Consumer interest in vegetarian, ethical, and clean-label foods is reviving the use of plant-derived milk coagulants. Cardosins from Cynara cardunculus (“thistle”) are aspartic proteases with strong clotting activity, yet their technological impact in cheese remains under-explored. This study compared a commercial thistle extract (PC) with traditional bovine rennet rich in chymosin (AC) during manufacture and 60-day ripening of Caciotta cheese. Classical compositional assays (ripening index, texture profile, color, solubility) were integrated with scanning electron microscopy, three-dimensional surface reconstruction, and descriptive sensory analysis. AC cheeses displayed slower but sustained proteolysis, yielding a higher and more linear ripening index, softer body, greater solubility, and brighter, more yellow appearance. Imaging revealed a continuous protein matrix with uniformly distributed, larger pores, consistent with a dairy-like sensory profile dominated by milky and umami notes. Conversely, PC cheeses underwent rapid early proteolysis that plateaued, producing firmer, chewier curds with lower solubility and darker color. Micrographs showed a fragmented matrix with smaller, heterogeneous pores; sensory evaluation highlighted vegetal, bitter, and astringent attributes. The data demonstrate that thistle coagulant can successfully replace animal rennet but generates cheeses with distinct structural and sensory fingerprints. The optimization of process parameters is therefore required when targeting specific product styles. Full article

Goat milk has lower allergenicity and high commercial value but faces storage limitations, often leading to waste. Converting it into powder increases costs, making blending with non-dairy ingredients, such as rice flour, a viable alternative to reduce costs and potentially improve nutrition. In this this study, we developed five dairy compounds by replacing 10–49% of goat milk powder with rice flour. We evaluated their nutritional and physical properties compared to pure goat milk powder and rice flour. Analyses included water activity, total solids, protein, lipids, energy value, color, flowability, wettability, polyphenol content, mineral profile, and morphology. Higher rice flour content increased water activity and improved wettability but reduced flowability, classifying most compounds as reasonable to fair in flow, except for the 10% rice flour sample. All samples met Brazilian standards, which require ≥13 g/100 g of protein. The dairy compounds showed a yellow-greenish color, with significant color differences compared to goat milk powder, particularly at 49% rice flour. Goat milk powder had higher mineral contents (Ca, K, Mg, Na, P, Zn). Total polyphenol content was highest in the 10% rice flour compound, while individual polyphenols were undetectable. Overall, the formulation proved viable for cost reduction while maintaining nutritional quality. Full article

Hemp (Cannabis sativa L.) is a versatile and multipurpose crop with a wide range of industrial, commercial, environmental and medicinal applications. This study aimed to evaluate the effects of hemp hay on the oxidative and inflammatory status of grazing goats. The high content of polyunsaturated fatty acids with anti-inflammatory activity makes hemp hay a promising feed that can be used to improve animals’ health by means of its metabolic profile, as well as its oxidative and inflammatory status. Studies on hemp hay suggest that its usage could replace alfalfa hay to obtain beneficial effects in terms of the milk yield and health status of goats. Indeed, because of the various biological compounds in hemp hay, the effects of its administration should be accurately explored. A total of 20 multiparous goats were equally divided immediately after calving (February) into two groups (C: control vs. H: hemp) that were homogeneous in terms of live weight (49 ± 2 kg) and milk yield at the previous lactation (2250 ± 200 g/head/day). The animals remained in the pasture but received 500 g/head/day of a barley and corn meal mixture. In addition, group H received 250 g/head/day of hemp hay, and group C was fed the same amount of alfalfa hay. Blood samples were taken before the trial and four months later and analyzed for clinical biochemistry, oxidative status, and anti-inflammatory profile. Reactive oxygen metabolites, biological antioxidant potential, Interleukins 6 (IL-6) and 10 (IL-10), and TNF-α were determined. In group H, a significant decrease in TNF-α level (22.7 vs. 12.7 pg/mL, p = 0.01) and an opposite but not significant trend for IL-10 was detected. In addition, there was a significant decrease (184.6 vs. 111.4 UCARR, p = 0.001) in d-ROM content and an increase in the biological antioxidant potential (BAP) values (3238.1 vs. 3886 μmol/L). A significant anti-inflammatory effect was only registered for TNF-α, whereas IL6 and IL10 were not affected. Full article

