Insect Mating Behaviors: A Review of the Regulatory Role of Neuropeptides

Insect mating behaviors are complex, diverse, and primarily regulated by neuropeptides and their receptors. Neuropeptides are peptide signaling molecules mainly secreted by insects’ central nervous system (CNS) to reach target organs. A substantial body of research on the role of neuropeptides in regulating mating behaviors in insects has been undertaken. This review aims to (1) synthesize existing knowledge on insect mating behaviors, (2) elucidate the neuropeptidergic mechanisms governing these behaviors, and (3) identify knowledge gaps and propose future research directions. The mating process, covering mate attraction, courtship rituals, copulation, and post-mating behaviors, was elucidated with appropriate examples. Additionally, specific neuropeptides involved at each stage of the mating process, their functions, and mechanistic aspects were discussed as demonstrated in research. The review highlights that insects display behavioral dimorphism in the mating process driven by a complex underlying neuropeptidergic mechanism. While previous publications have generally addressed the role of neuropeptides in insect behavior, none has intensively and methodically examined their role in mating behaviors. In this review, we synthesized 18 neuropeptides that we found to regulate mating behaviors in insects. We note that some of the neuropeptides are malfunctional in their regulatory roles, while others are specific. We also note that these neuropeptides execute their regulatory functions through the G protein-coupled receptor (GPCR) signaling pathway but may take different routes and messengers downstream to effect behavioral change. Neuropeptides also interact with other regulatory systems, such as the endocrine system, to discharge their functions. Given their significance in mediating mating and reproduction, targeted manipulation of the signaling system of neuropeptides could serve as viable targets in the production of ecologically friendly pest management tools. Tools that could disrupt the mating process would be applied in crop production systems to reduce the population pressure of destructive pests, consequently reducing the urge to use chemical pesticides that are ecologically unfriendly. Our findings not only advance the understanding of neuropeptide-mediated mating regulation but also highlight their potential as eco-friendly pest control targets. Full article

Tropical forest hornbills display complex social and reproductive behaviours that make them hard to reliably and frequently breed in ex situ facilities. This study investigated potential behavioural indicators of pair compatibility for two wreathed hornbills (Rhyticeros undulatus) at ARTIS Amsterdam Royal Zoo. We focused on behaviours linked to reproductive investment, such as time spent in proximity, vocalisation, and courtship display. Through systematic observations using ZooMonitor, we recorded behavioural and enclosure use data, as well as visitor presence. Key findings revealed that the male hornbill was more active and exploratory than the female. Courtship behaviours were more frequently performed by the male bird and were positively associated with perching at elevated positions. However, this behaviour decreased when the birds were in close proximity or inactive. Visitor presence influenced enclosure use, with the hornbills preferring higher perches during peak visitor times. We conclude that behaviours such as proximity, vocalisations, and courtship displays could potentially serve as indicators of compatibility in hornbill pairs but more data across more birds are needed for validation. Finally, we advocate for enclosure designs that consider hornbills’ natural behaviours and social needs to help ex situ facilities promote an environment conducive to pair bonding. Full article

Social experiences can shape adult behavior and cognition. Here, we use El Abra swordtails (Xiphophorus nigrensis) to assess how life-long experience with different male mating tactics shapes coercion evasion ability and female spatial cognition. We raised females from birth to adulthood in environments that varied by male mating tactic: coercers only, courtship displayers only, coercers and displayers together, mixed-strategists, and female only. In adulthood, we tested females’ behavioral responses to a coercive male and spatial cognition in a maze. Females reared with only displayers were significantly worse at distancing themselves from the coercive male than females raised with coercers and displayers and females raised with only coercers. Females raised with a single mating tactic (either courtship display or coercion) exhibited significantly higher accuracy in the spatial maze than females from other rearing groups, and showed significant reduction in total errors (courtship display group) or latency to reward (coercion group) over successive trials. These more predictable environments (one tactic), and not the more complex environments (two tactics), showed evidence for spatial learning. The results are discussed in light of the existing literature on two components of environmental change (environmental predictability and the certainty with which cues predict the best behavioral response) and their effect on the development of cognitive abilities. Flamingos are colonial species commonly