This article explores the concept of textuality as embedded within contemporary architecture, understood as the capacity of buildings to generate meanings, narratives, and interpretations that transcend their physical and functional dimensions. An interdisciplinary approach is adopted, integrating architectural theory, semiotics, hermeneutics, and cultural studies, positioning architecture as a form of symbolic production deeply intertwined with current social and technological contexts. The primary aim is to demonstrate how certain paradigmatic buildings operate as open texts that engage in dialogue with their users, urban surroundings, and cultural frameworks. The methodology combines theoretical analysis with an in-depth study of three emblematic cases: the Guggenheim Museum in Bilbao, the Centre Pompidou in Paris, and the Seattle Public Library. The findings reveal that these buildings articulate multiple layers of meaning, fostering rich and participatory interpretive experiences that influence both the perception and construction of public space. The study concludes that contemporary architecture functions as a narrative and symbolic device that actively contributes to the shaping of collective imaginaries. The article also identifies the study’s limitations and proposes future research directions concerning architectural textuality within the context of emerging digital technologies. Full article

Organoids allow to model healthy and diseased human tissues. and have applications in developmental biology, drug discovery, and cell therapy. Traditionally cultured in immersion/suspension, organoids face issues like lack of standardization, fusion, hypoxia-induced necrosis, continuous agitation, and high media volume requirements. To address these issues, we developed an air–liquid interface (ALi) technology for culturing organoids, termed AirLiwell. It uses non-adhesive microwells for generating and maintaining individualized organoids on an air–liquid interface. This method ensures high standardization, prevents organoid fusion, eliminates the need for agitation, simplifies media changes, reduces media volume, and is compatible with Good Manufacturing Practices. We compared the ALi method to standard immersion culture for midbrain organoids, detailing the process from human pluripotent stem cell (hPSC) culture to organoid maturation and analysis. Air–liquid interface organoids (3D-ALi) showed optimized size and shape standardization. RNA sequencing and immunostaining confirmed neural/dopaminergic specification. Single-cell RNA sequencing revealed that immersion organoids (3D-i) contained 16% fibroblast-like, 23% myeloid-like, and 61% neural cells (49% neurons), whereas 3D-ALi organoids comprised 99% neural cells (86% neurons). Functionally, 3D-ALi organoids showed a striking electrophysiological synchronization, unlike the heterogeneous activity of 3D-i organoids. This standardized organoid platform improves reproducibility and scalability, demonstrated here with midbrain organoids. The use of midbrain organoids is particularly relevant for neuroscience and neurodegenerative diseases, such as Parkinson’s disease, due to their high incidence, opening new perspectives in disease modeling and cell therapy. In addition to hPSC-derived organoids, the method’s versatility extends to cancer organoids and 3D cultures from primary human cells. Full article

Effects of a Positive Psychology Intervention on Mental Health and Well-Being Among Mothers in Riyadh, Saudi Arabia

Background: Concerns over women’s mental health have intensified globally, especially among mothers managing dual careers and family responsibilities. Positive Psychology Interventions (PPIs), such as gratitude journaling and well-being workshops, have demonstrated promise in enhancing mental health; however, their applicability in Arab contexts remains underexplored. This study aims to investigate the effectiveness of PPIs on mothers’ well-being, gratitude, depression, anxiety, and stress in Saudi Arabia. Methods: This quasi-experimental, one-group pretest–posttest study assessed the effects of a four-week PPI on 37 Saudi working mothers (aged 21–50 years) employed at a private school in Riyadh. The intervention included guided gratitude journaling thrice weekly and two workshops on positive psychology and gratitude. Pre- and post-intervention assessments used validated Arabic versions of the Depression Anxiety Stress Scale (DASS-21), WHO-5 Well-being Index, and Gratitude Questionnaire (GQ-6). Results: Significant improvements were found post-intervention: depression, anxiety, and stress scores decreased (p < 0.001), while well-being and gratitude increased (p = 0.001). However, participants with lower household income (<50,000 SAR) showed less improvement, indicating a potential moderating effect of socioeconomic status. Conclusions: The intervention demonstrated promising short-term improvements in mental health and well-being among Saudi mothers. The findings underscore the importance of culturally appropriate PPIs and highlight the need for further research using controlled, long-term designs. Limitations include the small, non-random sample, absence of a control group, and restriction to a single geographic region. Full article