The objective of the present study was to determine whether the addition of a blend based on the essential oils of cinnamon, oregano, and eucalyptus to the liquid diets of calves would stimulate the immune system combined with anti-inflammatory action, minimize oxidative responses, and alter the intestinal microbiota, consequently enhancing animal growth. Twenty-four male Holstein calves (approximately five days old) were suckled for 60 days, underwent a weaning process, and were followed up until day 75 of the experiment. The calves were divided into control (n = 12) and phytobiotic (n = 12) groups, receiving commercial milk replacer and pelleted concentrate ad libitum. For the phytobiotic group, we added the blend to the liquid diet twice daily at 5 mL/feeding in the first 15 days and 10 mL/feeding until day 60. We detected no differences in weight gain, but animals in the phytobiotic group tended to consume less feed. Calves treated with phytobiotics showed better conversion and feed efficiency than the animals in the control group. Lower leukocyte and lymphocyte counts were observed, as was a higher cholesterol concentration. Immunoglobulin A, ceruloplasmin, and transferrin also differed between groups, with higher IgA and lower levels of acute-phase proteins (ceruloplasmin and transferrin) in calves that consumed the phytobiotic. Higher glutathione S-transferase activity was found in the serum of calves in the treatment group. The intestinal microbiota did not differ between the groups; however, the genera Acinetobacter, Pseudomonas, and Psychrobacter were the most abundant regardless of treatment. We concluded that the blend of oils based on cinnamon, oregano, and eucalyptus improved the calves’ immune and antioxidant systems, improving feed efficiency without affecting the intestinal microbiota. Background: Polar lipids from dairy are novel sources of energy that may replace other dietary lipids and impact plasma lipidomic profiles in piglets. This study evaluated the impact of feeding diets rich in polar lipids on the plasma lipidome of piglets during the weaning period. Material and Methods: Weaned male piglets (n = 240; 21 days of age; 6.3 ± 0.5 kg of BW) were blocked by initial weight and distributed into 48 pens of five animals each in a complete randomized block design with a 2 × 3 factorial arrangement of treatments as follows: a plant-based diet rich in neutral lipids from soybeans (24 pens; SD) or a polar lipid-rich diet by-product of cheese making (24 pens; PD) from weaning until the 21st day of the nursery phase. Within each diet group, animals received one of three milk replacers (MR; 0.5 L/d/animal) for the first 7 days after weaning: (1) commercial MR containing animal and coconut lipids (CO); (2) polar lipid-based MR (PO); or (3) soybean lipids-based MR (SO). Results: The PD diet group increased the plasma concentrations of sphingolipids, phospholipids, and cholesterol esters, but did not impact the concentrations of glycerolipids (GLs). Both the PO and CO milk replacers increased the plasma concentrations of ceramide, acyl-chain phosphatidyl choline, and cholesterol esters. The plasma concentrations of GLs containing 18-carbon fatty acids such as 18:0, 18:1, 18:2, and 18:3, were higher in SD, whereas GLs containing 16:0 and 20:3 were higher in PD. Conclusions: In summary, the diet lipid type significantly modulated the plasma lipid composition in piglets 7 days after weaning. The dietary inclusion of polar lipids in diets for growing pigs can modulate the plasma lipidomic profile, relative to plant-based diets rich in soybean lipids. Cost may be a major consideration when using these lipids in pig diets. Their health benefits need to be further characterized in other models of stress and inflammation. Full article

We compared the effects of milk-feeding in 288 Holstein calves (72 per group) which were fed twice (2F) or thrice (3F) daily, with or without the addition of hydrogenated fat-embedded calcium gluconate (G) supplemented in the starter food and in the daily diet up to the age of 9 months, on the calves’ metabolism, growth, health, and reproductive efficiency up to first pregnancy. The calves received 6 L of milk replacer (130 g/L) and had ad libitum access to water and textured calf starter with or without gluconate. Gluconate supplementation promoted a “catch-up” in growth in supplemented calves compared to their counterparts that did not receive gluconate. Gluconate appeared to reduce animal metabolic stress during key events, such as weaning and transfer into open-door pens, reducing fructosamine (352.61 vs. 303.06 in 3FG and 3F, respectively; p = 0.028) and urea (3F revealed the highest values compared with the other three groups: 19.06 for 3F vs. 13.9 (2F), 13.7 (2FG), and 14.3 (3FG), respectively, p = 0.002) from weaning onwards. The feeding of dairy calves with milk replacer three rather than two times per day tended to be associated with better health from weaning to 4 months old; parameters such as ultrasound lung score and calf health score improved over time (p < 0.001). Thrice-daily feeding with milk replacer tended to reduce the number of artificial inseminations per pregnancy in heifers by 0.2 points (p = 0.092). We confirmed significant correlations between early health and growth parameters and reproductive efficiency and a positive correlation between body weight and average daily weight gain and the thickness of the back fat layer in young heifers (r = 0.245; p < 0.0001; r = 0.214; p < 0.0001 respectively). Our study was conducted on a commercial farm with reasonably effective animal management, so baseline welfare was likely satisfactory. Full article