kept in zoos, well known for their bright plumage and elaborate courtship displays. This project aimed to determine the differences in flock position and association preferences of juvenile Greater Flamingos (Phoenicopterus roseus) and Caribbean Flamingos (P. ruber) housed in the same zoological collection. Little research has been conducted on the association preferences of juvenile flamingos, especially in captive flocks, and therefore this study collected data using photographs taken throughout 2014 and 2015 to further understand association patterns. Data were collected on the age category of each juvenile flamingo observed, the age of their nearest neighbour and their position within the flock, and the location within an enclosure zone at different times of the day. The results showed that Greater Flamingo juveniles mainly associated with individuals of their own age and were most likely positioned at the periphery of their flock significantly more of the time until approximately 24 months of age. Sub-adult Greater Flamingos spent significantly more time associating with adult flamingos at the centre of the flock. In contrast, data collected on Caribbean Flamingos indicated that juveniles did not segregate themselves from the adults as distinctively. Birds aged 13–24 months were observed significantly more at the centre of the flock and had more associations with adult flamingos, in a similar manner to that observed in Greater Flamingos. Due to population management needs, juvenile Caribbean Flamingos were removed from the flock at the start of 2015 and this may have influenced the association and location preferences of the remaining young flamingos. In conclusion, these results indicated that captive juvenile flamingos were often seen away from adult birds and that sub-adult flamingos returned to the heart of their natal flock to associate significantly more with other adult individuals, potentially preparing for mate selection and breeding. Captive enclosure should therefore be spacious enough to enable young flamingos to remove themselves from adult birds so that behavioural development can be unaffected by artificially high rates of aggression. Flamingos (Phoenicopteriformes) are obligate colonial species that nest in large colonies, with monogamous pairs rearing a single chick following a synchronised group courtship display. Within this relatively simplistic behavioural description, deviations from these social and reproductive norms are apparent. Same sex pairings, multi-bird relationships and extra pair copulations are documented in the literature. Flamingos display highly sexually selected characteristics of plumage colour, carotenoid accumulation and diversity of display movements that underpin mate choice decisions. The brightest birds in best body condition are more successful at breeding. Therefore, documented mate guarding of female birds by male partners, is a relevant response to maximise investment in a pair bond. Limited information that describes the action of forced copulation by the male flamingo and the response of the female bird is available in the literature. This paper describes an occurrence of an attempted forced copulation by an older male Greater Flamingo to a younger female bird. Such behaviour may be an artefact of the captive environment, and limited mate choice when compared to the sizes of wild flocks, or it could be regularly apparent in the wild and therefore worthy of more scrutiny and empirical study. Male guppies (Poecilia reticulata) have multiple colored spots and perform courtship displays near the edges of streams in Trinidad in shallow water flowing through rainforest. Depending upon the orientation of the pair, the female sees the male displays against gravel or other stream bed substrates or against the spacelight—the roughly uniform light coming from the water column away from the bank. We observed courting pairs in two adjacent natural streams and noted the directions of each male display. We found that the female sees the male more often against spacelight than against gravel when females either faced the spacelight from the opposite bank or from downstream, or both. Visual modelling using natural substrate reflectances and field light measurements showed higher chromatic contrast of males against spacelight than against substrates independent of the two ambient light environments used during displays, but achromatic contrast depended upon the ambient light habitat. This suggests that courtship involves both chromatic and achromatic contrast. We conclude that the orientation of courting pairs and the ambient light spectrum should be accounted for in studies of mate choice, because the visual background and light affect visibility, and these differ with orientation. The rusty grain beetle, Cryptolestes ferrugineus (Stephens) (Coleoptera: Laemophloeidae), is a serious secondary pest of stored and processed food commodities. In the present study, we investigated the lateralization of males during courtship and mating, attempting to understand if it can be linked with a high likelihood of successful copulation. Most males exhibited left-biased (41%) approaches towards females, and turned 180° to their left, with 37% mating success. Right-biased males (i.e., approaching from the right and then turning 180°) were fewer than left-biased ones; 26% out of 34% managed to copulate