load-shifted energy demand response intermittent lighting schedule (LSI-14L10D/stress) and finally plants grew under 13 h photoperiod following of a load-shifted energy demand response intermittent lighting schedule (LSI-13L11D/stress). Plants were subjected also under two different light spectra for all the treatments, specifically WHITE and Blue/Red/Far-red light composition. The aim was to develop flexible, energy-efficient lighting protocols that maintain crop productivity while reducing electricity consumption in indoor settings. Results indicated that short periods of disruptive light did not negatively impact physiological responses, and plants exhibited tolerance to abiotic stress induced by intermittent lighting. Post-harvest data indicated that intermittent lighting regimes maintained or enhanced growth compared to continuous lighting, with spectral composition further influencing productivity. Plants under LSI-14L10D and B/R/FR spectra produced up to 93 g fresh fruit per plant and 30.4 g dry mass, while consuming up to 16 kWh less energy than continuous lighting—highlighting the potential of flexible lighting strategies for improved energy-use efficiency. Speaker burn-in is a controversial practice in the audio world, based on the belief that new devices reach optimal performance only after a certain period of use. Supporters claim it improves component flexibility, reduces initial distortion, and enhances sound quality—especially in the low-frequency range. Critics, however, emphasize the lack of scientific evidence for audible changes and point to the placebo effect in subjective listening tests. They argue that modern manufacturing and strict quality control minimize differences between new and “burned-in” devices. This study cites a standard describing a preliminary burn-in procedure, specifying the exact conditions and duration required. Objective tests revealed slight changes in speaker impedance and amplitude response after burn-in, but these differences are inaudible to the average listener. Notably, significant variation was observed between speakers of the same series, attributed to production line tolerances rather than use-related changes. The study also explored aging processes in speaker materials to better understand potential long-term effects. However, subjective listening tests showed that listeners rated the sound consistently across all test cases, regardless of whether the speaker had undergone burn-in. Overall, while minor physical changes may occur, their audible impact is negligible, especially for non-expert users. Full article