Lactoferrin (LF) stands as one of the extensively investigated iron-binding glycoproteins within milk, exhibiting diverse biological functionalities. The global demand for LF has experienced consistent growth. Biotechnological strategies aimed at enhancing LF productivity through microbial expression systems offer substantial cost-effective advantages and exhibit fewer constraints compared to traditional animal bioreactor technologies. This study devised a novel recombinant plasmid, wherein the AOX1 promoter was replaced with a glucose-inducible G1 promoter (PG1) to govern the expression of recombinant porcine LF (rpLF) in Pichia pastoris GS115. High-copy-number PG1-rpLF yeast clones were meticulously selected, and subsequent induction with 0.05 g/L glucose demonstrated robust secretion of rpLF. Scaling up production transpired in a 5 L fermenter, yielding an estimated rpLF productivity of approximately 2.8 g/L by the conclusion of glycerol-fed fermentation. A three-step purification process involving tangential-flow ultrafiltration yielded approximately 6.55 g of rpLF crude (approximately 85% purity). Notably, exceptional purity of rpLF was achieved through sequential heparin and size-exclusion column purification. Comparatively, the present glucose-inducible system outperformed our previous methanol-induced system, which yielded a level of 87 mg/L of extracellular rpLF secretion. Furthermore, yeast-produced rpLF demonstrated affinity for ferric ions (Fe3+) and exhibited growth inhibition against various pathogenic microbes (E. coli, S. aureus, and C. albicans) and human cancer cells (A549, MDA-MB-231, and Hep3B), similar to commercial bovine LF (bLF). Intriguingly, the hydrolysate of rpLF (rpLFH) manifested heightened antimicrobial and anticancer effects compared to its intact form. In conclusion, this study presents an efficient glucose-inducible yeast expression system for large-scale production and purification of active rpLF protein with the potential for veterinary or medical applications. Mycotoxins are toxic mold metabolites that can adversely affect human and animal health. More than 400 mycotoxins have been identified so far. Cereals and nuts are the predominant mycotoxin-contaminated foodstuffs. Plant-based drinks produced from cereals, nuts, and legumes have grown in popularity. The mycotoxins accumulated in these crops may transfer to these beverages. A liquid chromatography–tandem mass spectrometry method was developed and optimized for the assessment of 22 mycotoxins in commercially available plant-based drinks in Latvia and Lithuania. A total of 64% of the seventy-two analyzed beverages were positive for one to sixteen mycotoxins, with deoxynivalenol, beauvericin, and enniatins A, B, B1, T-2, and HT-2 toxins detected most frequently. The European Commission has not yet set guidelines for the maximum mycotoxin concentrations in plant-based beverages, nor has the European Food Safety Authority conducted a risk assessment. Therefore, acute exposure studies were provided for the Latvian population based on the assumed replacement of dairy milk with plant-based beverages to ascertain the safety of plant-based milk substitutes. Based on the observed levels of mycotoxin prevalence and contamination levels and assumed exposure, it can be concluded that tested plant-based beverages may be relatively safe. Scotta is an underutilized whey by-product of ricotta making. In this study, we investigated the utilization of scotta in beer production. To understand the quality of regional scotta, samples from eight Sicilian ricotta makers were analyzed for pH, total protein, fat, lactose, titratable acidity, % salt, total plate count, lactic acid bacteria counts, and minerals. Overall, the samples had low amounts of residual protein and fat. The average lactose content was 4.81 g/100 mL ± 0.52 g, with a pH of 6.12 ± 0.17 and a salt content of 1.05% ± 0.24. The majority of lactic acid bacteria counts were below the limit of detection. The total plate counts were more variable, ranging between 102 to 103 CFU/mL, suggesting occasional post-processing contamination during handling. Scotta was then used to replace some of the water and sugar in the production of two beer styles: (i) a Gose, a salty and acidified German beer style, and (ii) a sweet milk stout. A trained panel used for sensory analysis found that these prototypes fit within the sensory profiles of commercial beers of these styles. This work highlights opportunities to upcycle dairy by-products into novel fermented beverages that would be appealing to consumers. Full article

