Cover Story: This study describes the fabrication of hierarchical scaffolds aimed at mimicing the native osteochondral tissue architecture. A complex 3D bilayered structure was composed of two layers, (i) chitosan/β-TCP, which resembles bone, and (ii) silk fibroin, which resembles cartilage, whereas genipin was applied as a natural crosslinking agent to produce the composite architectures. The findings revealed that genipin crosslinking promoted a structural rearrangement in the scaffolds, enhancing the interaction between layers, and improved the composite scaffold properties. The biological outcomes corroborated that the developed hierarchical scaffolds show good ATDC5 and MC3T3 cell adherence and proliferation for 14 days. Overall, the physicochemical properties and biological data suggest that they may be potential candidates for osteochondral tissue strategies.