with females. Only 9% out of 13% and 7% out of 11% of the back side- and front side-approaching males succeeded in mating, respectively. Directional asymmetries in approaching a potential mate, as well as the laterality of side-biased turning 180°, significantly affected male copulation success, with left-biased males achieving higher mating success if compared to right-biased males. Copula duration was significantly lower for left-biased-approaching males (1668.0 s) over the others (i.e., 1808.1, 1767.9 and 1746.9 for right-biased, front and back side-males, respectively). Left-biased males performed shorter copulation attempts and copula compared to right-biased males. Overall, our study adds basic knowledge to the lateralized behavioral displays during courtship and copula of C. ferrugineus. Full article

From courtship rituals, to prey identification, to displays of rivalry, a spider’s web vibrates with a symphony of information. Examining the modality of information being transmitted and how spiders interact with this information could lead to new understanding how spiders perceive the world around them through their webs, and new biological and engineering techniques that leverage this understanding. Spiders interact with their webs through a variety of body motions, including abdominal tremors, bounces, and limb jerks along threads of the web. These signals often create a large enough visual signature that the web vibrations can be analyzed using video vibrometry on high-speed video of the communication exchange. Using video vibrometry to examine these signals has numerous benefits over the conventional method of laser vibrometry, such as the ability to analyze three-dimensional vibrations and the ability to take measurements from anywhere in the web, including directly from the body of the spider itself. In this study, we developed a method of three-dimensional vibration analysis that combines video vibrometry with stereo vision, and verified this method against laser vibrometry on a black widow spiderweb that was experiencing rivalry signals from two female spiders. Lateralization has been documented in many insect species, but limited information on courtship and mating lateralization in wild conditions is available. We conducted field investigation on the courtship and mating behavior of the neem bug, Halys dentatus, a polyphagous insect mainly infesting Azadirachta indica, with particular attention to lateralization of mating displays. We investigated the presence of population-level behavioral asymmetries during H. dentatus sexual interactions and their influence on male mating success. Two lateralized traits were found: left or right-biased male approaches to the female and left or right-biased male turning displays. Males approaching females from their left side were mainly right-biased in the 180° turning display, and males that approached females from their right side were mainly left-biased. Right-biased males by turning 180° to carry out end-to-end genital contact, performed a lower number of copulation attempts, thus starting copula earlier than left-biased males. Mating success was higher when males approached the left side of females during sexual interactions. A higher number of successful mating interactions was observed in right-biased males when turning 180°. Our results add useful knowledge on the reproductive behavior of H. dentatus in the field, with potential applications for identifying useful benchmarks to monitor the quality of individuals mass-reared for pest control purposes over time. Butterflies possess impressive cognitive abilities, and investigations into the neural mechanisms underlying these abilities are increasingly being conducted. Exploring butterfly neurobiology may require the isolation of larval, pupal, and/or adult brains for further molecular and histological experiments. This procedure has been largely described in the fruit fly, but a detailed description of butterfly brain dissections is still lacking. Here, we provide a detailed written and video protocol for the removal of Bicyclus anynana adult, pupal, and larval brains. This species is gradually becoming a popular model because it uses a large set of sensory modalities, displays plastic and hormonally controlled courtship behaviour, and learns visual mate preference and olfactory preferences that can be passed on to its offspring. The extracted brain can be used for downstream analyses, such as immunostaining, DNA or RNA extraction, and the procedure can be easily adapted to other lepidopteran species and life stages. Mate choice that is based on behavioural traits is a common feature in the animal kingdom. Using the Trinidadian guppy, a species with mutual mate choice, we investigated whether males use female swimming activity—a behavioural trait known to differ consistently among individuals in many species—as a trait relevant for their mate choice. In the first experiment, we assessed male and female activity in an open field test alone (two repeated measures) and afterwards in heterosexual pairs (two repeated measures). In these pairs, we simultaneously assessed males’ mating efforts by counting the number of sexual behaviours (courtship displays and copulations). Male and female guppies showed consistent individual differences in their swimming activity when tested both alone and in a pair, and these differences