The objective of this research was to determine the effect of cassava (Manihot esculenta Crantz) supplementation on enteric methane (CH4) emissions, carbon footprint, and production parameters in dairy cows. Daily concentrate supply for Jersey and Jersey * Holstein breeds was evaluated in four treatments (T): T1: 100% commercial concentrate; T2: 70% concentrate + 30% cassava leaves; T3: 70% concentrate + 30% cassava roots; and T4: 70% concentrate + 15% cassava leaves + 15% cassava root chips. Measurements of CH4 emissions were performed using the polytunnel technique. Average daily dry matter intake ranged from 7.8 to 8.5 kg dry matter (DM). Cassava leaves were characterized by a high crude protein (CP) content (171 g CP/kg DM), with 5 times more neutral detergent fiber (NDF) content than cassava root (587 vs. 108 g NDF/kg DM). Average enteric CH4 emissions per animal ranged from 194 to 234 g/d (p > 0.05). The carbon footprint was reduced by replacing 30% of the concentrate with cassava leaves and/or roots. Energy-corrected milk production was 1.15 times higher in Jersey * Holstein animals than Jersey cows (47 vs. 55 kg). Therefore, supplementation with cassava leaves and/or roots is a nutritionally and environmentally sustainable strategy. Full article

This assay is potentially also useful to analyse serum samples from first-season grazing (FSG) calves, providing a more cost-effective and robust diagnostic technique than the current serum pepsinogen assay. However, a comprehensive evaluation of its use in serum samples from FSG calves has not yet been conducted. In this study, we first reviewed the available scientific literature in which the O. ostertagi-Ab ELISA was applied to serum samples from FSG calves. Then, a field study was conducted to compare results from the O. ostertagi-Ab ELISA assay with a serum pepsinogen assay on a set of 230 serum samples from 11 commercial dairy herds (seven in Belgium and four in Sweden). The literature review showed an increase in mean antibody levels, expressed as optical density ratio (ODR) values, from <0.4 (early grazing season) to values of 0.7–1.1 (late grazing season). Three out of five studies found a negative correlation between O. ostertagi antibody levels measured during the late grazing season and weight gain, while the other two studies found no correlation between the two variables. Our field studies showed a good correlation between O. ostertagi antibody levels and the results from the pepsinogen assay. Both indicators were negatively related to average daily weight gain in the Belgian herds, but not in the Swedish herds. Overall, the results suggest that the O. ostertagi-Ab ELISA test can be a useful tool in FSG calves and could replace the use of the serum pepsinogen assay at the end of the grazing season for general monitoring purposes. The omasum is the third compartment of the ruminant stomach, which is also considered a water absorption organ and participates in the absorption of volatile fatty acids (VFA), minerals, electrolytes, and fluids. The most important morphological parameter of the omasum is the available absorption area, which depends on the size and number of the omasal laminae, and is variable among different ruminants and based on differences in their daily diets. Optimal omasum development in the transition period to ruminant life can enhance animal performance, so identifying the best diet for this period is crucial for producers. The objective of this study was to determine the effect of two diets based on 8 L of milk replacer with the inclusion of concentrate or forage on the development of the omasum in twenty newborn male Holstein calves divided into two groups. The first group was fed alfalfa hay, and the second was administered a balanced commercial starter feed, both groups ad libitum. After standard dissection of the omasum of both calf groups, the omasal laminae were classified as primary, secondary, and tertiary, and their surface area was calculated. Regarding the number of first-, second-, and third-order laminae, a significant difference was only observed in the number of third-order laminae in favor of the forage-fed group (p = 0.04). The laminar surface area indicated that the area of the primary, secondary, and tertiary sheets, and the total laminar area, were greater in the forage group (p < 0.05). The length of the omasal papillae recorded with the scanning electron microscope showed that the papillae near the ostium reticulo-omasicum tended to be significantly longer in the forage-fed group (p = 0.05). In conclusion, this study demonstrates that significant anatomical differences can be observed between two groups of animals of the same species and rearing stage that were fed with two different diets within a period of less than two months. This highlights the remarkable plasticity and adaptability of the ruminant stomach. Full article