Modern life, characterized by constant exposure to artificial light from electronic devices, such as light-emitting diodes (LEDs), disrupts the natural circadian rhythm and induces important metabolic changes. The impact of blue light exposure on male and female rat’s onset of puberty, hormonal and biochemical parameters was assessed by comparison between the four study groups: the control group (CTRL) maintained under normal light conditions, the group exposed to blue light from a mobile phone (MP), the group subjected to blue light from a computer screen (PC), and the group exposed to blue light from an LED lamp (LED). Both female and male rats exposed to PC and LED failed to thrive, with a significantly lower body weight intake than the CTRL group. All three distinct sources of blue light interfered with the cyclicity of the estrous cycle in female rats. A marked decrease in the number of complete estrous cycles and the highest incidence of incomplete cycles were noticed in the LED group. Elevated ALT, AST, glucose, and insulin levels were influenced in a gender-specific manner, and depending on the source of emitted light. Prolonged blue light exposure induces significant metabolic disruptions and possesses important future research potential in identifying explicit pathways regarding this environmental stressor. Manual assembly remains essential in modern manufacturing, yet the increasing complexity of customised production imposes significant cognitive burdens and error rates on workers. Existing Spatial Augmented Reality (SAR) systems often operate passively, lacking adaptive interaction, real-time feedback and a control system with gesture. In response, we present Gest-SAR, a SAR framework that integrates a custom MediaPipe-based gesture classification model to deliver adaptive light-guided pick-to-place assembly instructions and real-time error feedback within a closed-loop interaction instance. In a within-subject study, ten participants completed standardised Duplo-based assembly tasks using Gest-SAR, paper-based manuals, and tablet-based instructions; performance was evaluated via assembly cycle time, selection and placement error rates, cognitive workload assessed by NASA-TLX, and usability test by post-experimental questionnaires. Quantitative results demonstrate that Gest-SAR significantly reduces cycle times with an average of 3.95 min compared to Paper (Mean = 7.89 min, p < 0.01) and Tablet (Mean = 6.99 min, p < 0.01). It also achieved 7 times less average error rates while lowering perceived cognitive workload (p < 0.05 for mental demand) compared to conventional modalities. In total, 90% of the users agreed to prefer SAR over paper and tablet modalities. These outcomes indicate that natural hand-gesture interaction coupled with real-time visual feedback enhances both the efficiency and accuracy of manual assembly. By embedding AI-driven gesture recognition and AR projection into a human-centric assistance system, Gest-SAR advances the collaborative interplay between humans and machines, aligning with Industry 5.0 objectives of resilient, sustainable, and intelligent manufacturing. Background: Etiologic diagnosis in suspected viral meningitis is not always achievable, yet it can play a significant role in patient management. Our study aimed to do a comprehensive analysis of current practices regarding etiologic diagnosis in these cases and compare patients with and without known etiologic diagnosis in order to visualize if and how having an etiological diagnosis can impact patient management and/or outcome. Methods and results: We conducted a convergent mixed-methods study. Quantitative data was obtained from 118 patients hospitalized during a one-year period. There were 40.7% (n = 48) cases with unknown etiology. The length of hospitalization was longer in the group with unknown etiology vs. known etiology (12.6 days vs. 9.8 days p = 0.01). Thematic analysis was used for the qualitative approach in order to evaluate physicians’ overall perceptions regarding this subject. Conclusions: Our mixed-methods study shows that while clinicians consider etiologic diagnosis very important, it remains a diagnostic challenge even in modern times. Continued efforts are needed to optimize diagnostic strategies and address existing gaps in the etiologic workup of viral meningitis. There may be overlooked pathogens that have cost-efficient testing methods, like TBEV, that can be introduced in a testing protocol and may enhance patient management and reduce unnecessary hospital stays. High-speed trains have revolutionized modern transportation with their exceptional speeds, yet the essence of this technological breakthrough resides in the train’s wheels. These components are engineered to endure extreme mechanical stresses while ensuring high safety and reliability. In this paper, we selected the rim and web as representative components of the wheel and conducted a comprehensive and systematic study on their microstructure and mechanical properties. The wheels are typically produced through integral forging. To improve the mechanical performance of the wheel/rail contact surface (i.e., the tread), the rim is subjected to surface quenching or other heat treatments. This endows the rim with strength and hardness second only to the tread and lowers its ductility. This results in a more isotropic structure with improved fatigue resistance in low-cycle and high-cycle regimes under rotating bending. The web connects the wheel axle to the rim and retains the microstructure formed during the forging process. Its strength is lower than that of the rim, while its ductility is slightly better. The web satisfies current property standards, although the microstructure suggests further optimization may be achievable through heat treatment refinement. Full article

Obstructive sleep apnea (OSA) is a prevalent and heterogeneous sleep disorder associated with significant health and societal burdens. While continuous positive airway pressure (CPAP) remains the gold standard treatment, its limitations in adherence and patient tolerance have highlighted the need for alternative therapies. Positional therapy (PT), which targets apneas that occur predominantly in the supine position, has emerged as a promising option for individuals with positional OSA (POSA). This narrative review synthesizes the current literature on PT, examining its clinical indications, typologies, comparative efficacy with CPAP, oral appliances, and hypoglossal nerve stimulation, as well as data on adherence and barriers to long-term use. Traditional methods such as the tennis ball technique have largely been replaced by modern vibrotactile devices, which demonstrate improved comfort, adherence, and comparable short-term outcomes in selected POSA subjects. While PT remains inferior to CPAP in reducing overall AHI and oxygen desaturation, it performs favorably in terms of mean disease alleviation (MDA) and sleep continuity. Importantly, treatment effectiveness is influenced by both anatomical and non-anatomical traits, underscoring the need for accurate phenotyping and individualized care. PT should be considered within a broader patient-centered model that incorporates preferences, lifestyle, and motivational factors. Further research is needed to validate long-term efficacy, optimize selection criteria, and integrate PT into personalized OSA management strategies. Full article

