First Detection and Molecular Identification of Rhabditis (Rhabditella) axei from the Chinese Red Panda (Ailurus styani)

Rhabditis (Rhabditella) axei is a predominantly free-living nematode commonly found in sewage systems and decomposing organic matter. While primarily saprophytic, it has been documented as an opportunistic pathogen in human urinary and gastrointestinal tracts. The Chinese red panda (Ailurus styani), a rare and protected species in China, has not previously been reported as a host for Rhabditis (Rhabditella) spp. infections. This study reports the first documented occurrence of R. axei in red panda feces, unambiguously confirmed through integrative taxonomic approaches combining morphological and molecular analyses. The nematodes exhibited key morphological features consistent with R. axei, including a cylindrical rhabditiform esophagus, sexually dimorphic tail structures, and diagnostic spicule morphology. Molecular analysis based on 18S-ITS-28S rDNA sequencing confirmed their identity, showing >99% sequence similarity to R. axei reference strains (GenBank: PP135624.1, PP135622.1). Phylogenetic reconstruction using 18S rDNA and ITS rDNA sequences placed the isolate within a well-supported R. axei clade, clearly distinguishing it from related species such as R. blumi and R. brassicae. The findings demonstrate the ecological plasticity of R. axei as a facultative parasite capable of infecting non-traditional hosts and further highlight potential zoonotic risks associated with environmental exposure in captive wildlife populations. Our results emphasize the indispensable role of molecular diagnostics in accurately distinguishing morphologically similar nematodes within the Rhabditidae family, while providing essential baseline data for health monitoring in both in situ and ex situ conservation programs for this endangered species. Most research on domestic dog (Canis familiaris) behavior has focused on pets with restricted movement. However, free-ranging dogs exist in diverse cultural contexts globally, and their interactions with humans are less understood. Tourists can facilitate unrestricted dog movement into wilderness areas, where they may negatively impact wildlife. This study investigated which stimuli—namely, voice, touch, or food—along with inherent factors (age, sex, sociability) motivate free-ranging dogs to follow a human stranger. We measured the distance (up to 600 m) of 129 free-ranging owned and stray dogs from three villages in southern Chile as they followed an experimenter who presented them one of the above stimuli or none (control). To evaluate the effect of dog sociability (i.e., positive versus stress-related or passive behaviors), we performed a 30 s socialization test (standing near the dog without interacting) before presenting a 10 s stimulus twice. We also tracked whether the dog was in the company of other dogs. Each focus dog was video-recorded and tested up to three times over five days. Generalized linear mixed-effects models revealed that the food stimulus significantly influenced dogs’ motivation to follow a stranger, as well as a high proportion of sociable behaviors directed towards humans and the company of other dogs present during the experiment. Juveniles tended to follow a stranger more than adults or seniors, but no effects were found for the dog’s sex, whether an owner was present, the repetition of trials, the location where the study was performed, or for individuals as a random variable. This research highlights that sociability as an inherent factor shapes dog–stranger interactions in free-ranging dogs when food is given. In the context of wildlife conservation, we recommend that managers promote awareness among local communities and tourists to avoid feeding dogs, especially in the context of outdoor activities close to wilderness. Porcine circovirus type 3 (PCV3) has been detected in wild boars across many countries in Europe, Asia, and South America. However, data regarding the presence of porcine circoviruses in wild boars and ticks remain limited. In this study, we investigated the presence and genetic characteristics of PCV3 in wild boars and parasitizing ticks in Jiangsu, China. Samples, including whole blood, serum, tissues, feces, and oral fluids from wild boars, as well as ticks collected from 47 wild boars, were obtained between March 2021 and November 2022. PCR results indicated that 34.0% (16/47) of wild boars tested positive for PCV3, while ELISA detected 41.9% (18/43) seropositivity. RT-qPCR results showed that 7.2% (6/83) were positive for PCV3 in 83 analyzed tick samples, with all positive samples identified as Amblyomma testudinarium. The PCV3 genome obtained from wild boars was classified as PCV3a and was closely related to the strain identified in domestic pigs in Nanjing, Jiangsu Province. Collectively, these findings confirm the presence of PCV3 in wild boars in Jiangsu and suggest a possible link of PCV3 infection among domestic pigs, wild boars, and ticks, providing new insights into the transmission risk of PCV3 at wildlife–livestock–human interfaces and highlighting the genetic homology