Dear Colleagues,

The historical paradigm of cancer metabolism, long dominated by the Warburg effect’s emphasis on aerobic glycolysis, has undergone a fundamental transformation. We now recognize that mitochondria are not merely dysfunctional bystanders but are dynamic, biosynthetic, and signaling hubs that actively drive tumorigenesis, metastasis, and therapeutic resistance. This Special Issue, “Harnessing Mitochondrial Rewiring for Cancer Therapy: Novel Strategies in the Era of Precision Medicine”, aims to crystallize the rapid advancements in our understanding of mitochondrial plasticity. It addresses the critical need to decipher how cancer cells reprogram mitochondrial networks to survive distinct microenvironmental stresses, including hypoxia, nutrient deprivation, and immune surveillance. The scope of this Special Issue encompasses the full spectrum of mitochondrial biology in oncology: from the dysregulation of the electron transport chain and the accumulation of oncometabolites like 2-hydroxyglutarate and succinate to the macroscopic dynamics of fission and fusion that propel metastatic invasion. We invite authors to submit original research and comprehensive reviews that illuminate these mechanisms and propose innovative strategies to dismantle the mitochondrial engine of malignancy, and particularly welcome contributions that bridge the gap between their basic biology and clinical application.

Submitted manuscripts should not have been published previously, nor be under consideration for publication elsewhere (except conference proceedings papers). All manuscripts are thoroughly refereed through a single-blind peer-review process. Life is an international peer-reviewed open access monthly journal published by MDPI.
