Effect of Plant Extracts Combinations on TNF-α, IL-6 and IL-10 Levels in Serum of Rats Exposed to Acute and Chronic Stress

Oxydative stress, anxiety and depression are associated with changes in cytokine levels. Natural products, including individual and combined plant extracts, have the potential to be used in the treatment of neuropsychiatric disorders. The goal of this study is to investigate the effects of two combined plant extracts, rich in flavonoids, on the levels of the cytokines TNF-α, IL-6, and IL-10 in rats subjected to models of acute cold stress and chronic unpredictable stress. The study utilized common medicinal plants such as Valeriana officinalis, Melissa officinalis, Crataegus monogyna, Hypericum perforatum, and Serratula coronata, which were combined in two unique combinations—Antistress I and Antistress II. The compositions of the used extracts were determined by HPLC methods. Pro- and anti-inflammatory cytokines in rats’ serum were measured with Enzyme-linked immunosorbent assay. The results from the acute stress model revealed that the individual extract of Crataegus monogyna decreased levels of TNF-α, while Serratula coronata, Hypericum perforatum, and Valeriana officinalis effectively reduced IL-6 levels. Both combinations, Antistress I and Antistress II, were effective in reducing TNF-α and IL-6 levels, with Antistress II also increasing IL-10 levels. In the chronic stress model, Hypericum perforatum extract decreased the levels of the pro-inflammatory cytokines TNF-α and IL-6, whereas extracts of Serratula coronata and Valeriana officinalis only reduced TNF-α levels. The two combined extracts, Antistress I and Antistress II, decreased TNF-α and IL-6 levels, while Antistress I also reduced the levels of the anti-inflammatory cytokine IL-10. The combinations of plant extracts used in our experiment have not been previously studied or documented in the available literature. However, based on our own experimental results, we can draw the conclusion that the combinations exhibit a more pronounced effect in reducing cytokine levels compared to the individual plant extracts. Phytoecdysteroids exert significant anti-inflammatory effects, which makes them valuable ingredients in pharmaceutical and cosmetic products. However, data on their use in cosmetics are limited. Here, a new formulation with the extract of the Serratula coronata herb containing phytoecdysteroids was developed. The aim of this study was to perform physicochemical characteristics and evaluate the safety of the creams with S. coronata. Chromatography was used to detect the dominant phytoecdysteroids in the extract. The chemical and physical description of the creams was performed using the following parameters: viscosity, pH, and stability. The microbiological purity (pharmacopoeial methods) and transdermal permeability (Raman spectroscopy) were assessed to ensure the safety of the plant extracts used in the creams. The study confirmed the presence of phytoecdysteroid fractions of the S. coronata herb in the creams (20-hydroxyecdysone, polypodine B, and ajugasterone C). The results indicated that the cosmetics containing the S. coronata extract were chemically and microbiologically stable, thereby contributing to their safety. Their effectiveness is the result of transdermal permeability of 20-hydroxyecdysone. In this study, we demonstrated the importance of the S. coronata extract as a source of bioactive phytoecdysteroids and proved that the extract’s characteristics may make it the key ingredient of safe and stable skincare products that support the treatment of various inflammatory skin diseases. Serratula L. and Klasea Cass. are two systematically related genera of the family Asteraceae, which are distributed in most of the Eurasia area and are used as food and colorants and in traditional medicines as a drug. Since 1967, 261 metabolites have been isolated and identified from five Serratula species and 21 Klasea species. This review provides information on the chemodiversity of the terpenes, penolics, lipids, and other compounds found in both genera and their occurrence in individual species. Among the studied species, the most studied are S. coronata subsp. coronata, K. centauroides, and K. centauroides subsp. centauroides. This review also provides information on the methods of extraction, isolation, and analysis of ecdysteroids and flavonoids as the most valuable metabolites. For the first time, we provide general information about the biological activity of these extracts and individual compounds. The data presented in this review demonstrate the prospects of Serratula and Klasea species as sources of bioactive metabolites. Full article