Adult trees of pedunculate oak (Quercus robur L.) are recalcitrant to vegetative propagation. In this study, we investigated the micropropagation of five oak genotypes corresponding to trees aged 60–800 years in a liquid medium. We used commercial RITA bioreactors to study the influence of the explant type, the culture medium, shoot support and number of immersions. Variables evaluated included the number of normal and hyperhydric shoots, shoot length, multiplication coefficient and number of rootable shoots per explant. All genotypes could be cultured in temporary immersion. Basal stem sections attached to callus grew better than apical sections and developed less hyperhydricity. For long-term cultivation, Gresshoff and Doy medium was the best of the three media evaluated. All genotypes produced vigorous shoots suitable for rooting and acclimation. This is the first protocol to proliferate adult oak trees in bioreactors, representing significant progress towards large-scale propagation of this and other related species. Full article

Sociotechnical systems (STSs) are generally assumed to be systems that incorporate humans and technology, strongly depending on a sustainable equilibrium between the following nondeterministic social context ingredients: social structures, roles, and rights, as well as the designers’ Holy Grail, the deterministic nature of the underlying technical system. The fact that the relevant social concepts are more mature than the supporting technologies qualifies the digital transformation of sociotechnical systems as a reengineering rather than an engineering endeavor. Preserving the social mission throughout the digital transformation process in varying social contexts is mandatory, making the digital twins (DT) methodology application a contemporary research hotspot. In this research, we combined continuous transformation STS theory principles, an observer-based system-of-sociotechnical-systems (SoSTS) architecture model, and digital twinning methods to address common STS context representation challenges. Additionally, based on model-driven systems engineering methodology and meta-object-facility principles, the research specifies the universal meta-concepts and meta-modeling templates, supporting the creation of arbitrary sociotechnical systems’ external context digital twins. Due to the inherent diversity, significantly influenced by geopolitical, economic, and cultural influencers, a higher education external context specialization illustrates the reusability potentials of the proposed universal meta-concepts. Substituting higher-education-related meta-concepts and meta-models with arbitrary domain-dependent specializations further fosters the proposed universal meta-concepts’ reusability. Full article

This study evaluated biomass accumulation and phenolic compound production in seven Hypericum species (H. androsaemum, H. calycinum, H. hirsutum, H. kalmianum, H. olympicum, H. perforatum, and H. triquetrifolium) cultivated in vitro under varying growth regulator treatments and culture periods. Shoots were grown on Murashige and Skoog (MS) medium supplemented with benzyladenine (BA) or meta-topoline (mT) and analyzed after 40 and 60 days. MS medium supplemented with 0.2 mg/L BA was the most effective condition for promoting biomass across all species, with shoot fresh weight increasing significantly at 60 days, particularly in H. olympicum, H. perforatum, and H. triquetrifolium. High-performance liquid chromatography coupled with diode array detection and electrospray ionization mass spectrometry (HPLC-DAD-ESI-MS) identified 13 phenolic compounds, including flavonols, hydroxycinnamic acids, anthocyanins, phloroglucinols, and naphthodianthrones. Phenolic profiles were species-specific and influenced by culture period. H. kalmianum accumulated the highest total phenolic content (37.6 mg/g DW), while H. olympicum was the top producer of hypericin and pseudohypericin. These results highlight the crucial role of culture conditions in regulating both biomass and phytochemical production and provide a promising approach for producing bioactive metabolites in Hypericum species through in vitro systems. Although beer fermentation has traditionally been carried out with Saccharomyces, the boom in craft brewing has led to the use of non-conventional yeast species for beer production. This group also includes non-Saccharomyces starters, which are commonly used in winemaking and which have different technological characteristics compared to standard representatives of the Saccharomyces genus. One of the important characteristics of the non-Saccharomyces group is the richer enzyme profile, which leads to the production of beverages with different taste and aroma profiles. The aim of this study was to investigate sweet and hopped wort fermentation with seven strains of active dry non-conventional yeasts of Lachancea spp., Metschnikowia spp., Torulaspora spp. and a mixed culture of Saccharomyces cerevisiae and Torulaspora delbrueckii. One ale and one lager active dry yeast strain were used as control strains. The extract consumption, ethanol production, degree of fermentation, pH drop, as well as the yeast secondary metabolites formed by the yeast (higher alcohols, esters and aldehydes) in sweet and hopped wort were investigated. The results indicated that all of the studied types of non-conventional yeasts have serious potential for use in beer production in order to obtain new beer styles. For the purposes of this study, statistical methods, principle component analysis (PCA) and correlation analysis were used, thus establishing the difference in the fermentation kinetics of the growth in the studied species in sweet and hopped wort. It was found that hopping had a significant influence on the fermentation kinetics of some of the species, which was probably due to the inhibitory effect of the iso-alpha-acids of hops. Directions for future research with the studied yeast species in beer production are presented. Full article