Feeding increased volumes of milk in the preweaning phase has been shown to improve growth, morbidity and mortality rates in calves (Bos Taurus). This experiment enlisted 20 Holstein-Friesian dairy replacement calves from birth until weaning (at 10 weeks of age) and assessed the effect of feeding either 4 L (Low) or 8 L (High) of milk per calf per day on their growth, immune competence and metabolic characteristics. The responsiveness of these systems was compared through a vaccination immune challenge. Calves in the High treatment group were significantly heavier from two weeks of age and were 19 kg heavier than calves in the Low treatment group at weaning. Calves in the High treatment group also exhibited greater immune responses, with significantly higher white cell counts and neutrophil counts than calves in the Low treatment group post-vaccination. Calves in the High treatment group also had lower beta-hydroxybutyrate both pre- and post-vaccination, and higher glucose and insulin levels post-vaccination, indicating superior metabolic characteristics. Calves had ad libitum access to lucerne hay (Medicago sativa) and a commercial concentrate. Solid feed intakes were mostly the same between treatments, with differences in hay intake only detected at 7 and 8 weeks of age. Results from this experiment are indicative of a positive influence of accelerated preweaning nutrition on growth, immune response and metabolic characteristics. Full article

The continuous increase in the cost of feeds and the need to improve the sustainability of animal production require the identification of alternative feeds, such as those derived from the agro-industrial sector, that can be effectively used for animal nutrition. Since these by-products (BP) are sources of bioactive substances, especially polyphenols, they may play an important role as a new resource for improving the nutritional value of animal-derived products, being effective in the modulation of the biohydrogenation process in the rumen, and, hence, in the composition of milk fatty acids (FA). The main objective of this work was to evaluate if the inclusion of BP in the diets of dairy ruminants, as a partial replacement of concentrates, could improve the nutritional quality of dairy products without having negative effects on animal production traits. To meet this goal, we summarized the effects of widespread agro-industrial by-products such as grape pomace or grape marc, pomegranate, olive cake, and tomato pomace on milk production, milk composition, and FA profile in dairy cows, sheep, and goats. The results evidenced that substitution of part of the ratio ingredients, mainly concentrates, in general, does not affect milk production and its main components, but at the highest tested doses, it can depress the yield within the range of 10–12%. However, the general positive effect on milk FA profile was evident by using almost all BP at different tested doses. The inclusion of these BP in the ration, from 5% up to 40% of dry matter (DM), did not depress milk yield, fat, or protein production, demonstrating positive features in terms of both economic and environmental sustainability and the reduction of human–animal competition for food. The general improvement of the nutritional quality of milk fat related to the inclusion of these BP in dairy ruminant diets is an important advantage for the commercial promotion of dairy products resulting from the recycling of agro-industrial by-products. The aim of the current study was to investigate using a customized profit and carbon total merit index to identify sustainable milking cows and herd replacements within a commercial dairy herd. Balancing the economic, social and environmental aspects of milk production has gained interest given the increasing global demand for milk products. Furthermore, a farm-level customized breeding index with farm-derived weightings for biological traits would incorporate the effect of the farm environment. This study used a Markov chain approach to model a commercial dairy herd in the UK between the years 2017 and 2022. Production, financial, genetic and nutritional data for the herd were used as input data. The model derived the economic (GBP per unit) and carbon values (kilograms CO2-eq. emissions per unit) for a single phenotypic increase in milk volume, milk fat yield, milk protein yield, somatic cell count, calving interval and lifespan, which were used in a profit and carbon index. The study proposed a methodology for selecting individual milking cows and herd replacements based on their potential to increase herd profitability and reduce carbon emissions as a means to identify more sustainable animals for a given farm environment. Of the 370 cows and herd replacements studied, 76% were classified as sustainable with a desirable increase in profit and reduction in carbon emissions. Customized breeding indices with trait weightings derived from the farm environment and selecting individual animals on economic and carbon metrics will bring permanent and cumulative improvements to the sustainability of milk production with appropriate nutrition and management. The approach used can be applied to any commercial farm to select animals that are more sustainable. Full article