were maintained across both test situations. When controlling for male swimming behaviour and both male and female body size, males performed more courtship displays towards females with higher swimming activity. In a second experiment, we tested for a directional male preference for swimming activity by presenting males video animations of low- and high-active females in a dichotomous choice test. In congruence with experiment 1, we found males to spend significantly more time in association with the high-active female stimulus. Both experiments thus point towards a directional male preference for higher activity levels in females. We discuss the adaptive significance of this preference as activity patterns might indicate individual female quality, health or reproductive state while, mechanistically, females that are more active might be more detectable to males as well. Comparative behavioural studies help reconstruct the phylogeny of closely related species. In that respect, the serows Capricornis spp. occupy an important position as they have been assumed to be the closest forms to the ancestors of Caprinae. In spite of that, information on the behavioural repertoire of the mainland serow Capricornis sumatraensis is exceedingly poor. In this paper, we report data on the activity rhythms and social behaviour of rutting mainland serows in captivity (Central Thailand, January 1986; January–February 1987). Activity was bimodal with peaks in mid-afternoon and late night. Resting and ruminating peaked at noon and twilight. Four patterns of marking behaviour were observed out of a total of 1900 events. Males and females were found to use different marking sites and frequencies. A total of 33 social behaviour patterns were observed: 18 patterns concerned agonistic behaviour, whereas 15 patterns were relevant to courtship behaviour. A comparison across Caprinae species with unritualised piercing weapons (i.e., Capricornis, Naemorhedus, Rupicapra, Budorcas, and Hemitragus) has shown that inter-sexual direct forms of aggressive behaviour are used significantly more often than indirect ones, but for chamois, confirming Rupicapra spp. as the most advanced genus among them in terms of an early ritualisation of weapons. Conversely, horns of the goral Nemorhaedus spp. and the serow lie on the same plane of the frontal bones, thus making possible the usage of a dominance display through frontal pushing. Full article

Green tea has been consumed as the most popular drink in East Asia for centuries, and is believed to have a wide range of health benefits. l-Theanine, the major component of the free amino acids in green tea, has been reported to display neuronal protection and tumor inhibition in vitro, but its physiological effects on animal development and behavior remain elusive. In this report, we used Drosophila melanogaster, the fruit fly, as a model organism to investigate the physiological effects of L-theanine. Flies were fed with three different concentrations of theanine as a dietary supplement after eclosion, and were examined for a variety of physiological parameters at different time points. We found theanine treatment results in significantly increased locomotion and courtship ability, and decreased resistance against wet and dry starvation in males, but not in females. Furthermore, theanine application diminished UV tolerance in females, but not in males. However, we did not perceive distinguishable effect of theanine on animal development, life span, weight, and tolerance of heat and anoxia. This work represents the first comprehensive physiological investigation of L-theanine at the whole animal level, and shall shed light on the mechanistic study of theanine in the future. Full article

The emphasis in the published literature has mostly been on symmetry as the critical source for beauty judgment. In fact, both symmetry and asymmetry serve as highly aesthetic sources of beauty, whether the context is perceptual or conceptual. The human brain is characterized by symbolic cognition and this type of cognition facilitates a range of aesthetic reactions. For example, both art and natural scenery contain asymmetrical elements, which nevertheless render the whole effect beautiful. A further good case in point is, in fact, human faces. Normally, faces are structurally left-right symmetrical content-wise but not size-wise or function-wise. Attractiveness has often been discussed in terms of content-wise full-face symmetry. To test whether or not attractiveness can be gleaned only from the presence of left-right full-faces we tested half faces. Three separate groups of participants viewed and rated the attractiveness of 56 full-faces (women’s and men’s), their 56 vertical left hemi-faces and 56 vertical right hemi-faces. We found no statistically significant differences in the attractiveness ratings of full- and hemi-faces (whether left or right). Instead, we found a strong and significant positive correlation between the ratings of the hemi- and full-faces. These results are consistent with the view that the underpinning of human facial beauty is complex and that bilateral symmetry does not constitute a principle factor in beauty assessment. We discuss that the highly evolved human brain, compared to other animals, as well as symbolic and abstract cognition in humans enable a wide variety of aesthetic reactions. Full article