between strains from wild and domestic pigs. Canine distemper is caused by a morbillivirus similar to others that affect livestock and humans. The increase in host range and its persistence in wildlife reservoirs complicate eradication considerably. Canine distemper virus has been reported in wildlife in Mexico since 2007. Dogs were previously considered the main reservoirs, but high vaccination coverage in the USA has helped control the disease, and racoons (Procyon lotor) are now recognized as the main reservoirs of the agent in the USA, since they live in high densities in urban environments (peridomestic), where contact with domestic and wildlife species is common. Racoons are now considered to spread CDV in wildlife species and zoo animals. Mexico is home to at least two wildlife species that have been reported as carriers of the CDV infection in studies in the USA. Raccoons and Coyotes are distributed in several Mexican states and could play the same reservoir role as for the US. In addition, the increase in non-traditional pets expands the availability of susceptible individuals to preserve CDV in domiciliary and peri-domiciliary environments, contributing to the spread of the disease. Combined with incomplete vaccination coverage in domestic canids, this could contribute to maintaining subclinical infections. Infected pets with incomplete vaccination schedules could also spread CDV to other canines or wildlife coexisting species. In controlled habitats, such as flora and fauna sanctuaries, protected habitats, zoo collections, etc., populations of wildlife species and stray dogs facilitate the spread of CDV infection, causing the spilling over of this infectious agent. Restricting domestic pets from wildlife habitats reduces the chance of spreading the infection. Regular epidemiological surveillance and specific wildlife conservation practices can contribute to managing threatened species susceptible to diseases like CDV. This may also facilitate timely interventions in companion animals which eventually minimize the impact of this disease in both scenarios. Aim: The review discusses the circulation of CDV in wildlife populations, and highlights the need for epidemiological surveillance in wildlife, particularly in endangered wildlife species from Mexico. Through an extensive review of recent scientific literature about CDV disease in wildlife that has been published in local and international databases, the findings were connected with the current needs of information from a local to global perspective, and conclusions were made to broaden the context of Mexican epidemiological scenarios as closely related to the neighboring regions. The Eurasian lynx (Lynx lynx) is listed in CITES Appendix II and is protected under the Bern Convention and the EU Habitats Directive, yet it remains a frequent target of wildlife crime, highlighting the urgent need for reliable identification methods. This study focuses on determination and DNA quantification of the Lynx spp. using quantitative real-time PCR (qPCR). The Llynx Qplex quantification multiplex system effectively distinguishes Lynx spp. from other Feliformia species by targeting mitochondrial and nuclear markers. Additionally, we present the results of the developmental validation of the Llyn STRplex system for individual identification and databasing using six STR loci. This study followed ISFG recommendations for non-human DNA testing and developmental validation guidelines. Both systems demonstrate high sensitivity (5 pg genomic DNA for Llynx Qplex and 30 pg of mtDNA for Llyn STRplex) and high specificity to Lynx spp., confirmed by testing against 16 related Feliformia species. Robustness was evaluated, showing sensitivity to temperature variation, and both repeatability and reproducibility were successfully tested across replicates and conditions. Given that forensic casework often involves degraded and limited biological material, molecular tools must be both sensitive and specific to ensure accurate results. Developing precise and efficient tools is essential for supporting investigations of wildlife crime involving the Eurasian lynx, as well as efforts aimed at conserving the species. Full article

Drones are becoming increasingly valuable tools in wildlife studies due to their ability to access remote areas and offer high-resolution information with minimal human interference. Their application is, however, causing concern regarding wildlife disturbance. This review synthesizes the existing literature on how animals within terrestrial, aerial, and aquatic environments are impacted by drone disturbance in relation to operational variables, sensory stimulation, species-specific sensitivity, and physiological and behavioral responses. We found that drone altitude, speed, approach distance, and noise levels significantly influence wildlife responses, with some species exhibiting increased vigilance, flight responses, or physiological stress. Environmental context and visual cues are also involved in species detection of drones and disturbance thresholds. Although the short-term response to behavior change has been