Tripterygium wilfordii has extremely important pharmaceutical value in both traditional and modern medicine. The mitogenome of T. wilfordii was subjected to assembly and annotation with Nanopore long reads and Illumina short reads in this study. The mitogenome is 720,306 bp in length and is responsible for encoding 55 specific genes, including 35 protein-coding genes (PCGs), 17 transfer RNA (tRNA) genes, and 3 ribosomal RNA (rRNA) genes. Upon repetitive sequence analysis, 223 simple sequence repeats (SSRs), 24 long tandem repeats (LTRs), and 47 dispersed repetitive sequences (DRSs) were identified. The 24 common PCGs were used for phylogenetic analysis, which revealed that T. wilfordii is more closely related to Euonymus alatus. Moreover, mitochondrial plastid DNA (MTPT) analysis revealed eight MTPTs in the mitochondrial genome. Furthermore, 600 RNA-editing sites were detected in the protein-coding genes according to RNA-seq results. Among these genes, the ccmB gene contained the greatest number of sites, followed by the nad4 gene. This is the first study to report the T. wilfordii mitogenome and illustrate its linear structure. The findings of this study will help elucidate the evolution of the T. wilfordii mitogenome and facilitate its potential application in genetic breeding. Full article

Despite the progressive development of modern technology, there is a lack of adequate and reliable training and information materials. Analyses carried out in this area show that most drivers do not have the skills to use safety systems or acquire these skills while driving. Drivers often demonstrate an incomplete understanding of them and overestimate their skills, which can jeopardize road safety. In this context, the authors of the paper set out to develop a model for evaluating the effectiveness of practical training using a high-end simulator. The research goal of the paper was to objectively measure drivers’ progress in operating safety systems after receiving practical training conducted in the AS1200-6 driving simulator located at the Motor Transport Institute. The main findings indicate that the innovative model was highly effective, with 81% of participants achieving a level rated as “acceptable” or “very good”. The model enabled more accurate and objective assessments of skills in comparison with the traditional subjective assessment of trainers, especially where driver progress was not clearly evident. The results suggest that combining the data model with trainer evaluation can be an effective evaluation mechanism in ADAS training. Full article

Modern fly-by-wire (FBW) aircraft demand high-fidelity simulation systems for research and training, yet existing force-sensing solutions are often prohibitively expensive. This study presents the design, development, and validation of a low-cost, reconfigurable force-sensing sidestick. The system utilizes four strain-gauge load cells to capture pure pilot force inputs, integrated with a 6-DoF non-linear flight model. To evaluate its performance, a pitch-angle tracking task was conducted with 16 participants (pilots and non-pilots). Objective metrics revealed that the control strategy was a primary determinant of performance. Participants employing a proactive feedforward control strategy exhibited roughly an order of magnitude lower tracking-error variance than those relying on reactive corrections. Subjective assessments using the Cooper-Harper scale and NASA-TLX corroborated the objective data, confirming the sidestick’s ability to differentiate control techniques. This work demonstrates an open-source platform that makes high-fidelity FBW simulation accessible for academic research, pilot training, and human factors analysis at a fraction of the cost of commercial systems. Focusing on the Pārijāta Tree on the summit of Mount Sumeru, the centre of Asian cosmology, this study analyses its depictions and images. These include Chinese translations of Buddhist texts, the most notable depictions in the handscrolls from the Dūn Huáng and Harvard Art Museums, its representations in Japanese classical literature and early modern Japanese illustrations of Mount Sumeru. Finally, drawing from the discussions on trees in the Buddhist texts, A Forest of Pearls from the Dharma Garden [法苑珠林, Fǎ yuàn zhū lín], the study also addresses various issues surrounding tree felling, which are relevant to the current concerns of environmental protection. I argue that the Pārijāta Tree, positioned as the heavenly king of trees, holds significance as a core figure at the centre of the cosmos. The Pārijāta Tree can be said to serve as a metaphor for the supreme state pursued by Buddhist practitioners. Furthermore, this study suggests that issues related to Asian cosmology or worldviews should be pursued as important subjects in future research on environmental literature. Full article

Is Cardiopulmonary Fitness Related to Attention, Concentration, and Academic Performance in Different Subjects in Schoolchildren?