Cystic fibrosis (CF), the most common hereditary lung disease in Caucasians, is caused by dysfunction of the cystic fibrosis transmembrane conductance regulator (CFTR). We evaluated CFTR function using a newly developed Ussing chamber system, the Multi Trans Epithelial Current Clamp (MTECC), in an in vitro model of human airway epithelia. Air–liquid interface (ALI) cultures were established from nasal brushings of healthy controls (HC) and CF patients with biallelic CFTR variants. ALI layer thickness was similar between groups (HC: 62 ± 13 µm; CF: 55 ± 9 µm). Immunofluorescence showed apical CFTR expression in HC, but reduced or absent signal in CF cultures. MTECC enabled continuous measurement of transepithelial resistance (Rt), potential difference (PD), and conductance (Gt). Gt was significantly reduced in CF cultures compared to HC (0.825 ± 0.024 vs. −0.054 ± 0.016 mS/cm2), indicating impaired cAMP-inducible ion transport by CFTR. Treatment of CF cultures with elexacaftor, tezacaftor, and ivacaftor (Trikafta®) increased Gt, reflecting partial restoration of CFTR function. These findings demonstrate the utility of MTECC in detecting functional differences in CFTR activity and support its use as a platform for evaluating CFTR-modulating therapies. Our model may contribute to the development of personalized treatment strategies for CF patients. Full article

Socio-Economic and Environmental Trade-Offs of Sustainable Energy Transition in Kentucky

A just and sustainable energy transition in historically coal-dependent regions like Kentucky requires more than the adoption of new technologies and market-based solutions. This study uses a stated preferences approach to evaluate public support for various attributes of energy transition programs, revealing broad backing for moving away from coal, as indicated by a negative willingness to pay (WTP) for the status quo (–USD 4.63). Key findings show strong bipartisan support for solar energy, with Democrats showing the highest WTP at USD 8.29, followed closely by Independents/Others at USD 8.22, and Republicans at USD 8.08. Wind energy also garnered support, particularly among Republicans (USD 4.04), who may view it as more industry-compatible and less ideologically polarizing. Job creation was a dominant priority across political affiliations, especially for Independents (USD 9.07), indicating a preference for tangible, near-term economic benefits. Similarly, preserving cultural values tied to coal received support among Independents/Others (USD 4.98), emphasizing the importance of place-based identity in shaping preferences. In contrast, social support programs (e.g., job retraining) and certain post-mining land uses (e.g., recreation and conservation) were less favored, possibly due to their abstract nature, delayed benefits, and political framing. Findings from Kentucky offer insights for other coal-reliant states like Wyoming, West Virginia, Pennsylvania, Indiana, and Illinois. Ultimately, equitable transitions must integrate local voices, address cultural and economic realities, and ensure community-driven planning and investment. Full article