well documented, long-term consequences of repeated drone exposure remain poorly known. This paper identifies the necessity for continued research into drone–wildlife interactions, with an emphasis on the requirement to minimize disturbance by means of improved flight parameters and technology. The global spread of antimicrobial resistance (AMR) threatens to plummet society back to the pre-antibiotic era through a resurgence of common everyday infections’ morbidity. Thus, studies investigating antibiotic resistance genes (ARGs) and antibiotic-resistant bacteria (ARB) in urban, agricultural, and clinical settings, as well as in extreme environments, have become increasingly relevant in the One Health perspective. Since the Antarctic and Arctic regions are considered amongst the few remaining pristine environments on Earth, the characterization of their native resistome appears to be of the utmost importance to understand whether and how it is evolving as a result of anthropogenic activities and climate change. In the present review, we report on the phenotypic (e.g., disk diffusion test) and genotypic (e.g., PCR, metagenomics) approaches used to study AMR in the aquatic environment of polar regions, as water represents one of AMR main dissemination routes in nature. Their advantages and limits are described, and the emerging trends resulting from the analysis of ARB and ARGs diffusion in polar waters discussed. The resistome detected in these extreme environments appears to be mostly comparable to those from more anthropized areas, with the predominance of tetracycline, β-lactam, and sulfonamide resistance (and related ARGs). Indeed, AMR is, in all cases, more consistently highlighted in sites impacted by human and wildlife activities with respect to more pristine ones. Surprisingly, aminoglycoside and fluroquinolone determinants seem to have an even higher incidence in the Antarctic and Arctic aquatic environment compared to that from other areas of the world, corroborating the need for a more thorough AMR surveillance in these regions. Due to continuing worldwide urban expansion, research into how urban environments affect local flora/fauna has grown significantly. Studies on the impacts of urbanization on birds have explored a wide variety of behaviors (e.g., foraging, breeding, migratory), but there is little research on the impacts of cities on avian coloniality. Various urban-environmental factors may impact colonial birds. The predominance of impervious surfaces in cities, for example, has been associated with the decline of several bird species due to negative effects on availability and quality of habitat. The urban heat island effect and shifts in resource availability (e.g., food, water) may also affect colonial birds. Here, we used five years of community-science data available in eBird to investigate urban impacts on group size in Cliff Swallows (Petrochelidon pyrrhonota), an abundant colonial bird species that now breeds readily under bridges and other built structures over or near water in Phoenix, Arizona, USA. We hypothesized that, based on the colonial breeding habits of these neotropical migratory birds in this desert environment, swallows in Phoenix would form larger groups in areas with more food and water sources and with more built structures. In fact, we found that proximity to water sources and cropland, but not impervious surface density, was positively and significantly related to group size. These results suggest that, in this desert ecosystem, an abundance of food/water resources provided by humans permits Cliff Swallows to form larger social groups during breeding. Although many studies show harmful impacts of cities on local wildlife, our findings highlight how urban and/or agricultural ‘oases’ may relieve some native species from natural resource limitations and permit them to thrive and increase in group size in human-impacted environments. Traditional methods for managing free-roaming cats in Australia primarily depend on legislation and enforcement to achieve compliance. State laws and local regulations mandate confinement, sterilization, registration, and identification and limit the number of cats kept, with penalties for breaches. However, these strategies fail to address underlying issues like financial constraints in low-income areas and the prevalence of semi-owned cats. Containment mandates often result in increased complaints, shelter intake, and euthanasia, without effectively reducing cat-related problems. Research shows that these approaches are expensive, difficult to enforce, and place a disproportionate burden on disadvantaged communities. Moreover, they negatively affect the mental health of shelter staff and animal management officers, who are frequently exposed to euthanasia and ongoing challenges. An alternative “One Welfare” framework, which recognizes the interconnectedness of animal, human, and environmental welfare, has proven more effective. Programs that provide support and resources, particularly for cat sterilization and microchipping, while fostering the human–animal bond improve outcomes for both cats and