Background: The perceived importance of physical practice and its contribution to students’ academic success have evolved considerably throughout the history of the modern educational system. Aim: The purpose of this study was to understand the relationship between physical fitness (measured as VO2max) and cognitive abilities (attention and concentration) and academic performance in different subjects: sciences, letters, language, arts, and physical education. Method: Fifty Spanish male students who participated in extracurricular sports activities (mean age (SD): 11.59 ± 1.30; range: 9–15 years) were included in the analysis. The 6 min walk test was used to assess physical fitness (6MWT), while for selective attention and concentration, the students completed the D2 test, which is usually considered to analyse the visual ability to select the most relevant stimulus of an exercise and ignore precisely the most irrelevant stimuli. Results: Correlation the individual contribution analyses revealed no significant associations between VO2max and academic performance in sciences (r = 0.04, p = 0.77), humanities (r = 0.00, p = 0.98), language (r = 0.03, p = 0.83), or arts (r = 0.04, p = 0.76). Similarly, no relationship was found between VO2max and overall academic performance (r = 0.10, p = 0.46), or cognitive abilities. However, a small positive correlation was observed between VO2max and physical education scores. Conclusions: Physical fitness showed no significant association with cognitive abilities or academic performance in most subjects, although a small positive correlation with physical education scores was observed. These findings emphasise the importance of promoting physical activity for its health and physical benefits. However, future research should explore broader cognitive outcomes and include more diverse and representative samples. Full article

The Power of Old Hats: Rediscovering Inosine-EpPCR to Create Starting Libraries for Whole-Cell-SELEX

Shaking off the forgetfulness towards the methodological power of inosine-mediated error-prone PCR (epPCR), this study reintroduces an often-underappreciated method as a considerably powerful approach for generating aptamer libraries from a single decameric ATCG-repeat-oligonucleotide. The aim was to demonstrate that this simple way of creating sequence diversity was suitable for delivering functional starting libraries for a set of ten whole-cell-SELEX (Systematic Evolution of Ligands by Exponential Enrichment) processes. This epPCR method uses inosine to introduce targeted mutations, avoiding the need for commercial oligo pools or large-scale synthesis. We applied this method to a “universal aptamer” and subjected the three resulting libraries to two rounds of selection against ten diverse targets including probiotic and pathogenic bacteria (Gram-negative and -positive) as well as human cell lines. The enriched aptamers exhibited new binding specificities, demonstrating that the approach supports functional selection. Much like dusting off an old tool and finding it perfectly suited for a modern task, this work shows that revisiting established techniques can address current challenges in aptamer development. Our main finding is that epPCR provides a robust, cost-effective strategy for generating starting libraries and lowers the barrier for initiating successful SELEX campaigns. Full article

Serum Levels of IL-21 and IL-22 in Breast Cancer Patients—A Preliminary Study

Breast cancer is one of the most commonly diagnosed malignant tumours in women worldwide. Although modern medicine has led to advanced diagnostic methods and therapies that allow for increasingly effective treatment, the mechanisms underlying breast cancer development and progression remain the subject of intensive research. In the pathogenesis of this cancer, significant importance is attributed to interactions between tumour cells and the tumour microenvironment, in which soluble immune system mediators—cytokines—play a key role, including IL-21 and IL-22. These interleukins, by modulating the immune response, can both promote and inhibit tumour progression, and analysing their concentrations may prove helpful in diagnosis, disease progression prognosis, and the development of new therapies, including immunotherapy. The aim of this study was to determine the concentrations of IL-21 and IL-22 in a group of patients with invasive cancer, depending on the biological type of the tumour and its malignancy grade. The study involved 60 women with breast cancer and 20 women with benign breast lesions, and the analysis of IL-21 and IL-22 protein concentrations was performed using the enzyme-linked immunosorbent assay (ELISA) method. The analysis shows that the concentrations of IL-21 and IL-22 do not differ significantly depending on the malignancy grade of the tumour. However, a statistically significant negative correlation between the concentrations of IL-21 and IL-22 was observed exclusively in the group of patients with benign breast lesions. Due to the high heterogeneity of breast cancers, further research with a larger study group is necessary to better understand these parameters and possibly apply them clinically in patients with breast cancer.