This work explores the application of robotic systems in archaeology, highlighting their transformative role in excavation, documentation, and the preservation of cultural heritage. By combining technologies such as LiDAR, GIS, 3D modeling, sonar, and other sensors with autonomous and semi-autonomous platforms, archaeologists can now reach inaccessible sites, automate artifact analysis, and reconstruct fragmented remains with greater precision. The study provides a systematic overview of underwater, aerial, terrestrial, and other robotic systems, drawing on scientific literature that showcases their innovative use in both fieldwork and museum settings. Selected examples illustrate how robotics is being applied to solve key archaeological challenges in new and effective ways. While the paper emphasizes the potential of these technologies, it also addresses their technical, economic, and ethical limitations, concluding that successful adoption depends on interdisciplinary collaboration, careful implementation, and a balanced respect for cultural integrity. In vitro cartilage explant culture has been used to assess nutraceuticals on cartilage responses to inflammatory stimuli. However, applying extracts of nutraceuticals directly to cartilage explants does not account for effects of digestion and hepatic biotransformation, or selective exclusion of product metabolites from joint fluid by the synovial membrane. The current study produced a simulated biological extract of a common nutraceutical (glucosamine; Gsim) by exposing it to a simulated upper gastrointestinal tract digestion, hepatic biotransformation by liver microsomes, and purification to a molecular weight cut-off of 50 kDa. This extract was then used to condition cartilage explants cultured for 120 h in the presence or absence of an inflammatory stimulus (lipopolysaccharide). Media samples were analyzed for prostaglandin E2 (PGE2), glycosaminoglycan (GAG), and nitric oxide (NO). Tissue was digested and analyzed for GAG content and stained for viability. Conditioning of explants with Gsim significantly reduced media GAG in stimulated and unstimulated explants and reduced nitric oxide production in unstimulated explants. These data provide evidence for the value of glucosamine in protecting cartilage from deterioration following an inflammatory challenge, and the model improves applicability of these in vitro data to the in vivo setting. Results show that 8-shot similarity-based demonstration selection achieves optimal performance. GPT-4 achieved 20.2 BLEU for Tarifit–Arabic, 14.8 for Tarifit–French, and 10.9 for Tarifit–English. Linguistic proximity significantly impacts translation quality, with Tarifit–Arabic substantially outperforming other language pairs by 8.4 BLEU points due to shared vocabulary and morphological patterns. Error analysis reveals systematic issues with morphological complexity (42% of errors) and cultural terminology preservation (18% of errors). This work establishes baseline benchmarks for Tarifit translation and demonstrates the viability of in-context learning for morphologically complex low-resource languages, contributing to linguistic equity in AI systems. This article provides a critical review of the current design and research challenges in contemporary timber architecture. Conducted from the perspective of a designer-researcher, the review focuses on the role of wood as a material at the intersection of environmental performance, cultural meaning, and spatial practice. The study adopts a conceptual, problem-oriented approach, eschewing the conventional systematic aggregation of existing data. The objective of this study is to identify, interpret and categorise the key issues that are shaping the evolving discourse on timber architecture. The analysis is based on peer-reviewed literature published between 2020 and 2025, sourced from the Scopus and Web of Science Core Collection databases. Fifteen thematic challenges have been identified and classified according to their recognition level in academic and design contexts. The subjects under discussion include well-established topics, such as life cycle assessment and carbon storage, as well as less commonly explored areas, such as symbolic durability, social acceptance, traceability, and the upcycling of low-grade wood. The review under consideration places significant emphasis on the importance of integrating technical, cultural, and perceptual dimensions when evaluating timber architecture. The article proposes an interpretive framework combining design thinking and transdisciplinary insights. This framework aims to bridge disciplinary gaps and provide a coherent structure for understanding the complexity of timber-related challenges. The framework under discussion here encourages a broader understanding of wood as not only a sustainable building material but also a vehicle for systemic transformation in architectural culture and practice. The study’s insights may support designers, educators, and policymakers in identifying strategic priorities for the development of future-proof timber-based design practices. Full article

Ensuring bacterial safety of blood transfusions remains a critical focus in medicine. We investigated a novel pathogen reduction technology utilizing nylon functionalized with synthetic dyes (nylon affinity networks) to capture and remove bacteria from plasma. In the initial screening process, we spiked phosphate buffer solution (PBS) and human plasma (1 mL each) with 10 or 100 colony forming units (cfu) of either Escherichia coli or Staphylococcus epidermidis, exposed the suspensions to affinity networks and assessed the extent of bacterial reduction using agar plate cultures as the assay output. Nineteen synthetic dyes were tested. Among these, Alcian Blue exhibited the best performance with both bacterial strains in both PBS and plasma. Next, bacterial suspensions of approximately 1 and 2 cfu/mL in 10 and 50 mL, respectively, were treated with Alcian Blue affinity networks in three sequential capture steps. This procedure resulted in complete bacterial depletion, as demonstrated by the lack of bacterial growth in the remaining fraction. The viability of the captured bacteria was confirmed by plating the post-treatment affinity networks on agar. Alcian Blue affinity networks captured and sequestered a few plasma proteins identified by liquid chromatography tandem mass spectrometry. These findings support the potential applicability of nylon affinity networks to enhance transfusion safety, although additional investigations are needed. Full article

Although numerous studies have examined the spatial patterns of traditional villages and their driving factors, limited attention has been devoted to the transformation of tourism. This study focused on traditional villages in Shaanxi Province, employing geodetector and grounded theory methods to analyze their spatial distribution characteristics and influencing factors. First, most traditional villages have not developed tourism. Only 11.98% reached the relatively mature tourism stage. Second, the spatial distribution of mature traditional tourism villages is scattered and primarily clustered in Liuba County, Mizhi County, and Jia County. Third, the factors influencing spatial distribution characteristics include resource endowment, transportation accessibility, and regional economic conditions. Among these factors, the level of traditional villages, village heritage values, and the local tourism environment show the strongest explanatory power. These findings can help enhance cultural resilience, promote economic transformation and upgrading, and support the sustainable development of traditional villages. Full article