caregivers. Shifting from punitive measures to collaborative, community-driven strategies is crucial for managing free-roaming cats in a way that benefits animals, people, and the broader community, while protecting wildlife. West Nile virus (WNV) is an important zoonotic pathogen belonging to the Flaviviridae family, which is endemic in some areas and emerging in others. WNV is transmitted by blood-sucking mosquitoes of the genus Culicoides, Aedes, and Anopheles, and the infection can cause different clinical symptoms. The most common and benign illness in humans is West Nile fever (WNF), but a lethal neurological disease (WNND), related to the neuro-invasiveness of WNV lineage 2, represents the highest health risk of WNV infection. The neuro-clinical form is recognized in mammals (land and cetaceans), particularly in humans (elderly or immunosuppressed) and in horses, avian species, and wildlife animals ranging free or in a zoological setting. This review highlights the most relevant data regarding epidemiology, virology, pathogenesis and immunity, clinical signs and differential diagnosis, pathology and imaging, histopathology and gross pathology, economic impact, influence of climate change, and surveillance of WNV. Climate change has favored the wide spread of WNV in many areas of the globe and consequent One-Health and Eco-Health emergencies, influencing the health of human beings, animals, and ecosystems. Human activities impact ecosystems globally, and understanding human–wildlife coexistence is crucial for species conservation. This study analyzed trends in local wildlife populations before and during the COVID-19 pandemic to assess their response to human disturbance. From 2017 to 2022, 60 camera sites were monitored, and seven species with the largest population size—excluding rodents—were selected for analysis. The results revealed that the presence of humans (p = 0.025) and domesticated animals (cats and dogs, p = 0.002) significantly decreased during the pandemic. Conversely, five species (except the Tibetan macaque and mainland serow) showed habitat expansion and population growth (p < 0.05), which may be related to their avoidance of human presence or artificial structures such as roads and tourism facilities. In addition, the analysis showed that most species, except the Tibetan macaque and wild boar, adjusted their activity patterns, showing increased diurnal activity when human disturbances were reduced (RR > 0). These findings suggest that species may adapt their behaviors to avoid human presence. This study highlights the negative impacts of human activities on local wildlife and emphasizes the need for stronger conservation and management efforts to mitigate human disturbances in scenic areas. Human–wildlife conflict poses significant ecological and socio-economic challenges, particularly in rural communities where agriculture and livestock rearing form the backbone of livelihoods. Despite the growing importance of this issue, District Lakki Marwat remains an unexplored area of northwest Pakistan. This study aims to fill this gap by systematically assessing the status, economic impacts, and community perceptions of five wildlife species: wild boar (Sus scrofa), grey wolf (Canis lupus), golden jackal (Canis aureus), striped hyena (Hyaena hyaena), and red fox (Vulpes vulpes). Using semi-structured surveys with 117 respondents, we analyzed species prevalence, perceived danger levels, crop damage patterns, and predation impacts on livestock and poultry. The findings revealed that wild boars were identified as the primary contributors to agricultural damage, with total annual crop losses surpassing the economic impacts attributed to the studied carnivores. On average, each surveyed household experienced an annual loss of PKR 4510.38. For the 39% of households reporting crop damage, the annual loss per reported household was PKR 11,727, which was higher than the average annual loss across all households, underscoring the severity of the impact on those specifically affected by the wild boar-related crop damage. Notably, community attitudes were most negative toward wild boars, a pattern driven by the economic burden of crop losses, challenging the conventional focus on carnivores as the primary conflict species. A Pearson’s X2 test confirmed strong associations between species and perceived danger levels, while regression analysis demonstrated an association between crop damage and negative attitudes. Traditional deterrents like thorn fences were found ineffective against wild boars. More advanced methods, including game-proof fencing, trenches, bio-fencing, crop rotation, audio and visual deterrents, taste and order repellents, and watchtowers combined with group vigilance, are recommended to reduce crop damage. Integrating these approaches with community-based education, habitat management, and government-supported compensation schemes can mitigate wild boar impacts. This study contributes new insights into multi-species HWC dynamics, demonstrating that community perceptions are primarily shaped by the economic impact of a species, regardless of whether it is a carnivore or an omnivore. The attitudes of local communities are driven by the financial losses incurred, rather than the species' behavior or ecological role. This study underscores the need for collaborative efforts to reduce human–wildlife conflict, foster coexistence, and ensure ecological balance in vulnerable rural areas. Full article

