Special Issue Information

Dear Colleagues,

Brain and other central nervous system (CNS) tumors are the leading cause of cancer-related death in children, and frequently result in substantial long-term morbidity as well as disability. Recent years have seen remarkable progress in brain cancer biology across a large number of tumor types and histologies; however, improvements in long-term survival and quality of life for CNS tumors have remained elusive compared to other childhood cancers. Although the classification of CNS tumors was historically based on histologic features only, advances in our understanding of the molecular features of CNS tumors have led to the incorporation of molecular alterations into the diagnostic criteria. The 2021 revised fifth edition of the World Health Organization (WHO) Classification of Tumors of the CNS (WHO CNS5) further integrates molecular alterations for CNS tumor categorization, updating the current understanding of the pathophysiology of many of these disease entities with better-characterized natural histories as well as introducing new tumor types and subtypes, especially in the pediatric population. Optimizing the treatment of childhood CNS tumors remains a tremendously challenging task, requiring a multidisciplinary approach involving many pediatric specialists as well as the support of molecular biologists and pharmacologists.