In this study, the potential of biogenic zinc oxide nanoparticles (ZnO NPs) in the removal of alizarin yellow R (AY) from aqueous solutions by photocatalytic degradation, as well as adsorption, was investigated. The synthesized ZnO NPs were prepared by the simple wet-combustion method using the plant extract of Serratula coronata L. as a reducing and stabilizing agent and characterized by powder X-ray diffraction, scanning electron microscopy, energy dispersive X-ray and X-ray photoelectron spectroscopy. Photocatalytic degradation of AY was monitored by UV–visible spectroscopy and the effects of parameters, such as light source type (UV-, visible- and sunlight), incubation time, pH, catalyst dosage and temperature on degradation were investigated. It was demonstrated that the source of light plays an important role in the efficiency of the reaction and the UV-assisted degradation of AY was the most effective, compared to the others. The degradation reaction of AY was found to follow the Langmuir-Hinshelwood mechanism and a pseudo-first-order kinetic model. The degradation kinetics of AY accelerated with increasing temperature, and the lowest activation energy (Ea) was calculated as 3.4 kJ/mol for the UV-light irradiation system, while the Ea values were 4.18 and 7.37 kJ/mol for visible light and sunlight, respectively. The dye removal by the adsorption process was also affected by several parameters, such as pH, sorbent amount and contact time. The data obtained in the kinetics study fit the pseudo-second-order equation best model and the rate constant was calculated as 0.001 g/mg·min. The isotherm analysis indicated that the equilibrium data fit well with the Freundlich isotherm model. The maximum adsorption capacity of AY on biogenic ZnO NPs was 5.34 mg/g. Full article

Phytoecdysones from Serratula coronata seem to be promising agents for skincare in patients with psoriasis. The aim of the study was to determine the effects of creams containing the extract of S. coronata on psoriatic lesions. Creams with different formulas were prepared: 0-Lekobaza®, 1-Lekobaza®, S. coronata, 2-Lekobaza®, Salicylic acid, 3-Lekobaza®, S. coronata, Salicylic acid. After examination of skin penetration and biosafety, the designated cream was applied twice daily for 6 weeks on 72 psoriatic plaques located on elbows or knees. The lesions were assessed at baseline and follow-up of 6 weeks. The lesions area was measured, and severity of scaling, erythema, and infiltration was assessed using a 5-point scale (from 0—none to 4—very severe). Skin hydration and structure, pH, transepidermal water loss, erythema, and melanin index were analyzed instrumentally. Creams 1, 2, and 3 significantly reduced the area of psoriatic plaques. Improvement in erythema and infiltration was observed for creams 1 and 3. Creams 1–3 reduced scaling. Our study confirmed a beneficial effect of creams containing S. coronata extract on psoriatic lesions. Full article

Phytoecdysteroids are natural compounds with therapeutic benefits in both humans and animals. The effectiveness of natural products with health potential is based on the activities and potencies of their active ingredients. In this study, dominant ecdysteroids—ajugasterone C, 20-hydroxyecdysone and polypodine B—from the Serratula coronata (S. coronata) herb were separated by column chromatography, identified by spectroscopic data and quantified by high-performance liquid chromatography with a diode array detector (HPLC-DAD). The obtained concentration of ecdysteroids (approximately 23%) in the S. coronatae herb extract enhances the possibility of their use in pharmaceutical and cosmetic products with high levels of phytoecdysteroids. Moreover, this study has shown a positive effect of ecdysteroids-containing cream on changes in quality of life and a beneficial effect in reducing the symptoms of seborrheic dermatitis. It has been demonstrated that the application of the cream with phytoecdysteroids resulted in a statistically significant alleviation of symptoms (p < 0.05), especially in terms of itching, pain or burning sensations in the affected areas in comparison to previous symptoms.