Background/Objectives: The spread of antimicrobial resistance (AMR) is a growing global health concern. Wild animals can play an important role in the amplification and dissemination of AMR and in conservation efforts aiming at controlling diseases in vulnerable wild animal populations. These animals can serve as reservoirs for antibiotic resistance genes and are key in the spread of AMR across ecosystems and hosts. Therefore, monitoring AMR in wild animals is crucial in tackling the spread of resistance in the environment and human population. This study investigated the phenotypic and genotypic resistance of Escherichia coli (E. coli) isolated from semi-wild oryx (Oryx leucoryx) in Qatar. Methods: One hundred fecal samples were collected from oryx in diverse natural reserves across Qatar. A selective agar medium was used to isolate E. coli, and the identity of the isolates was further confirmed using the VITEK® 2 Compact system. The Kirby–Bauer disk diffusion method was used to test antibiotic susceptibility. Genetic resistance determinants were identified through polymerase chain reaction (PCR) analyses and sequencing using the Oxford Nanopore Technology (ONT). Results: The results revealed that 18% (n = 18) of the samples harbored E. coli with resistance to a single antibiotic, 28% (n = 28) were resistant to at least one antibiotic, and 2% (n = 2) were multidrug-resistant (MDR). No resistance was observed against colistin. tetA and tetB encode tetracycline resistance were the most frequently detected genes (57.7%). Whole genome sequencing (WGS) was used to expand on AMR gene-PCR analyses and analyze the resistome of 12E. coli isolates. WGS identified several important antibiotic resistance determinates, including blaCTX-M-encoding Extended Spectrum Beta-Lactamase (ESBL) resistance, soxR associated with tetracycline target alteration, and mdtE, emrB, AcrE, mdtF, and marA related to ciprofloxacin efflux pump resistance. Conclusions: This study provides essential information regarding AMR in Qatari semi-wild animals, which will guide conservation strategies and wildlife health management in a world experiencing increasing antibiotic-resistant infections. Furthermore, these findings can inform policies to mitigate AMR spread, improve ecosystems, and enhance public and environmental health while paving the way for future research on AMR dynamics in wildlife. Urban biodiversity is essential for enhancing liveability for both humans and wildlife by providing a range of ecosystem services. Therefore, it is crucial to raise awareness among citizens, particularly schoolchildren, about the significance of biodiversity in urban environments. To this end, an Open-Air Laboratory was established in an urban park of an Italian Mediterranean city (Livorno), where natural vegetation flourishes. This initiative engaged schoolchildren in experimental campaigns to collect ecophysiological data on local wild woody species. Specifically, the students were tasked with identifying various wild woody species and, under the guidance of researchers, recording specific leaf ecophysiological traits, such as leaf mass per area, leaf pigments, and chlorophyll fluorescence. The results, which highlighted seasonal variations in leaf ecophysiological traits and interspecific differences, were analysed in relation to the environmental conditions documented by the schoolchildren. This analysis revealed distinct plant strategies for coping with winter and summer stressful periods. The methodology employed in this project, which involved schoolchildren in research activities, not only fostered environmental awareness among young participants but also serves as a pilot model for public engagement in scientific research. Wildlife sanctuaries have gained popularity in recent years as settings for research into human–animal relations, captive wildlife tourism, and rehabilitation. While scholars from animal studies, ethics, geography, and ethnography disciplines have turned their attention to sanctuaries, there is still limited engagement from the fields of tourism, conservation, and animal welfare sciences. Adopting an interdisciplinary approach, this paper addresses the ‘sanctuary gap’ by offering a synthesis of the existing research related to wildlife sanctuary tourism. To this end, the paper suggests preliminary definitions for wildlife sanctuary tourism and wildlife sanctuary attractions, as well as a typology of sanctuaries along a spectrum from greenwashed to just. It aims to illuminate the shadowy presence of wildlife sanctuaries across multiple disciplines and identify areas for future research. The discussion considers how sanctuaries are well-suited to research on multispecies communities, as well as the conservation and welfare of wild animals. Full article
